The transfusion of naturally derived or modified cellular therapies, referred to as adoptive cell therapy (ACT), has demonstrated clinical efficacy in the treatment of hematologic malignancies and metastatic melanoma. Additionally, cellular vaccination, such as dendritic cell (DC) based cancer vaccines, continues to be actively explored. The manufacturing of these therapies presents a considerable challenge to expanding the use of ACT as a viable treatment modality, particularly at academic production facilities. Furthermore, the expanding commercial interests in ACT presents new opportunities as well as strategic challenges for the future vision of cellular manufacturing in academic centers. Current trends in the production of ACT at tertiary care centers and prospects for improved manufacturing practices that will foster further clinical benefit are reviewed herein.
Since before the COVID-19 pandemic, hospital-acquired infections (HAIs) represented a global healthcare crisis. Few studies suggested that COVID-19-related basic hygiene measures (BHM) could lower HAIs rates, reaching inconclusive results. The aim of this study was to investigate the hypothetical benefit on HAIs rate of COVID-19-enhanced BHM systematic introduction after major elective urological surgery.
Since the pandemic began, our hospital has implemented BHM to limit the spread of COVID-19. We compared patients operated in the pre-COVID-19 era (no-BHM period) with those operated after the pandemic started (BHM period). Outcomes were the incidence of HAIs and postoperative complications, and the length of hospital stay (LOS). Two balanced groups were generated by propensity score 11 matching.
Of 1053 major urological interventions, 604 were performed in the no-BHM period, and 449 in the BHM period. After matched analysis, the comparison groups consisted of 310 patients each. Of 107 recorded HAIs, 43 occurred during the BHM period (13.9%), and 64 during the no-BHM period (20.7%), with a statistically significant difference in multivariable analysis (OR 0.5 [95% CI 0.3-0.8], P=0.004). Postoperative complications rate was significantly lower in the BHM period than in the no-BHM period (29.0% versus 36.5%, OR 0.6 [95% CI 0.4-0.9], P=0.01). The LOS differed significantly between BHM and no-BHM periods a median of 5 (5-8) days versus 6 (5-8), respectively (P<0.001).
The risk of infections, postoperative complications, and prolonged LOS after major urological surgery was significantly reduced with the systematic introduction of COVID-19-related BHM, their application could, therefore, be prolonged with lasting benefits.

Data is lacking about long-term impact of JJ stents (JJst) on renal parenchyma. The aim of the study was to assess the evolution of renal parenchyma in patients with JJst indwelling for more than two years, and to find predictive factors for the development of renal atrophy.
Consecutive patients with JJst indwelled for more than 24 months, with a history of cancer, were retrospectively included. Replacements of JJst were scheduled every six months, or earlier in case of premature obstruction. Patient characteristics at the time of insertion of JJst, history of indwelling JJst and most recent data (serum creatinine, cancer status, definite JJst removal, renal volume (RV) with3D software) were recorded.
With a median follow-up of 4 years, 73 patients were included. The indication of JJst insertion was mostly external compression (65.8%). CT scans were available to assess RV evolution in 66 patients (90.4%). Median shrinkage of RV was higher when JJ stenting was unilateral versus bilateral -40% (-63; -15) versus -16% (-36; -3), P<0.001. The duration of indwelling JJst was the only statistically significative predictive factor of renal shrinkage in multivariate analysis (OR [CI 95%] 1.35 [1.10-1.66] P=0.004). Median relative change from baseline in eGFR was -22% (-45%; -5%.). No statistically significant predictive factors of eGFR evolution were found in univariate and multivariate analysis.

In living organisms, sensitivity to radiation increases in the presence of oxygen (O
) compared with that under anoxic or hypoxic conditions. Here, we investigated whether O
concentration affected the response of mitochondria to X-rays radiation, which is associated with tumor microenvironment formation via fibroblast activation in radiation-related tumors.

concentrations were controlled at <5% (internal environmental oxygen condition) or anoxic levels during culture of normal human diploid lung fibroblasts TIG-3 and MRC-5. Protein expression associated with the response of mitochondria to radiation was assessed using immunostaining or western blotting.
Induction of DNA damage (marker γ-H2A histone family member X) and mitochondrial signaling (AMP-activated protein kinase), suppression of mitochondrial metabolic activity, and generation of reactive oxygen species occurred with radiation in cells cultured under 5% and 20% O

In this study, we employed three types of signal amplification strategies using size enlargement and/or increase in mass of gold and titanium dioxide nanoparticles. Since the basic principle of the QCM sensor is to measure the change in resonance frequency according to the mass change caused by the molecular interactions on the sensor surface, we were able to quantitatively analyze AFP by sandwich immunoassay using gold or titanium dioxide nanoparticles conjugated with anti-AFP detection antibodies and the subsequent three signal amplification techniques in a similar manner. The signal amplification technologies provide the size expansion of gold nanoparticles by gold or silver staining reaction and mass enhancement by photocatalytic silver staining of titanium dioxide nanoparticles. The limit of detections (LODs) of the AFP immunoassay in human serum by the gold and silver staining-mediated signal amplifications for gold nanoparticles were 56 and 87 pg mL-1, respectively, but by the photocatalytic silver staining signal amplification for titanium dioxide nanoparticles was 118 pg mL-1. This means that the signal amplification method through size enhancement by gold staining of gold nanoparticles further improved the detection ability of the QCM immunosensor.Cardiovascular diseases (CVDs) represent the leading cause of mortality worldwide. As a complex matrix of micronutrients, phytochemicals and sugars, the effects of orange juice (OJ) on CVD risk factors in overweight and obese adults warrant investigation. Hence, this systematic review and meta-analysis of randomized controlled trials (RCTs) evaluated the effect of chronic OJ consumption on the lipid profile, blood pressure, glycemic indices and inflammatory biomarkers in overweight and obese adults. Four databases were searched from inception until May 2022 and eight studies were included in the final analysis. Pooled results demonstrated the beneficial effect of OJ on systolic blood pressure (MD -0.98 mmHg, 95% CI [-1.52, -0.44], p = 0.000, I2 = 0.0%) and HDL-C (SMD 0.29, 95% CI [0.03, 0.54], p = 0.027, I2 = 0.0%) compared to the control group. However, total cholesterol, TG, LDL-C, glucose, insulin, HOMA-IR and CRP did not change following chronic OJ consumption. This meta-analysis demonstrates the beneficial effects of chronic OJ consumption on blood pressure and HDL-C in overweight and obese adults.We present explainable machine learning approaches for the accurate prediction and understanding of solvation free energies, enthalpies, and entropies for different salts in various protic and aprotic solvents. As key input features, we use fundamental contributions from the conceptual density functional theory (DFT) of solutions. The most accurate models with the highest prediction accuracy for the experimental validation data set are decision tree-based approaches such as extreme gradient boosting and extra trees, which highlight the non-linear influence of feature values on target predictions. The detailed assessment of the importance of features in terms of Gini importance criteria as well as Shapley Additive Explanations (SHAP) and permutation and reduction approaches underlines the prominent role of anion and cation solvation effects in combination with fundamental electronic properties of the solvents. These results are reasonably consistent with previous assumptions and provide a solid rationale for more recent theoretical approaches.
Technological advances have enabled us to profile single-cell multi-omics data from the same cells, providing us with an unprecedented opportunity to understand the cellular phenotype and links to its genotype. The available protocols and multi-omics datasets [including parallel single-cell RNA sequencing (scRNA-seq) and single-cell ATAC sequencing (scATAC-seq) data profiled from the same cell] are growing increasingly. However, such data are highly sparse and tend to have high level of noise, making data analysis challenging. The methods that integrate the multi-omics data can potentially improve the capacity of revealing the cellular heterogeneity.
We propose an adaptively weighted multi-view learning (scAWMV) method for the integrative analysis of parallel scRNA-seq and scATAC-seq data profiled from the same cell. scAWMV considers both the difference in importance across different modalities in multi-omics data and the biological connection of the features in the scRNA-seq and scATAC-seq data. It generates biologically meaningful low-dimensional representations for the transcriptomic and epigenomic profiles via unsupervised learning. Application to four real datasets demonstrates that our framework scAWMV is an efficient method to dissect cellular heterogeneity for single-cell multi-omics data.
