Sweet-potato breeding programs need to integrate sensory characterization and hedonic perception to provide global markets with widely accepted products. The present work aimed to characterize a series of purple-skin sweet potatoes differing in flesh coloration (from pale yellow to slight orange) from a sensory and physicochemical perspective. Uruguayan consumers' perceptions of the genotypes were also explored. Special focus was placed on the influence of flesh color on consumers' expectations and hedonic perception of purple-skin sweet potatoes.
Sensory characterization based on projective mapping showed that the evaluated genotypes largely differed in their sensory characteristics in terms of flavor, texture, and color. Soluble solids and the concentration of individual sugars were not good predictors of sensory quality. In the appearance evaluation, consumers associated orange flesh with positive expected sensory characteristics and stronger liking. Although there are 4 types of IPMN, among which intestinal-type IPMN is likely to progress into invasive cancer known as colloid carcinoma, no information regarding the involvement of the intestinal phenotype in the carcinogenesis of IPMN exists. The present study was conducted to explore how the intestinal differentiation system is maintained during the tumor progression of intestinal-type IPMN using surgical resection specimens. Results showed that Atoh1, a critical transcriptional factor for intestinal differentiation toward the secretory lineages of intestinal epithelial cells, was expressed in an invasive-grade IPMN. To determine the function of Atoh1 in pancreatic cancer, we generated a pancreatic ductal adenocarcinoma (PDAC) cell line overexpressing Atoh1. In a xenograft model, we successfully induced an IPMN phenotype in PDAC cells via Atoh1 induction. Finally, for the first time, we discovered that GPA33 is expressed in intestinal-type IPMN, thereby suggesting a novel target for cancer therapy. In conclusion, the intestinal differentiation system might be maintained during tumor progression of intestinal-type IPMN. Further analysis of the function of Atoh1 in IPMN might be useful for understanding the molecular mechanism underlying the malignant potential during the tumor progression of IPMN.The standard technique for pancreas preservation for transplantation is static cold storage (SCS). In this experimental study, we compare SCS to hypothermic machine perfusion (HMP) of the pancreas to assess if the latter could safely prolong the ischaemia period prior to transplantation. We worked in two phases, first with organ preservation for 24 h and second, preservation for either 2 or 6 h before allotransplantation. In phase 1, exocrine injury markers were found to be nonsignificantly lower, in the HMP group (n = 3) vs. SCS (n = 3) after 24 h of preservation; amylase (P = 0.2), lipase (P = 0.3) and lactate dehydrogenase (P = 0.1). In phase 2, 14 recipient diabetic pigs (after total pancreatectomy) received allotransplantations with n = 4 and n = 4 pancreases after HMP for 2 and 6 h vs. n = 3 and n = 3 pancreases after SCS for 2 and 6 h, respectively. There were no differences in recipient survival (P = 0.7), and mean survival was 14 days (0-53 days). All recipients had allograft function defined as detectable C-peptide and independent normoglycemia. We have not highlighted vascular thrombosis in all allotransplantations. This study reports the first successful pancreas allotransplantation after HMP preservation for up to 6 h with no evidence of graft thrombosis.
Chili is one of the most important and high-value vegetable crops worldwide. However, pest and disease infections are among the main limiting factors in chili cultivation. These diseases cannot be eradicated but can be handled and monitored to mitigate the damage. Hence, the use of an automated identification system based on images will promote quick identification of chili disease. The features extracted from the images are of utmost importance to develop such an accurate identification system.
In this research, chili pest and disease features extracted using the traditional approach were compared with features extracted using a deep-learning-based approach. A total of 974 chili leaf images were collected, which consisted of five types of diseases, two types of pest infestations, and a healthy type. Six traditional feature-based approaches and six deep-learning feature-based approaches were used to extract significant pests and disease features from the chili leaf images. find more The extracted features were fed into three machine learning classifiers, namely a support vector machine (SVM), a random forest (RF), and an artificial neural network (ANN) for the identification task. The results showed that deep learning feature-based approaches performed better than the traditional feature-based approaches. The best accuracy of 92.10% was obtained with the SVM classifier.
