We performed extensive analysis of model parameters correlations to understand the crucial factors and enable extrapolation of a simplified model. We found good agreement between the simulation and the experimental data. The proposed model is a potent tool in the optimisation of electrospun structures and scaffolds for enhanced regenerative therapies.Nitric oxide (NO) binds to soluble guanylyl cyclase (sGC), activates it in a reduced oxidized heme iron state, and generates cyclic Guanosine Monophosphate (cGMP), which results in vasodilatation and inhibition of osteoclast activity. In inflammation, sGC is oxidized and becomes insensitive to NO. NO- and heme-independent activation of sGC requires protein expression of the α1- and β1-subunits. Inflammation of the periodontium induces the resorption of cementum by cementoclasts and the resorption of the alveolar bone by osteoclasts, which can lead to tooth loss. As the presence of sGC in cementoclasts is unknown, we investigated the α1- and β1-subunits of sGC in cementoclasts of healthy and inflamed human periodontium using double immunostaining for CD68 and cathepsin K and compared the findings with those of osteoclasts from the same sections. In comparison to cementoclasts in the healthy periodontium, cementoclasts under inflammatory conditions showed a decreased staining intensity for both α1- and β1-subunits of sGC, indicating reduced protein expression of these subunits. Therefore, pharmacological activation of sGC in inflamed periodontal tissues in an NO- and heme-independent manner could be considered as a new treatment strategy to inhibit cementum resorption.The aim of this research was to find out the morphometric differentiation of six native freshwater species in the Guayas Hydrographic Basin (Ecuador) by means of discriminant analysis. A total of 1355 mature fishes (Cichlasoma festae, Andinoacara rivulatus, Dormitator latifrons, Bryncon dentex, Hoplias microlepis and Leporinus ecuadorensis) were captured and 27 morphometric measurements and 20 landmarks were used. Two-way analysis of variance with species and sex as fixed factors and discriminant analysis were applied. The selection of the most discriminant variables was made applying the F of Snedecor, Wilks'-Lambda and the 1-Tolerance. While sex within species had no significant effect on the morphology, differences among species were significant. Twenty-seven morphological variables showed highly significant differences among six native freshwater species. Nine biometric variables with high discriminant power were selected. The six species analyzed were discriminated by the morphometric models generated, thus showing that discriminant analysis was useful for differentiating species. The morphometric differentiation by discriminant analysis is a direct, simple and economic methodology to be applied in situ in rural communities. It favors the implementation of a livestock development program and it could be used with other native freshwater species in the Guayas Hydrographic Basin.A prominent circadian variation is present in atrial fibrillation (AF) attacks that may be related to the expression of circadian clock genes. Little is known about the expression of circadian clock genes in AF. We prospectively enrolled 73 patients who had received pacemaker implantation, in order to define the burden of atrial high-rate episodes (AHREs) accurately. AF was diagnosed clinically in 43 (59%) patients (15 with persistent AF and 28 with paroxysmal AF). The expression levels of circadian clock genes of peripheral blood leukocytes were checked. There were more males and patients with a larger left atrial (LA) size and lower expression levels of BMAL1, CRY2, NR1D1, NR1D2, PER2, RORA, RORC, and TIM genes in persistent AF group than in other groups. There was a significant correlation between higher AHRE burden and larger LA size and between higher AHRE burden and decreased expression of circadian clock genes in patients with AF. LA volume and the expression of CRY1, NR1D1, and RORA are significantly associated with AHRE burden. However, the underlying mechanism needs to be elucidated in further studies.Reformulating recipes of ready-to-eat meat products such as salami to reduce salt content can mitigate the negative health impacts of a high salt diet. We evaluated the potential of potassium lactate (KL) as a sodium chloride (NaCl) replacer during salami production. NaCl and KL stress tolerance comparisons showed that four food-derived Listeria innocua isolates were suitable as biologically safe Listeria monocytogenes surrogates. Effects of the high salt (4% NaCl) concentration applied in standard salami recipes and a low salt (2.8% NaCl) plus KL (1.6%) combination on product characteristics and growth of contaminating Listeria and starter culture were compared. Simulated salami-ripening conditions applied in meat simulation broth and beef showed that the low salt plus KL combination retained similar to superior anti-Listeria activity compared to the high salt concentration treatment. Salami challenge tests showed that the low NaCl plus KL combination had comparable anti-Listeria activity as the high NaCl concentration during ripening and storage. No significant differences were detected in starter culture growth profiles and product characteristics between the high NaCl and low NaCl plus KL combination treated salami. In conclusion, KL replacement enabled a 30% NaCl reduction without compromising the product quality and antimicrobial benefits of high NaCl concentration inclusion.During early lactation in dairy cows, metabolic processes are adopted to provide energy and nutrients for the synthesis of milk compounds.
