Tanycytes are radial glial cells located in the mediobasal hypothalamus. Recent studies have proposed that tanycytes play an important role in hypothalamic control of energy homeostasis, although this has not been directly tested. Here, we report the phenotype of mice in which tanycytes of the arcuate nucleus and median eminence were conditionally ablated in adult mice. Although the cerebrospinal fluid-hypothalamic barrier was rendered more permeable following tanycyte ablation, neither the blood-hypothalamic barrier nor leptin-induced pSTAT3 activation in hypothalamic parenchyma were affected. We observed a significant increase in visceral fat distribution accompanying insulin insensitivity in male mice, without significant effect on either body weight or food intake. A high-fat diet tended to accelerate overall body weight gain in tanycyte-ablated mice, but the development of visceral adiposity and insulin insensitivity was comparable to wildtype. Adavosertib Thermoneutral housing exacerbated fat accumulation and produced a shift away from fat oxidation in tanycyte-ablated mice. These results clarify the extent to which tanycytes regulate energy balance, and demonstrate a role for tanycytes in regulating fat metabolism. © 2020 Wiley Periodicals, Inc.The objective of this paper was to investigate the effects of hydrophobicity and molecular weight (MW) on the transepithelial transport permeability of oligopeptides across Caco-2 cell monolayers. Results showed that oligopeptides with different N-terminal amino acids had a wide range of permeability values and could be divided into three levels according to their correlations with log D and MW. At a good level of permeability, the permeability was positively correlated with log D, but negatively correlated with MW (p less then .001); at an intermediate level of permeability, the permeability was negatively correlated with log D and MW (p less then .001); and at a low level of permeability, the permeability was positively correlated with log D and MW (p less then .01). These results suggest for the first time that the transport of oligopeptides across Caco-2 cell monolayers might be closely related to their molecular properties of log D and MW. PRACTICAL APPLICATIONS A great number of food-derived bioactive peptides display health-promoting effects and show potential as bioactive ingredients in functional foods. However, the poor absorption in the intestine limits the application of food bioactive peptides, especially for the oligopeptides containing more than three amino acids. Although the transepithelial transport of food-derived oligopeptides in the intestinal epithelium has been widely reported, its transport mechanism is still obscure. Our study shows a three-level relationship between the transport permeability and log D and MW of oligopeptides across Caco-2 cell monolayers and provides a novel evidence for the coexistence of transcellular and paracellular pathways for the transport of oligopeptides through the intestine. This result will contribute to the understanding of the transport mechanisms of oligopeptides in the intestine. © 2020 Wiley Periodicals, Inc.AIMS To appraise the quality of current nursing clinical practice guidelines (N-CPGs) in China and explore the methodology for N-CPGs development. BACKGROUND Implementation of quality improvement projects based on N-CPGs has becoming an hot topic for nursing with proliferation in the number of N-CPGs in China in recent years. The methodology for developing N-CPGs is worthy of exploration. EVALUATION A systematic literature search was performed using PubMed, CINAHL, Web of Science, CNKI, Wanfang, VIP and CBM and relevant representative guidelines repositories from inception to July 31, 2019. Two authors independently selected eligible guidelines and performed data extraction. Four appraisers independently assessed the quality of the N-CPGs using the AGREE II tool. 20 N-CPGs were eventually included in this review. KEYISSUE (S) After AGREE II appraisal, the final domain scores ranged between 0.00% and 83.33%. When comparing the total domain scores, "Scope and purpose" and "Clarity of presentation" scored highest with a total of 63.89 (59.37-69.79) (%, median, interquartile range (IQR)), and 63.89 (58.33-75.70) (%), respectively. "Editorial independence" obtained the lowest ranking with a total score of 0 (0-81.25)(%). The total scores of "Stakeholder involvement", "Rigour of development", "applicability" and "editorial independence" were lower than 50%. CONCLUSION The quality of N-CPGs in China is not very high and the process of guideline development still needs to improve. IMPLICATIONS FOR NURSING MANAGEMENT N-CPGs are important documents used to guide nursing quality improvement. High quality N-CPGs are beneficial for nursing management. All rights reserved.BACKGROUND The impact of psoriasis on quality of life (QoL), sexuality and empathy requires better understanding in patient-partner relationships. OBJECTIVES To evaluate the influence of psoriasis on partner QoL, presence of sexual dysfunction (SDy) in couples and empathy in partners of psoriasis patients. METHODS A total of 183 adult psoriasis patients and their partners participated in this observational, cross-sectional and non-comparative study. Severity of psoriasis was measured using the Psoriasis Area and Severity Index. Patient QoL was assessed using the Dermatology Life Quality Index and the Short Form-12 (SF12). The impact of psoriasis on partner QoL was measured with the Family Pso and the SF12. Presence of SDy and empathy in partners were assessed using the Family Pso. RESULTS Overall, 49.7% of the patients had moderate-to-severe psoriasis. Patient psoriasis severity and patient QoL were correlated with partner psychological distress. The largest QoL impairment was observed in female patients withrmatology and Venereology.Mechanically guided, 3D assembly has attracted broad interests, owing to its compatibility with planar fabrication techniques and applicability to a diversity of geometries and length scales. Its further development requires the capability of on-demand reversible shape reconfigurations, desirable for many emerging applications (e.g., responsive metamaterials, soft robotics). Here, the design, fabrication, and modeling of soft electrothermal actuators based on laser-induced graphene (LIG) are reported and their applications in mechanically guided 3D assembly and human-soft actuators interaction are explored. Over 20 complex 3D architectures are fabricated, including reconfigurable structures that can reshape among three distinct geometries. Also, the structures capable of maintaining 3D shapes at room temperature without the need for any actuation are realized by fabricating LIG actuators at an elevated temperature. Finite element analysis can quantitatively capture key aspects that govern electrothermally controlled shape transformations, thereby providing a reliable tool for rapid design optimization.
