The importance of policy for promoting physical activity (PA) is increasingly recognized by academics, and there is a push by national governments and international institutions for PA policy development and monitoring. However, our knowledge about which policies are actually effective to promote PA remains limited. This article summarizes the currently available evidence by reviewing existing reviews on the subject. Building on results from a previous scoping review on different types of PA-related evidence, we ran searches for combinations of the terms "physical activity", "evidence", "effect", "review", and "policy" in six different databases (PubMed, Scopus, SportDiscus, PsycInfo, ERIC, and IBSS). We used EPPI Reviewer 4 to further process the results and conduct an in-depth analysis. We identified 57 reviews providing evidence on 53 types of policies and seven broader groups of policies. Reviews fell into four main categories 1) setting- and target group-specific; 2) urban design, environment and transport; 3) economic instruments; and 4) broad-range perspective. Results indicate that there is solid evidence for policy effectiveness in some areas (esp. school-based and infrastructural policies) but that the evidence in other areas is insufficient (esp. for economic policies). The available evidence provides some guidance for policy-makers regarding which policies can currently be recommended as effective. However, results also highlight some broader epistemological issues deriving from the current research. This includes the conflation of PA policies and PA interventions, the lack of appropriate tools for benchmarking individual policies, and the need to critically revisit research methodologies for collating evidence on policies. © 2020 The Authors.A comprehensive series of optimization studies including pH, solvent and temperature were completed on the nitrile hydrolyzing Rhodococcus erythropolis bacterium SET1 with the substrate 3-hydroxybutyronitrile. These identified temperature of 25 °C and pH of 7 as the best conditions to retain enantioselectivity and activity. The effect of the addition of organic solvents to the biotransformation mixture was also determined. The results of the study suggested that SET1 is suitable for use in selected organo-aqueous media at specific ratios only. The functional group tolerance of the isolate with unprotected and protected β-aminonitriles, structural analogues of β-hydroxynitriles was also investigated with disappointingly poor isolated yields and selectivity obtained. The isolate was further evaluated with the α- aminonitrile phenylglycinonitrile generating acid in excellent yield and ee (>99 % (S) - isomer and 50 % yield). KGaA.This work investigated the properties of Paracoccus yeei VKM B-3302 bacteria isolated from activated sludge and immobilized in an N-vinylpyrrolidone-modified poly(vinyl alcohol) matrix. The developed hydrogel formed a network structure to enable the entrapment of microbial cells with their viability and biocatalytic properties preserved, which ensured the technological possibility of replicating expendable biosensor receptor elements. A new ratio of the components for the synthesis selected in this work enabled producing a copolymer of an earlier undescribed chemical structure, which can be efficiently used for immobilization of highly sensitive P. yeei bacteria. A biological oxygen demand (BOD) biosensor with these bacteria and matrix was shown to possess a long-time stability exceeding that described earlier, to have a broad substrate specificity and to exceed approximately tenfold the nearest analogues by its sensitivity and the lower boundary value of 0.05 mg/dm3. The biosensor enabled assays of water samples initially attributed to pure samples (the BOD range, 0.05-5.0 mg/dm3). BOD assays of water samples from various sources showed the use of the receptor element of this composition to enable the data that closely correlated with the standard method (R 2 = 0.9990). © King Abdulaziz City for Science and Technology 2020.MicroRNAs are endogenous small non-coding-RNAs that control gene expression and cancer development. Previous studies reported that Polyalthia longifolia treatment induced apoptotic cell death in HeLa cells by down-regulation of miR-221-5p. Hence, the current study was conducted to validate the down-regulated miR-221-5p in HeLa cells. Functional analysis of miR-221-5p was conducted through the gain-of-function, and loss-of-function approach and the miRNA expression was quantified by a real-time polymerase chain reaction. Trilaciclib clinical trial The P. longifolia treatment significantly (p less then 0.05) reduced miR-221-5p expression when compared to the untreated HeLa cells with a double delta Ct value of 6.32 and the expression fold change value was reduced up to 0.013. The transfection of miR-221-5p mimic significantly increased the expression of miR-221-5p with an expression fold change as high as 0.53 while anti-miR-221-5p transfected HeLa cells show the most significant decrease in miR-221-5p expression with an expression fold change of 0.011. The MTT assay results revealed that the over-expression of miR-221-5p increased the cell proliferation and viability of polyphenol-rich P. longifolia-treated HeLa cells and confirmed the role of downregulated miRNA 221-5p in HeLa cell death. The flow-cytometry analysis showed that the miR-221-5p over-expressed cells decreased the apoptosis of cells induced by polyphenol-rich P. longifolia treatment in HeLa cells, which proved the oncogenic role of miR-221-5p to inhibit apoptosis. Moreover, the depletion of caspase-3 in miR-221-5p-overexpressed HeLa cells showed the roles of downregulated miR-221-5p in the induction of apoptosis. In conclusion, these results suggest that the down-regulated miR-221-5p was involved in regulating apoptosis in HeLa cancer cells. © King Abdulaziz City for Science and Technology 2020.
