The studies were reviewed with regard to the investigated splicing regulatory events and the experimental strategy to construct and perform a splicing reporter minigene assay. We further elaborate on how analyses on the regulation of RNA splicing offer promising prospects for gaining important insights into disease mechanisms.Mycobacterium tuberculosis (MTB), the causative agent of tuberculosis (TB), avoids the host immune system through its virulence factors. MPT63 and MPT64 are the virulence factors secreted by MTB which regulate host proteins for the survival and proliferation of MTB in the host. Here, we found that MPT63 bound directly with TBK1 and p47phox, whereas MPT64 interacted with TBK1 and HK2. We constructed a MPT63/64-derived multifunctional recombinant protein (rMPT) that was able to interact with TBK1, p47phox, or HK2. rMPT was shown to regulate IFN-β levels and increase inflammation and concentration of reactive oxygen species (ROS), while targeting macrophages and killing MTB, both in vitro and in vivo. Furthermore, the identification of the role of rMPT against MTB was achieved via vaccination in a mouse model. Taken together, we here present rMPT, which, by regulating important immune signaling systems, can be considered an effective vaccine or therapeutic agent against MTB.Luminescent inorganic materials are used in several technological applications such as light-emitting displays, white LEDs for illumination, bioimaging, and photodynamic therapy. Low ionic diffusion and high melting points of the precursors lead to long processing times in these solid-state syntheses with a cost in energy consumption when conventional heating methods are applied. Microwave-assisted synthesis relies on selective, volumetric heating attributed to the electromagnetic radiation interaction with the matter. The microwave heating allows for rapid heating rates and small temperature gradients yielding homogeneous, well-formed materials swiftly. Luminescent inorganic materials can benefit significantly from the microwave-assisted synthesis for high homogeneity, diverse morphology, and rapid screening of different compositions. The rapid screening allows for fast material investigation, whereas the benefits of enhanced homogeneity include improvement in the optical properties such as quantum yields and storage capacity.Density estimation of pelagic fish was performed by means of single beam echosounding in 17 lakes within a period of 34 years, from 1985 to 2018. Surveys were performed repeatedly (two to fourteen times) in five lakes. The density estimates ranged from 34 to 4720 fish/ha and were significantly correlated with total phosphorus concentration. The high density in relatively phosphorus rich lakes (TP > 10 µg/L) was comprised of small fish ( less then 20 cm) and was partly due to the higher number of pelagic fish species. The number of pelagic species varied from one, Arctic charr, in the most elevated and oligotrophic lakes, and whitefish dominated in less elevated oligotrophic lakes. In lowland lakes characterized as mesotrophic or tending to mesotrophy, smelt, vendace, and two to three cyprinids comprised the pelagic fish stock. These fish species predate zooplankton effectively, and species composition and body size of planktonic cladocerans was affected by fish density. Large species of Daphnia were lacking in lakes with high fish density, and body size of present species, D. galeata, D. cristata, and Bosmina spp. were negatively correlated with pelagic fish density.We report the development of a laser gas analyzer that measures gas concentrations at a data rate of 100 Hz. This fast data rate helps eddy covariance calculations for gas fluxes in turbulent high wind speed environments. The laser gas analyzer is based on derivative laser absorption spectroscopy and set for measurements of water vapor (H2O, at wavelength ~1392 nm) and carbon dioxide (CO2, at ~2004 nm). This instrument, in combination with an ultrasonic anemometer, has been tested experimentally in both marine and terrestrial environments. First, we compared the accuracy of results between the laser gas analyzer and a high-quality commercial instrument with a max data rate of 20 Hz. We then analyzed and compared the correlation of H2O flux results at data rates of 100 Hz and 20 Hz in both high and low wind speeds to verify the contribution of high frequency components. The measurement results show that the contribution of 100 Hz data rate to flux calculations is about 11% compared to that measured with 20 Hz data rate, in an environment with wind speed of ~10 m/s. Therefore, it shows that the laser gas analyzer with high detection frequency is more suitable for measurements in high wind speed environments.Adverse food reactions include immune-mediated food allergies and non-immune-mediated intolerances. Special emphasis is placed on specific nutritional concerns for each of these conditions from the combined point of view of gastroenterology and immunology, in an attempt to offer a useful tool to practicing physicians in discriminating these diverging disease entities and planning their correct management. We conclude that a correct diagnostic approach and dietary control of both immune- and non-immune-mediated food-induced diseases might minimize the nutritional gaps in these patients, thus helping to improve their quality of life and reduce the economic costs of their management.
