Diabetic nephropathy is one of the most important complications in patients with diabetes. The etiology and pathogenesis of diabetic nephropathy remain unclear. Several studies have indicated that circular RNAs (circRNAs) play crucial regulatory roles in numerous human diseases and normal physiology; however, to date, no study has focused on the comprehensive expression profile of circRNAs in the kidneys of diabetic mice. Therefore, we aimed to identify differentially expressed circRNAs in diabetic mouse kidneys to explore the possible roles of dysregulated circRNAs in diabetic nephropathy development.

/Nju (BKS-DB/Nju) mice and their nondiabetic wild-type littermates of C57BL/KsJ wild-type (WT) mice were used as experimental animals. Epibrassinolide Among all circRNAs identified by high-throughput RNA sequencing, four circRNAs were upregulated and ten were downregulated in diabetic mouse kidneys compared to those in nondiabetic mouse kidneys. After verification using quantitative reverse transcriptase polymerase chain reaction assays, we found that circR_1084, circR_182, circR_4, circR_50, circR_596, circR_897, and circR_203 were downregulated, whereas circR_627, circR_628, circR_735, and circR_801 were upregulated in the renal tissues of 8- and 16-week-old BKS-DB/Nju mice compared to those in WT mice.

Bacterial plasmids often carry antibiotic resistance genes and are a significant factor in the spread of antibiotic resistance. The ability to completely assemble plasmid sequences would facilitate the localization of antibiotic resistance genes, the identification of genes that promote plasmid transmission and the accurate tracking of plasmid mobility. However, the complete assembly of plasmid sequences using the currently most widely used sequencing platform (Illumina-based sequencing) is restricted due to the generation of short sequence lengths. The long-read Oxford Nanopore Technologies (ONT) sequencing platform overcomes this limitation. Still, the assembly of plasmid sequence data remains challenging due to software incompatibility with long-reads and the error rate generated using ONT sequencing. Bioinformatics pipelines have been developed for ONT-generated sequencing but require computational skills that frequently are beyond the abilities of scientific researchers. To overcome this challenge, theine suitable for accurate plasmid assembly and the detection of anti-microbial resistance genes in (clinical) samples where multiple plasmids can be present.The ability to confidently perform fall-risk assessment on older adults is critical for Doctor of Physical Therapy (DPT) students prior to entering workforce. The complex nature of falls makes it challenging to teach it realistically in traditional classroom settings. This could lead to lack of confidence in performing effective assessments in real clinical situations. For this purpose, an evidence-based experiential fall-risk assessment activity was implemented in the curriculum. The purpose was to investigate if this activity improved students' confidence in performing fall-risk assessment. Twenty-eight students completed this activity on thirty-three older adults from a senior living community. A 13-item questionnaire was used to investigate confidence before and after the activity. Significant improvements in students' confidence were noted for administering client interview (p = .001, r = -0.43), 30-Second Chair Stand Test (p = .046, r = -0.34) and 10-Meter Walk Test (p = .011, r = -0.27). Additionally, students demonstrated excellent inter-rater reliability (ICC > 0.9) with the faculty experts for administering 5-Times Sit-to-Stand, 10-Meter Walk, Berg Balance Scale, 4-Stage Balance, Timed Up and Go and 30-Second Chair Stand tests, and good inter-rater reliability (ICC = 0.78) for Single-Limb Stance Time test. This activity had a positive impact on DPT students' confidence in conducting effective fall-risk assessment.The purposes of this study were to analyze Latino/a immigrant mushroom workers' perceptions of how the workplace environment shapes occupational safety and health, examine whether and how those perceptions differ by gender, and identify future areas for research on occupational safety and health in the mushroom industry. Researchers conducted structured interviews with 15 women and 45 men on 6 Pennsylvania mushroom farms to obtain their descriptions and perspectives of safety and health risk factors in their workplaces. Approximately one third of respondents had suffered an injury at work, and nearly half felt that there are workplace factors that affect their health and safety. The study found that Latino/a mushroom farmworkers perceive risks that are posed by the indoor infrastructure of mushroom production houses, including poorly maintained wooden walkways and cool indoor temperatures, and by the organization of mushroom production work, including the application of chemicals including pesticides, physical demands of the job, use of small knives, contact with compost, and the piece rate payment system. Workers commonly discussed back pain and believed it was associated with the organization of work. Women in the sample were more likely to be concerned about slips and falls than men and less likely to be concerned about aches and pains. Mushroom farm infrastructure and the specific demands of the jobs pose occupational safety and health risks to Latino/a farmworkers that merit further study to develop adequate public health interventions. Future research should obtain gender-disaggregated objective reports of injury, aches and pains, and discomfort and test for relationships between these reports and the indoor infrastructure and conditions of mushroom production work.
