To develop and characterize a new type of plasma rich in growth factors (PRGF) membrane for patients in which immune system is involved in the disease etiology.
Blood from three healthy donors was collected to obtain the different fibrin membranes by PRGF technology. PRGF obtained volumes were activated and divided into two groups PRGF membrane (mPRGF) obtained after incubation at 37°C for 30min (control); and is-mPRGF mPRGF obtained after incubation for 30min at 56°C. The concentration of several growth factors, proteins, immunoglobulin E and the complement activity was determined in the different mPRGF. The proliferative potential of heat-inactivated mPRGF were assayed on keratocytes (HK) and conjunctival fibroblasts (HConF). In addition, morphological and physical features of the inactivated mPRGF were evaluated in contrast to the control mPRGF.
Heat-inactivation of the mPRGF preserves the content of most of the growth factors involved in the ocular wound healing while reducing drastically the contenpreserved after heat-inactivation.In women, aging leads to reduced hair density and thinner fibers and can result in female-pattern hair loss. However, the impact of the aging dermal environment on female scalp hair follicles remains unclear. In this study, we document in situ changes in 22 women (aged 19-81 years) and primary cultures of dermal fibroblast and dermal sheath cells. In situ, the papillary reticular boundary was indistinguishable in the young scalp but prominent in the scalp of those aged >40 years, accompanied by reduced podoplanin (PDPN) expression, increased versican expression, and changes in collagen organization. Hair follicles were shorter, not reaching the adipose layer. Hyaluronic acid synthase 2 was highly expressed, whereas matrix metalloproteinase 1 was elevated in the dermal papilla and dermal sheath in situ. Primary dermal fibroblast cultures confirmed that matrix metalloproteinase 1 mRNA, MMP1, increased with aging, whereas in dermal sheath cells, hyaluronic acid synthase 2, HAS2, and PDPN increased and α-smooth muscle actin αSMA mRNA decreased. Both exhibited increased cartilage oligomeric protein, COMP mRNA expression. Proteomics revealed an increase in dermal sheath proteins in the dermal fibroblast secretome with aging. In summary, aging female scalp shows striking structural and biological changes in the hair follicle environment that may impact hair growth.
Lung cancer risk prediction models do not routinely incorporate imaging metrics available on low-dose CT (LDCT) imaging of the chest ordered for lung cancer screening.
What is the association between quantitative emphysema measured on LDCT imaging and lung cancer incidence and mortality, all-cause mortality, and airflow obstruction in individuals who currently or formerly smoked and are undergoing lung cancer screening?
In 7,262 participants in the CT arm of the National Lung Screening Trial, percent low attenuation area (%LAA) was defined as the percentage of lung volume with voxels less than -950 Hounsfield units on the baseline examination. Multivariable Cox proportional hazards models, adjusting for competing risks where appropriate, were built to test for association between %LAA and lung cancer incidence, lung cancer mortality, and all-cause mortality with censoring at 6 years. In addition, multivariable logistic regression models were built to test the cross-sectional association between %LAA and leveraged to improve lung cancer risk prediction and help diagnose COPD in individuals who currently or formerly smoked and are undergoing lung cancer screening.
Our previous work identified an aberrant expression of hsa_circ_0001946 in coronary atherosclerotic heart disease (CHD). Here we aimed to verify the role of hsa_circ_0001946 as a biomarker for CHD, and explore the clues of its downstream regulation.
The hsa_circ_0001946 expression in CHD patients (n=120) and controls (n=120) were confirmed with qRT-PCR. BLU-945 solubility dmso CircBank and miRDB were used for target analysis in silico. Spearman correlation test was performed to infer potential interrelationships among the nucleic acid molecular biomarkers, and their predictive abilities were examined using receiver operating characteristic (ROC) curves.
Hsa_circ_0001946 was validated to be significantly up-regulated in the peripheral blood mononuclear cells of CHD patients, and revealed as an independent indicator of increased CHD risk (odds ratio 2.364; 95% confidence interval [CI] 1.765-3.165) after adjusting for confounding factors. Hsa-miR-7-5p was found to own the largest number of binding sites in has_circ_0001946 sequence, and among its targets predicted, the poly ADP-ribose polymerase 1 (PARP1) has been implicated in the pathophysiology of CHD. Spearman analysis indicated negative correlations of hsa-miR-7-5p with hsa_circ_0001946 and PARP1, respectively; while hsa_circ_0001946 was positively correlated with PARP1. The prediction accuracy of hsa_circ_0001946 in CHD was evaluated, showing an area under the ROC curve of 0.897 (95% CI 0.791-0.961), which could further increase to 0.957 (95% CI 0.870-0.992) upon a combination of hsa-miR-7-5p and PARP1.
The present work demonstrated the predictive power of hsa_circ_0001946, hsa-miR-7-5p and PARP1 as combined biomarkers for CHD, and suggests a regulatory axis they consisted might contribute to the CHD development.

Iron deficiency (ID) is an important predictor of adverse outcomes in patients with heart failure, however it is unclear whether ID also affects prognosis in patients with acute coronary syndrome (ACS). The aim of this systematic review and meta-analysis was to assess the prognostic value of iron deficiency in patients with ACS.
We searched PubMed, Web of Science, and the Cochrane library and included cohort studies of patients with ACS that were stratified by ID status. There were no restrictions on definition of ACS or ID. Studies were systematically appraised and data extracted by two independent reviewers. Meta-analysis was performed where two or more studies reported on the same pre-determined outcome measure.
Seven studies with 2821 participants were identified, reporting a high prevalence of ID in the ACS population. Three studies reported worse long-term outcomes in the ID population, whereas short-term outcomes were heterogeneous across studies.
Patients with ID presenting with ACS may have a worse long-term prognosis but more studies are required for confirmation.
