These may be difficult to treat because of resistance-associated substitutions (RAS) emerging after treatment failure. Triple therapy with sofosbuvir (SOF)/velpatasvir (VEL)/voxilaprevir (VOX) is the recommended retreatment after DAA-based failure. However, in rare cases, failure to triple therapy occurs, and there is little information characterizing the viruses that relapse. To determine the RAS profile after failing SOF/VEL/VOX, and seek suitable alternatives for retreatment, samples from 5 patients were analysed using MiSeq Illumina deep sequencing before and after triple therapy. All patients were men, aged 59-78 years, 2 HCV genotype (G) 1b and 3 G3a. The most prevalent NS3 substitutions after SOF/VEL/VOX failure were Y56F and A166T. Four patients had the NS5A RAS, Y93H, after triple failure, and Y93H was observed in both G1b patients before retreatment and after SOF/ledipasvir failure. In 2 G3a patients, Y93H appeared at triple failure, and on the other G3a, A30K persisted in 100% of viral genomes. Finally, G1b patients showed C316N in NS5B, associated with SOF failure, but G3a patients had no known NS5B substitutions. HCV RAS analysis identified the following substitutions present at higher rates after triple failure Y56F in NS3 (G1b), A166T in NS3 (G3a), A30K or Y93H in NS5A, and C316N in NS5B (G1b). This is followed by a section highlighting special considerations when applying the methods to primates, and a practical guide to facilitate further landscape genetics studies using both existing and de novo datasets. We conclude by exploring future avenues of inquiry that could be facilitated by recent developments as well as underdeveloped applications of landscape genetics to primates.A conventional sequencing batch reactor (SBR) was upgraded using fixed biofilm carriers with a specific surface area around 18 m2 m-3 . The upgraded SBR was investigated to remove phenol from high strength wastewater operated under various operational conditions. The operational conditions used were variable volume exchange ratio (VER) up to 75%, hydraulic retention time (HRT) from (10.7-21.3 hr), aeration time (from 2 to 8 hr), and initial phenol concentration up to 600 mg L-1 . It was found that the upgraded SBR increased the removal efficiencies of biological oxygen demand (BOD5 ), chemical oxygen demand (COD), and total suspended solids (TSS) by about 10% using high strength wastewater without phenol compared to SBR. Furthermore, the removal rate of phenol for the upgraded SBR was higher than conventional SBR by about 18% at 600 mg L- of initial phenol concentration under the same operational conditions. Compared to the conventional SBR, the upgraded version reduced the aeration step by 25% and achieved higher removal efficiency of phenol. In the animal experiments, we found persistent activation of autophagy in the development of pulmonary fibrosis, which was also accompanied by tumor necrosis factor and transforming growth factor expression increased. Therefore, the autophagy signaling pathway may regulate the inflammatory response and affect the progression of fibrosis. Further, in vitro experiments, we used LY294002, RAPA, and N-acetylcysteine (NAC) intervened autophagy. Our results showed that PI3K/Akt/mTOR signaling pathway is involved in the autophagy changed mediated by SiO2 exposed, and autophagy might play a protective role in the progression of pulmonary fibrosis.
