This study investigated the effect of superchilled storage (-4 °C) on protein degradation and structural changes of beef steaks from M. longissimus lumborum compared with traditional chilling (2 °C) and frozen storage (-18 °C). Traditional chilling induced significantly greater degradation of troponin T and desmin, and more rapid loss of calpain activity, compared to superchilled or frozen storage treatments. The proteolysis of key myofibrillar proteins resulted in a sharp decline of WBSF values during traditional chilled storage. For frozen beef samples, no major changes were observed with respect to protein degradation or muscle structure during storage. However, superchilled samples exhibited wider gaps between muscle fibers at 12 weeks storage, associated with muscle fiber shrinkage. To reconcile the inconsistency of the association between the resting-state functional connectivity (RSFC) and cognitive performance in healthy and depressed groups due to high variance of both measures, we proposed a Bayesian spatio-temporal model to precisely and accurately estimate the RSFC in depressed and nondepressed participants. This model was employed to estimate spatially-adjusted functional connectivity (saFC) in the extended default mode network (DMN) that was hypothesized to correlate with cognitive performance in both depressed and nondepressed. Multiple linear regression models were used to study the relationship between DMN saFC and cognitive performance scores measured in the following four cognitive domains while adjusting for age, sex, and education. In ROI pairs including the posterior cingulate (PCC) and anterior cingulate (ACC) cortex regions, the relationship between connectivity and cognition was found only with the Bayesian approach. Moreover, only the Bayesian approach was able to detect a significant diagnostic difference in the association in ROI pairs, including both PCC and ACC regions, due to smaller variance for the saFC estimator. The results confirm that a reliable and precise saFC estimator, based on the Bayesian model, can foster scientific discovery that may not be feasible with the conventional ROI-based FC estimator (denoted as 'AVG-FC').A single nucleotide polymorphism (SNP) rs12415800 of the silent mating type information regulation 2 homolog 1 gene (SIRT1) has shown a genome-wide significant association with major depression disorder (MDD) in a recent GWAS using a large sample. Subsequent studies of SIRT1's biological functions were supportive of a possible role in the pathophysiology of MDD. However, SIRT1-mediated physiopathology of MDD may be brain region specific. In the present study, we investigated the impact of SIRT1 rs12415800 genotypes on gray matter volumes (GMV) among different brain regions in both MDD patients and healthy controls. The rs12415800 was genotyped in 170 patients with first-episode medication-naïve MDD (cases) and 170 healthy controls. Magnetic resonance imaging was conducted and the voxel-based morphometry (VBM) approach was employed to analyze obtained images. When compared with the cases carrying GG genotype, the cases carrying GA or AA genotypes (A for risk allele) showed decreased GMV in right precuneus, left cuneus/precuneus, and right frontal superior. In contrast, the rs12415800-associated GMV abnormalities were not observed in controls. IAP inhibitor The SIRT1-rs12415800 polymorphism may be associated with the changes of GMV in MDD patients. Background Systemic lupus erythematosus (SLE) is a multisystem autoimmune disease that often leads to situations of harm to the mother-fetus binomial. Given the potential for complications and morbidities in these pregnant women, it is essential that a multidisciplinary team be involved in pregnancy planning, as well as monitoring the course of the pregnancy and the postpartum period. Owing to the imminent risks of disease worsening along with consequent disabilities, these women may experience psychological and psychosocial impacts conflicting with the psychological demands of pregnancy. Objective To understand the meanings attributed to pregnancy by women with SLE. Design A qualitative design with face-to-face interview following a semi-structured script of open-ended questions. Setting A specialized outpatient clinic where during prenatal care, women with stable disease undergo scheduled appointments. Participants The sample was intentionally composed of women visiting a specialized outpatient clinic from as well as choosing the best contraceptive methods and making optimal reproductive decisions. The development of strategies to deal with changes in the perinatal period may be helpful, as these women are willing to take care of themselves. Background Gestational diabetes mellitus is a rising global public health problem that can have short- and long-term sequelae for both mother and offspring. However, there are limited evidences on the effect of gestational diabetes mellitus on adverse neonatal outcomes using the updated international diagnostic criteria on adverse effects on neonatal outcomes. Therefore, this study was aimed to examine the effects of gestational diabetes mellitus on the risk of adverse neonatal outcomes in Ethiopia. Methods A prospective cohort study was conducted among pregnant women recruited from antenatal clinics and followed through pregnancy to delivery. Gestational diabetes mellitus was diagnosed using a two-hour 75g oral glucose tolerance test strategy with recent criteria. Multivariable log-binomial model was used to identify the effects of gestational diabetes mellitus on the risk of adverse neonatal outcomes which includes macrosomia, low birth weight, large for gestational age, small for gestational age, preterm bCI 1.95, 7.45), large for gestational age (ARR= 2.38; 95% CI 1.41, 4.03), preterm birth (ARR= 2.03; 95% CI 1.17, 3.53), low Apgar score at the first minute (ARR= 1.71; 95% CI 1.02, 2.86), and fifth minute (ARR= 2.14; 95% CI 1.05, 4.36). However, no significant differences in the risk of low birth weight and small for gestational age by gestational diabetes mellitus status. Conclusions Gestational diabetes mellitus increases the risk of adverse neonatal outcomes particularly macrosomia, large for gestational age, preterm birth, and poor Apgar score. Hence, the role of preventing gestational diabetes mellitus is quite crucial to improve neonatal outcomes.
