Studies evaluating colorectal cancer (CRC) risk associated with chronic hepatitis C virus (HCV) infection are limited.

In this case-control study, we identify 67,670 CRC cases newly diagnosed from 2005 to 2011 and randomly selected 67,670 controls without HCV and CRC from the same database, frequency matched by age and sex of cases.

Results of logistic regression analysis revealed that the adjusted odds ratio (aOR) of CRC was 1.16 (95% confidence interval [CI] = 1.08-1.24, p < 0.001) in association with chronic HCV. The CRC risk was slightly greater for women than for men. The risk decreased with age, with the aOR decreased from 2.26 (95% CI = 1.32-3.87, p = 0.003) in patients under 45 years old to 1.31 (95% CI = 1.10-1.55, p = 0.03) in patients aged 50-59, and 1.10 (95% CI = 1.00-1.22, p = 0.061) in patients aged over 70.

Our findings suggested that patients with chronic HCV infection are at an elevated risk of developing CRC. Our data also imply that the CRC prevention programs are needed to target younger HCV patients.

REG4 is associated with a relatively unfavorable prognosis and clinicopathologic features in cancers, including advanced tumor and nodal stage, histological differentiation, and liver and peritoneal metastasis. Moreover, REG4-positive cancer cells show more frequent resistance to chemoradiotherapy, especially 5-FU-based chemotherapy. REG4 participates in many aspects of carcinogenesis, including cell proliferation, apoptosis, cell cycle, invasion, metastasis, and drug resistance. The underlying mechanisms are complex and involve a series of signaling mediators and multiple pathways. Thus, REG4 may be a potential diagnostic and prognostic biomarker as well as a candidate therapeutic target in cancer patients. In this review, we systematically summarize the advances about the clinical significance, biological functions, and mechanisms underlying REG4 in cancer to provide new directions for future cancer research.
To establish and validate a radiomics model to estimate the malignancy of mediastinal lymph nodes (LNs) based on contrast-enhanced CT imaging.

In total, 201 pathologically confirmed mediastinal LNs from 129 patients were enrolled and assigned to training and test sets. Radiomics features were extracted from the region of interest (ROI) delineated on venous-phase CT imaging of LN. Feature selection was performed with least absolute shrinkage and selection operator (LASSO) binary logistic regression. Multivariate logistic regression was performed with the backward stepwise elimination. A model was fitted to associate mediastinal LN malignancy with selected features. The performance of the model was assessed and compared to that of five other machine learning algorithms (support vector machine, naive Bayes, random forest, decision tree, K-nearest neighbor) using receiver operating characteristic (ROC) curves. Calibration curves and Hosmer-Lemeshow tests were used to assess the calibration degree. AMG 232 Decision cuected by different scanners or slice thicknesses. There was no significant difference (DeLong test, P > 0.05) between any two subgroups, which showed the generalization of the radiomics score across different factors.

The model we built could help assist the preoperative estimation of mediastinal LN malignancy based on contrast-enhanced CT imaging, with stability for different scanners and slice thicknesses.

This study aimed to investigate the prognostic value of a combination of preoperative plasma fibrinogen, prognostic nutritional index, and monocyte-to-lymphocyte ratio (Fib-PNI-MLR) in patients with non-metastatic RCC undergoing nephrectomy. We retrospectively collected medical data from 829 of the 1,019 cases of RCC. The optimal cutoff values of fibrinogen (≥3.54 vs. less then 3.54, mg/dl), PNI ( less then 47.03 vs. ≥47.03), and MLR (≥0.29 vs. less then 0.29) were defined using receiver operating characteristic (ROC) analysis and the Fib-PNI-MLR score (range, 0-3) was determined as the sum of points (0 or 1) assigned to each indicator. As a result, Fib-PNI-MLR was an independent risk factor for overall survival (OS), cancer-specific survival (CSS), and metastatic-free survival (MFS) (all P less then 0.05). The concordance-index and area under the curve (AUC) were larger for the Fib-PNI-MLR score than that for other clinical parameters. Subgroup analysis (Fuhrman grade G1+G2 and Fuhrman grade G3+G4; pathologic T1, T2, and T3-4 stage) revealed the significant association of a higher Fib-PNI-MLR score with poor urological outcomes (all P less then 0.05). Data indicated that patients with higher Fib-PNI-MLR might benefit from partial nephrectomy. The Fib-PNI-MLR score might serve as a promising prognostic factor in patients with non-metastatic RCC.
To validate the feasibility and efficiency of a fully automatic knowledge-based planning (KBP) method for nasopharyngeal carcinoma (NPC) cases, with special attention to the possible way that the success rate of auto-planning can be improved.

A knowledge-based dose volume histogram (DVH) prediction model was developed based on 99 formerly treated NPC patients, by means of which the optimization objectives and the corresponding priorities for intensity modulation radiation therapy (IMRT) planning were automatically generated for each head and neck organ at risk (OAR). The automatic KBP method was thus evaluated in 17 new NPC cases with comparison to manual plans (MP) and expert plans (EXP) in terms of target dose coverage, conformity index (CI), homogeneity index (HI), and normal tissue protection. To quantify the plan quality, a metric was applied for plan evaluation. The variation in the plan quality and time consumption among planners was also investigated.

With comparable target dose distributions, the KBP method achieved a significant dose reduction in critical organs such as the optic chiasm (p<0.
