Electrospinning of polyacrylonitrile/DMF dopes containing salts of nickel, cobalt, zirconium, cerium, gadolinium, and samarium, makes it possible to obtain precursor nanofiber mats which can be subsequently converted into carbon nanofiber (CNF) composites by pyrolysis at 1000-1200 °C. Inorganic additives were found to be uniformly distributed in CNFs. Metal states were investigated by transmission electron microscopy and X-ray photoelectron spectroscopy (XPS). According to XPS in CNF/Zr/Ni/Gd composites pyrolyzed at 1000 °C, nickel exists as Ni0 and as Ni2+, gadolinium as Gd3+, and zirconium as Zr4+. If CNF/Zr/Ni/Gd is pyrolyzed at 1200 °C, nickel exists only as Ni0. For CNF/Sm/Co composite, samarium is in Sm3+ form when cobalt is not found on a surface. For CNF/Zr/Ni/Ce composite, cerium exists both as Ce4+ and as Ce3+. Composite CNF mats were platinized and tested as cathodes in high-temperature polymer electrolyte membrane fuel cell (HT-PEMFC). Such approach allows to introduce Pt-M and Pt-MOx into CNF, which are more durable compared to carbon black under HT-PEMFC operation. For CNF/Zr/Ni/Gd composite cathode, higher performance in the HT-PEMFC at I >1.2 A cm-2 is achieved due to elimination of mass transfer losses in gas-diffusion electrode compared to commercial Celtec®P1000.The solubility values and thermodynamic parameters of a natural phytomedicine/nutrient piperine (PPN) in Transcutol-HP (THP) + water combinations were determined. The mole fraction solubilities (xe) of PPN in THP + water combinations were recorded at T = 298.2-318.2 K and p = 0.1 MPa by the shake flask method. Hansen solubility parameters (HSPs) of PPN, pure THP, pure water and THP + water mixtures free of PPN were also computed. The xe values of PPN were correlated well with "Apelblat, Van't Hoff, Yalkowsky-Roseman, Jouyban-Acree and Jouyban-Acree-Van't Hoff" models with root mean square deviations of less then 2.0%. The maximum and minimum xe value of PPN was found in pure THP (9.10 × 10-2 at T = 318.2 K) and pure water (1.03 × 10-5 at T = 298.2 K), respectively. In addition, HSP of PPN was observed more closed with that of pure THP. The thermodynamic parameters of PPN were obtained using the activity coefficient model. The results showed an endothermic dissolution of PPN at m = 0.6-1.0 in comparison to other THP + water combinations studied. In addition, PPN dissolution was recorded as entropy-driven at m = 0.8-1.0 compared with other THP + water mixtures evaluated.Protein folding often requires molecular chaperones and folding catalysts, such as peptidyl-prolyl cis/trans isomerases (PPIs). The Escherichia coli cytoplasm contains six well-known PPIs, although a requirement of their PPIase activity, the identity of their substrates and relative enzymatic contribution is unknown. Thus, strains lacking all periplasmic and one of the cytoplasmic PPIs were constructed. Measurement of their PPIase activity revealed that PpiB is the major source of PPIase activity in the cytoplasm. Furthermore, viable Δ6ppi strains could be constructed only on minimal medium in the temperature range of 30-37 °C, but not on rich medium. To address the molecular basis of essentiality of PPIs, proteins that aggregate in their absence were identified. Next, wild-type and putative active site variants of FkpB, FklB, PpiB and PpiC were purified and in pull-down experiments substrates specific to each of these PPIs identified, revealing an overlap of some substrates. Substrates of PpiC were validated by immunoprecipitations using extracts from wild-type and PpiC-H81A strains carrying a 3xFLAG-tag appended to the C-terminal end of the ppiC gene on the chromosome. Using isothermal titration calorimetry, RpoE, RseA, S2, and AhpC were established as FkpB substrates and PpiC's PPIase activity was shown to be required for interaction with AhpC.The Western diet (WD) and hyperlipidemia are risk factors for vascular disease, dementia, and cognitive impairment. However, the molecular mechanisms are poorly understood. This pilot study investigated the genomic pathways by which the WD and hyperlipidemia regulate gene expression in brain microvessels. Five-week-old C57BL/6J wild type (WT) control and low-density lipoprotein receptor deficient (LDL-R-/-) male mice were fed the WD for eight weeks. Differential gene expression, gene networks and pathways, transcription factors, and non-protein coding RNAs were evaluated by a genome-wide microarray and bioinformatics analysis of laser-captured hippocampal microvessels. The WD resulted in the differential expression of 1972 genes. Much of the differentially expressed gene (DEG) was attributable to the differential regulation of cell signaling proteins and their transcription factors, approximately 4% was attributable to the differential expression of miRNAs, and 10% was due to other non-protein coding RNAs, primarily long non-coding RNAs (lncRNAs) and small nucleolar RNAs (snoRNAs) not previously described to be modified by the WD. Lipotoxic injury resulted in complex and multilevel molecular regulation of the hippocampal microvasculature involving transcriptional and post-transcriptional regulation and may provide a molecular basis for a better understanding of hyperlipidemia-associated dementia risk.This paper investigates the ability of steel fibers to enhance the short-term behavior and flexural performance of realistic steel fiber-reinforced concrete (SFRC) structural members with steel reinforcing bars and stirrups using nonlinear 3D finite element (FE) analysis. Test results of 17 large-scale beam specimens tested under monotonic flexural four-point loading from the literature are used as an experimental database to validate the developed nonlinear 3D FE analysis and to study the contributions of steel fibers on the initial stiffness, strength, deformation capacity, cracking behavior, and residual stress. see more The examined SFRC beams include various ratios of longitudinal reinforcement (0.3%, 0.6%, and 1.0%) and steel fiber volume fractions (from 0.3% to 1.5%). The proposed FE analysis employs the nonlinearities of the materials with new and established constitutive relationships for the SFRC under compression and tension based on experimental data. Especially for the tensional response of SFRC, an efficient smeared crack approach is proposed that utilizes the fracture properties of the material utilizing special stress versus crack width relations with tension softening for the post-cracking SFRC tensile response instead of stress-strain laws.
