Our results indicate that EAMT is a feasible approach and may improve upper extremity motor impairment, activities of daily living and kinematics after stroke. However, fully powered randomized clinical trials are warranted to confirm the results of this pilot study and explore the underlying mechanisms by which EAMT therapy might work.
To assess the effectiveness of group wheelchair maintenance training and investigate participant characteristics associated with responsiveness to training.
Randomized control trial with an immediate group and waitlist control group (WLCG) who received the intervention after a 6-month delay.
Four Spinal Cord Injury Model Systems Centers.
Manual (MWC) and power wheelchair (PWC) users with spinal cord injury (n=80 and 67, respectively).
Two 90-minute structured wheelchair maintenance training program classes with 12-20 people per class and separate classes for MWC and PWC users. Each class included in-person hands-on demonstrations and practice of wheelchair maintenance.
Separate analysis was completed for MWC and PWC users using the Wheelchair Maintenance Training Questionnaire (WMT-Q) capacity (ability to complete), performance (frequency of completion) and knowledge at baseline, 1-month, 6-month, 6-month-pre-training (WLCG only), and 1-year (immediate only).
Following the intervention, participapacity to complete maintenance and performance of maintenance activities for MWC and PWC users, even in a cohort of experienced wheelchair users. For MWC users improvements were tied to lower WMT-Q scores at baseline, while PWC users improved in capacity and performance independent of baseline score. Delivering this training in a structured group format is lower cost and therefore might improve adoption into clinical practice. As mitochondria are vulnerable to oxidative damage and represent the main source of reactive oxygen species (ROS), they are considered key tuners of ROS metabolism and buffering, whose dysfunction can progressively impact neuronal networks and disease. Defects in DNA repair and DNA damage response (DDR) may also affect neuronal health and lead to neuropathology. A number of congenital DNA repair and DDR defective syndromes, indeed, show neurological phenotypes, and a growing body of evidence indicate that defects in the mechanisms that control genome stability in neurons acts as aging-related modifiers of common neurodegenerative diseases such as Alzheimer, Parkinson's, Huntington diseases and Amyotrophic Lateral Sclerosis. In this review we elaborate on the established principles and recent concepts supporting the hypothesis that deficiencies in either DNA repair or DDR might contribute to neurodegeneration via mechanisms involving mitochondrial dysfunction/deranged metabolism.
Melanoma is an aggressive cancer with a rapidly increasing incidence rate worldwide. Acteoside has been shown to have antitumor effects in multiple human cancers; however, the underlying function and mechanisms of acteoside in melanoma remain unclear.
This study explored the inhibitory effect of acteoside on melanoma and the possible mechanisms.
Acteoside (15mg/kg, 30mg/kg) was administered to mice daily for 21 days. ICI182,780 (0.5mg/kg) was intraperitoneally injected 30min before acteoside administration three times a week to evaluate whether the effects elicited by acteoside were mediated via the estrogen receptor. Tumor growth and metabolism, cardiac function, ROS and apoptosis levels in the spleen, serum inflammatory factors, and immune cells in the spleen were monitored. STAT3, p-STAT3, CD31, and survivin levels in tumor tissues were measured via immunofluorescence. Ras, Raf1, STAT3, p-STAT3, Bcl-2, Bax, cleaved caspase-3, and cleaved caspase-9 levels in tumor tissues were determined via Western Blotting.
The results showed that acteoside inhibited melanoma growth, alleviated inflammation levels in mice, attenuated ROS and apoptosis levels in the spleen, downregulated the levels of CD31, survivin, Ras, Raf1, p-STAT3, and Bcl-2, and upregulated the levels of ERβ, Bax, cleaved caspase-3, and cleaved caspase-9. JAK Inhibitor I ic50 Moreover, the effect of acteoside was blocked by ICI182,780.
Acteoside may promote the apoptosis of tumor cells by regulating the ERβ-Ras/Raf1-STAT3 signaling axis, thus inhibiting the occurrence and development of melanoma.

The purpose of this study is to explore head and neck injuries secondary to accidents involving pogo sticks.
We conducted a 20-year cross-sectional study using the National Electronic Injury Submission System (NEISS). Head and neck injuries resulting from the use of a pogo-stick were included. The predictor variables were demographics, type, site, context of injury, and season in which the injury occurred. The primary outcome variable was admission rate, used to estimate the severity of the injury. We employed chi-squared and independent sample tests to determine whether an association existed or not. We conducted a multivariate logistic regression model to determine the odds of admission for all significant variables.
A total of 619 pogo stick injuries were included, with 96.1% under 18 years of age. The decrease in the number of injuries from the year 2000 to 2019 was significant (P=.003). Nearly all patients were under 18 (96.1%), with the remaining few being 18 or older. Laceration (43.8%) was the m (P < .05).
Head and neck injuries secondary to pogo sticks illustrated a predilection to the head. Fractures were the culprit behind the significantly increased odds of admission. Hence, all sectors of society should conceive different measures and implement them to protect against skull fractures (ie, helmet). Despite the decline in popularity of pogo sticks over the last two decades, all remaining riders should wear a helmet as religiously as bicycles or scooter riders to protect against skull fractures.
