Background Platelet refractoriness remains a challenging clinical dilemma although significant advancements have been made in identifying human leukocyte antigen (HLA) matched or HLA compatible units. Antiplatelet antibodies are the major risk factor for immune-mediated platelet refractoriness, yet the role of antibody-initiated complement-mediated platelet destruction remains poorly understood. Study design and methods Human complement-mediated opsonization and killing of platelets was assayed ex vivo using antibody-sensitized human platelets incubated with complement-sufficient human sera. A new animal model of platelet refractoriness utilizing Wistar rats transfused with human platelets is described. Results Human platelets sensitized with anti-platelet antibodies were rapidly opsonized with iC3b upon incubation in human sera. This opsonization could be completely blocked with a classical pathway complement inhibitor, PA-dPEG24. Complement activation decreased platelet viability, which was also reversible with complement inhibitor PA-dPEG24. A new rat model of platelet refractoriness was developed that demonstrated some platelet removal from the blood stream was complement mediated. Conclusions Complement activation initiated by anti-platelet antibodies leads to complement opsonization and decreased platelet viability. A new rat model of platelet refractoriness was developed that adds a new tool for elucidating the mechanisms of platelet refractoriness.Hypothesis While the pinch-off dynamics of bubbles is known to be influenced by changes in surface tension, previous studies have only assessed changes due to liquid properties or surfactant effects at the air-liquid interface but not due to the presence of particles. The current study proposes that particles at the air-liquid interface play an important role in changing the surface tension and thus the pinch-off dynamics of particle-laden bubbles. Experiments High-speed photography was used to study the pinch-off dynamics of air bubbles coated by a monolayer of silica microparticles. The influence of bubble surface coverage and particle size classes on the bubble pinch-off dynamics were explored. Findings We identify that although the scaling exponent of the power law that governs the pinch-off of coated and uncoated bubbles is the same, the pinch-off dynamics is distinctly different when particles are present at the air-liquid interface due to a decrease in surface tension with time in the neck region. We suggest that the surface pressure generated by particle interaction reduces the pinch-off speed by reducing the apparent surface tension. We observe that the apparent surface tension is dependent on particle size but not on the percentage of bubble surface coated by particles.SIRT1, a class III histone/protein deacetylase (HDAC) has been associated with autoimmune diseases. There is a paucity of data about the role of SIRT1 in Graves' disease. The aim of this study was to investigate the role of SIRT1 in the pathogenesis of GD. Here we showed that SIRT1 expression and activity were significantly decreased in GD patients compared with healthy controls. The NF-κB pathway was activated in the peripheral blood of GD patients. The reduced SIRT1 levels correlated strongly with clinical parameters. In euthyroid patients, SIRT1 expression was markedly upregulated, and NF-κB downstream target gene expression was significantly reduced. SIRT1 inhibited the NF-κB pathway activity by deacetylating p65. These results demonstrate that reduced SIRT1 expression and activity contribute to the activation of the NF-κB pathway and may be involved in the pathogenesis of GD.Many different biofabrication approaches as well as a variety of bioinks have been developed by researchers working in the field of tissue engineering. A main challenge for bioinks often remains the difficulty to achieve shape fidelity after printing. In order to overcome this issue, a homogeneous pre-crosslinking technique, which is generally applicable to all alginate-based materials, was developed in this study. With this technique it was possible to markedly enhance the printability of a 2 % (w/v) alginate solution, without using a higher polymer content, fillers or support structures. Results Morphosyntactic score was affected negatively by the chronological age, on the other hand, it was not affected by the age of implantation, the gender, or the duration of using cochlear implant.
