Among brain tumors, Medulloblastoma (MB) is one of the most common, malignant, pediatric tumors of the cerebellum. It accounts for ~20% of all childhood central nervous system (CNS) tumors. We also highlight the latest treatment strategies available for medulloblastoma.Previous studies have demonstrated that transcription factor Etv5 plays an important role in the segregation between epiblast and primitive endoderm at the second fate decision of early embryo. However, it remains elusive whether Etv5 functions in the segregation between inner cell mass and trophectoderm at the first cell fate decision. In this study, we firstly generated Etv5 knockout mouse embryonic stem cells (mESCs) by CRISPR/Cas9, then converted them into extended potential stem cells (EPSCs) by culturing the cells in small molecule cocktail medium LCDM (LIF, CHIR99021, (S)-(+)-dimethindene maleate, minocycline hydrochloride), and finally investigated their differentiation efficiency of trophoblast stem cells (TSCs). The results showed that Etv5 knockout significantly decreased the efficiency of TSCs (CDX2+) differentiated from EPSCs. In addition, Etv5 knockout resulted in higher incidence of the differentiated cells with tetraploid and octoploid than that from wild type. Mechanistically, Etv5 was activated by extracellular-signal-regulated kinase (ERK) signaling pathway; in turn, Etv5 had a positive feedback on the expression of fibroblast growth factor receptor 2 (FGFR2) which lies upstream of ERK. Etv5 knockout decreased the expression of FGFR2, whose binding with fibroblast growth factor 4 was essentially needed for TSCs differentiation. Collectively, the findings in this study suggest that Etv5 is required to safeguard the TSCs differentiation by regulating FGFR2 and provide new clues to understand the specification of trophectoderm in vivo.A new Rhizopogon species associated with Pinus was discovered at local wild mushroom markets and Pinus armandii forests from March to July in Southwest China where it is considered a delicacy. Based on morphological and molecular phylogenetic analyses, the collections were described as Rhizopogon songmaodan sp. nov. belonging to the subgenus Versicolores. The ease of R. songmaodan inoculation onto pine species, and the high market demand of its sporocarps, could make R. songmaodan a good candidate for cultivation in Southwest China.As an important class of flavonoids, anthocyanin has been used to enhance plant-defensive mechanisms against heavy metal stress. However, there are few available reports regarding surface spraying of anthocyanin for reduction of Cd poisoning in rice and its practical applications in paddy fields. After rice growing, measurements were taken of rice growth, pigments, the antioxidant system, thiol compounds, and distribution of Cd in rice tissues. The results showed that surface spraying anthocyanin could promote rice growth, and relative to the control, total chlorophyll significantly increased by 22.62% after surface spraying of 7.5 g L-1 anthocyanin. Simultaneously, Cd accumulation in rice grains was 0.17 ± 0.02 mg kg-1, which was significantly decreased by 46.88% relative to the control. In the pot experiment (40-day-old rice), treatment with 7.5 g L-1 anthocyanin resulted in decreases of ·O2-, H2O2, and malondialdehyde contents in rice leaves, while the activities of superoxide dismutase, peroxidase, catalase, and ascorbate peroxidase were increased by 59.10, 23.81, 41.75, and 9.39%, respectively. Meanwhile, contents of glutathione, ascorbic acid, non-protein thiols, and phytochelatins showed respective increases of 7.24, 14.49, 42.81, and 41.13% compared with the control value. Subcellular analysis revealed that surface spraying of anthocyanin increased organelle and soluble fractions of Cd in leaf cells. In conclusion, surface spraying of 7.5 g L-1 anthocyanin was mainly attributed to increased antioxidant activities and subcellular sequestration of Cd in organelles and soluble fractions in rice leaves to reduce Cd accumulation in rice grains in the field.The effects of diazinon (DZN), an organophosphate pesticide, on lipid profiles have been extensively reported. However, controversy on this issue persists. Here, we performed a systematic and meta-analysis study to investigate the association between DZN exposure and dyslipidemia in rodents and fish species.
