The coordinated regulation of glycerol channels in adipose tissue (AQP7) and the liver (AQP9) has been suggested as an important contributor to the pathophysiology of type-2-diabetes mellitus, as it would provide glycerol for hepatic synthesis of glucose and triglycerides. The regulation of AQP7 and AQP9 is influenced by sex. This study investigates the effect of a high-fat diet (HFD) on glycerol metabolism in mice and the influence of sex and GLP-1-receptor agonist treatment. Female and male C57BL/6JRj mice were fed either a control diet or a HFD for 12 or 24 weeks. Liraglutide was administered (1 mg/kg/day) to a subset of female mice. After 12 weeks of HFD, females had gained less weight than males. In adipose tissue, only females demonstrated an increased abundance of AQP7, whereas only males demonstrated a significant increase in glycerol kinase abundance and adipocyte size. 24 weeks of HFD resulted in a more comparable effect on weight gain and adipose tissue in females and males. HFD resulted in marked hepatic steatosis in males only and had no significant effect on the hepatic abundance of AQP9. Liraglutide treatment generally attenuated the effects of HFD on glycerol metabolism. In conclusion, no coordinated upregulation of glycerol channels in adipose tissue and liver was observed in response to HFD. The effect of HFD on glycerol metabolism is sex-specific in mice, and we propose that the increased AQP7 abundance in female adipose tissue could contribute to their less severe response to HFD.
Central lymph node metastasis (CLNM) occurs frequently in patients with papillary thyroid cancer (PTC), but performing prophylactic central lymph node dissection is still controversial. There are no reliable models for predicting CLNM. This study aimed to develop predictive models for CLNM by machine learning (ML) algorithms.
Patients with PTC who underwent initial thyroid resection at our hospital between January 2018 and December 2019 were enrolled. A total of 22 variables, including clinical characteristics and ultrasonography (US) features, were used for conventional univariate and multivariate analysis and to construct ML-based models. A 5-fold cross validation strategy was used for validation and a feature selection approach was applied to identify risk factors.
The areas under the receiver operating characteristic curve (AUC) of 7 models ranged from 0.680 to 0.731. All models performed significantly better than US (AUC=0.623) in predicting CLNM (P<0.05). In decision curve, most of the models also performed better than US. Lurbinectedin solubility dmso The gradient boosting decision tree model with 7 variables was identified as the best model because of its best performance in both ROC (AUC=0.731) and decision curves. Based on multivariate analysis and feature selection, young age, male sex, low serum thyroid peroxidase antibody and US features such as suspected lymph nodes, microcalcification and tumor size > 1.1cm were the most contributing predictors for CLNM.
It is feasible to develop predictive models of CLNM in PTC patients by incorporating clinical characteristics and US features. The ML algorithm may be a useful tool for the prediction of lymph node metastasis in thyroid cancer.

Further, patients with T1D have 3-4-fold increased risk of developing micro- and macrovascular complications. However, the contribution of immune-related factors contributing to these diabetes complications are poorly understood. Individuals with long-term T1D who do not progress to vascular complications offer a great potential to evaluate end-organ protection. The aim of the present study was to investigate the association of inflammatory protein levels with vascular complications (retinopathy, nephropathy, cardiovascular disease) in individuals with long-term T1D compared to individuals who rapidly progressed to complications. We studied a panel of inflammatory markers in plasma of patients with long-term T1D with (n = 81 and 26) and without (n = 313 and 25) vascular complications from two cross-sectional Scandinavian cohorts (PROLONG and DIALONG) using Luminex technology. Individuals with diabetic peripheral neuropathy (DPN) have functional deficits that increase their risk of falling. However, psychological aspects such as loss of confidence in undertaking activities could also contribute to this risk. We examined correlations between balance confidence and fall risk among individuals with DPN.
This was a cross-sectional study of 146 individuals with DPN. Elevated fall risk was determined by timed up-and-go test with standard cut-off time of 13.5 seconds, and balance confidence was measured by 16-item Activities Specific Balance Confidence scale. Functional parameters assessed included functional reach, body sway velocity during quiet standing and muscle strength at ankle and toe.
Twenty percent of the DPN patients were at increased risk of falls. Every unit increase in balance confidence was associated with 9% (95% confidence interval 0.88, 0.95; p<0.001) reduced odds of falling, after adjusting for socio-demographic, health and functional characteristics. No other functional parameters had significant associations with fall risk in adjusted analyses.
