C57BL/6 animals and Fpr1 KO mice were subjected to TBI and sacrificed 24 h or four weeks after injury. Twenty-four hours after injury, TBI Fpr1 KO mice showed reduced histological impairment, tissue damage and acute inflammation (MAPK activation, NF-κB signaling induction, NRLP3 inflammasome pathway activation and oxidative stress increase). Conversely, four weeks after TBI, the Fpr1 KO mice showed reduced survival of the proliferated cells in the Dentate Gyrus compared to the WT group. Behavioral analysis confirmed this trend. Moreover, TBI Fpr1 KO animals displayed reduced neural differentiation (evaluated by beta-III tubulin expression) and upregulation of astrocyte differentiation (evaluated by GFAP expression). Other parameters such as absorption coefficient, refractive index and optical dielectric constant were also explored.Herein, 15 phenylpiperazine 3-benzyl-5,5-dimethylhydantoin derivatives (1-15) were screened for modulatory activity towards Msr(A) efflux pump present in S. epidermidis bacteria. Synthesis, crystallographic analysis, biological studies in vitro and structure-activity relationship (SAR) analysis were performed. The efflux pump inhibitory (EPI) potency was determined by employing ethidium bromide accumulation assay in both Msr(A) efflux pump overexpressed (K/14/1345) and deficient (ATCC 12228) S. epidermidis strains. The series of compounds was also evaluated for the capacity to reduce the resistance of K/14/1345 strain to erythromycin, a known substrate of Msr(A). The study identified five strong modulators for Msr(A) in S. epidermidis. The 2,4-dichlorobenzyl-hydantoin derivative 9 was found as the most potent EPI, inhibiting the efflux activity in K/14/1345 at a concentration as low as 15.63 µM. Crystallography-supported SAR analysis indicated structural properties that may be responsible for the activity found. This study identified the first synthetic compounds able to inhibit Msr(A) efflux pump transporter in S. epidermidis. Thus, the hydantoin-derived molecules found can be an attractive group in search for antibiotic adjuvants acting via Msr(A) transporter.This pilot study focuses on the design, implementation, optimization and verification of a novel solution of smart measuring of water consumption and crisis detection leading to a smart water management platform. The system implemented consists of a modular IoT platform based on a PCB (Printed Circuit Board) design using the M2.COM standard, a LoraWAN modem and a LoraWAN gateway based on the Raspberry Pi platform. (industry partners distributing drinking water in the Olomouc and South-Moravian regions) to depict the current situation in the Czech Republic, characterized by extreme weather fluctuations and increasingly frequent periods of drought. These drinking water distributors are also constantly placing new demands on these smart solutions. These requirements include, above all, reliability of data transmission, modularity and, last but not least, low cost. However, smart water management (water consumption, distribution, system identification, equipment maintenance, etc.) is becoming an important topic worldwide. The functionality of the system was first verified in laboratory conditions and, then, in real operation. The study also includes checking signal propagation in the municipal area of the village of Zdarna, where the radius of the proposed measuring system was tested. A laboratory test with simulation of water leakage is also part of this work. Subsequently, the system was tested in a residential unit by means of water leakage detection using the MNF method (minimum night flow); the detection success rate was 95%.
