The present study assesses the liver cells' stability during in vitro expansion, as a prerequisite for therapeutic use. Results The human liver cell cultures in this study were propagated efficiently in vitro for at least 12 passages. No significant changes in morphology, intracellular ultrastructures and characteristic markers expression were found during in vitro expansion of cells from all analyzed donors. However, expanded cells derived from male donors of >60 years old, lost the Y chromosome. Conclusion Liver-derived cell cultures adopt a proliferative, stable mesenchymal phenotype, through an epithelial to mesenchymal transition process. The molecular and phenotypic changes of the cells during propagation are uniform, despite the heterogeneity of the different donors. Loss of Y chromosome occurs after cells' propagation in elder male donors.An inherent challenge to clinical trials that aim to test the efficacy of experimental therapeutics for patients with amyotrophic lateral sclerosis (ALS) is the relative rarity of the disease. A promising solution to this problem is a multi-center approach that ideally includes sites distributed across a broad geographic area. In support of such an approach, the European E-RARE program and the United States National Institutes of Health (NIH) partnered to support the investigator-initiated ROCK-ALS trial (Eudra-CT-Nr. 2017-003676-31, NCT03792490) as a multi-national collaboration between centers in Europe and North America that is led by European investigators. During the set-up of this international trial, however, a number of unanticipated legal, administrative, and financial complexities emerged that required significant adaptation of the proposed trial scheme. Here, we report our experience navigating these obstacles and describe the potential solutions that we explored. Our experience may inform future efforts to implement multi-national investigator-initiated trials that involve both European and United States centers.The medicinal plant, Scutellaria orientalis, is used for the treatment of several diseases in North-western of Iran. The purpose of this study was to investigate the phytochemical content, cytotoxicity assay against SW-480 and HCT-116 cells and anti-haemolytic activities using HPLC-PDA, GC/MS, MTT and spectrophotometry methods, respectively. Phytochemical screening revealed the presence of different terpenoids and flavonoids such as baicalin, tricin and wogonin and also some of the other medicinally active compounds such as conhydrine and cannabidiol. MTT results revealed that HCT-116 cells were more sensitive to examined extracts compared with SW-480 cells. Methanol extract had the most anti-proliferative activity against HCT-116 and SW-480 cells at 48 h with IC50 values of 614.5 and 592.3 µg/mL, respectively. Also, all samples had no significant haemolytic effect on human erythrocytes. Our results revealed that S. orientalis root extract has a promising anticancer activity, indicating the presence of major anti-cancer agents on human colon cell lines.
