Objective was to elucidate the effects of heat stress (HS) on embryo development during first 16 gestational days (GD) and circulating hormone concentrations on GD-16 in lactating Holstein cows. Cows in HS and control (CON) groups were exposed to temperature humidity index (THI) of ≥ 73 and  less then  73, respectively, for 3 weeks before the experiment. GD-7 (67 vs 49%) and GD-16 (52 vs. 31%) conception rates following single insemination were greater (P  less then  0.01) for CON compared with HS cows. Control cows produced more GD-7 transferrable embryos following superovulation compared with HS cows (84.8 vs 53.1%; P  less then  0.001). Mean (± SEM) length (45.2 ± 10.6 vs. 59.2 ± 9.1 mm) and weight (31.4 ± 4.3 vs. 42.4 ± 6.2 mg) of GD-16 conceptus were greater for CON compared with HS cows (P  less then  0.05). Control cows yielded more filamentous conceptus (≥ 25 mm) compared with HS cows (71 vs 45%; P  less then  0.05). Progesterone (2.09-fold) was higher, and cortisol (1.86-fold), prolactin (1.60-fold), substance-P (1.55-fold), Isoprostane-8 (1.34-fold) and prostaglandin F metabolites (1.97-fold) were lower in CON compared with HS cows (P  less then  0.05). Progesterone positively, and substance-P, isoprostane-8 and the THI negatively were associated with GD-16 conceptus length (P  less then  0.05). In conclusion, altered hormones concentrations in heat-stressed cows plausibly resulted in lower GD-7 and GD-16 conception rates, fewer GD-7 transferable embryos, and stunted GD-16 conceptus elongation.The removal and chemical speciation changes of heavy metals in the sewage sludge during the single bioleaching and combined bioleaching/Fenton-like processes were compared in this study. The improvement in the dewaterability of the treated sludge was also investigated. The single bioleaching led to a removal of Zn, Cu, Cd, Cr, Mn, Ni, As and Pb of 67.28%, 50.78%, 64.86%, 6.32%, 56.15%, 49.83%, 20.78% and 10.52% in 10 days, respectively. The chemical speciation analysis showed that the solubilization of heavy metals in mobile forms (exchangeable/acid soluble and reducible forms) and oxidizable form was the main reason for their removal. Subsequent Fenton-like treatment was carried out at different bioleaching stages when the bioleached sludge dropped to certain pH values (4.5, 4.0 and 3.0), by adding H2O2 at different dosages. The highest removal ratio of Zn, Cu, Cd, Cr, Mn and Ni could reach 75.53%, 52.17%, 71.91%, 11.63%, 66.29% and 65.19% after combined bioleaching/Fenton-like process, respectively, with appropriate pH and H2O2 dosages in less than 6 days. The solubilization efficiencies of these heavy metals in mobile forms were further improved by Fenton-like treatment. The removal efficiencies of As and Pb decreased due to their transformation into insoluble forms (mostly residual fraction) after Fenton treatment. The capillary suction times (CST) of the raw sludge (98.7 s) decreased by 79.43% after bioleaching and 87.44% after combined process, respectively.An electromagnetic (EM) pulse falling on a plasma medium from vacuum can either reflect, get absorbed or propagate inside the plasma depending on whether it is overdense or underdense. In a magnetized plasma, however, there are usually several pass and stop bands for the EM wave depending on the orientation of the magnetic field with respect to the propagation direction. The EM wave while propagating in a plasma can also excite electrostatic disturbances in the plasma. In this work Particle-In-Cell simulations have been carried out to illustrate the complete transparency of the EM wave propagation inside a strongly magnetized plasma. The external magnetic field is chosen to be perpendicular to both the wave propagation direction and the electric field of the EM wave, which is the X mode configuration. Despite the presence of charged electron and ion species the plasma medium behaves like a vacuum. The observation is understood with the help of particle drifts. It is shown that though the two particle species move under the influence of EM fields their motion does not lead to any charge or current source to alter the dispersion relation of the EM wave propagating in the medium. Furthermore, it is also shown that the stop band for EM wave in this regime shrinks to a zero width as both the resonance and cut-off points approach each other. Thus, transparency to the EM radiation in such a strongly magnetized case appears to be a norm.It is well established that genetics, environment, and interplay between them play a crucial role in adult disease. We aimed to evaluate the role of genetics, early life nutrition, and the interaction between them, on optimal adult health. As part of a large international consortium (n ~ 154,000), we identified 60 SNPs associated with both birthweight and adult disease. Utilising the Raine Study, we developed a birthweight polygenic score (BW-PGS) based on the 60 SNPs and examined relationships between BW-PGS and adulthood cardiovascular risk factors, specifically evaluating interactions with early life nutrition. This provides an example of how precision medicine in early life can improve adult health.Tempol (4-hydroxy-2,2,6,6-tetramethylpiperidine-1-oxyl) is a potential redox agent in cells. The present study investigated changes in cellular reactive oxygen species (ROS) and glutathione (GSH) levels and in antioxidant enzymes, in Tempol-treated Calu-6 and A549 lung cancer cells, normal lung WI-38 VA-13 cells, and primary pulmonary fibroblasts. Results demonstrated that Tempol (0.5-4 mM) either increased or decreased general ROS levels in lung cancer and normal cells at 48 h and specifically increased O2•- levels in these cells. In addition, Tempol differentially altered the expression and activity of antioxidant enzymes such as superoxide dismutase, catalase, and thioredoxin reductase1 (TrxR1) in A549, Calu-6, and WI-38 VA-13 cells. In particular, Tempol treatment increased TrxR1 protein levels in these cells. Tempol at 1 mM inhibited the growth of lung cancer and normal cells by about 50% at 48 h but also significantly induced cell death, as evidenced by annexin V-positive cells. Furthermore, down-regulation of TrxR1 by siRNA had some effect on ROS levels as well as cell growth inhibition and death in Tempol-treated or -untreated lung cells.
