It is accepted that a subset of all diabetes patients, generally estimated to account for 1-2% of all diabetic cases, is attributed to mutations in single genes. As only a subset of these genes has been identified and fully characterized, there is a dramatic need to understand the pathophysiological impact of genetic determinants on β-cell function and pancreatic development but also on cell replacement therapies. Pluripotent stem cells differentiated along the pancreatic lineage provide a valuable research platform to study such genes. This review summarizes current perspectives in applying this platform to study monogenic diabetes variants.
Superior outcomes after surgical treatment over medical treatment for primary aldosteronism (PA) has been reported in small-scale clinical studies, but no solid conclusion has been drawn as results of large randomized trials are lacking.

We performed a search of PubMed, MEDLINE, Embase and Cochrane Library for randomized or observational studies that investigated cardiovascular outcomes in patients with PA undergoing medical
surgical treatment. Meta-analyses of both composite and individual outcomes were conducted. Risks of bias of the included studies were assessed with Risk Of Bias In Non-randomized Studies of Interventions (ROBINS-I) checklist. Trial sequential analysis (TSA) was performed to control the risk of random errors and assess whether the results in our meta-analysis were conclusive.

A total of 12 studies, including a total of 6148 PA patients, were included in the meta-analysis. The results of meta-analyses demonstrated lower incidence of composite cardiovascular outcomes among PA patieralized PA.
To confirm the efficacy of ultrasound (US) guided radiofrequency ablation (RFA) in the treatment of benign thyroid nodules, we evaluated as primary outcome the technical efficacy and clinical success in a single center dataset. The secondary outcome was to find a correlation between nodules' pre-treatment features and volume reduction rate (VRR) ≥75% at 12 months after RFA and during follow-up period.

This retrospective study included 119 consecutive patients (99 females, 20 males, 51.5 ± 14.4 years) with benign thyroid nodules treated in our hospital between October 2014 and December 2018 with a mean follow-up of 26.8 months (range 3-48). Clinical and US features before and after RFA were evaluated by a US examination at 1, 3, 6, 12 months and annually thereafter up to 48 months.

The median pre-treatment volume was 22.4ml; after RFA we observed a statistically significant volume reduction from the first month (11.7ml) to the last follow-up (p < 0.001 for all follow-up times). The median VRR was 47.1crocystic nodules smaller than 22.4ml better long-term response can be achieved, which is predicted by an early shrinkage of the nodule.

Current perspectives on the neuroendocrine substrates of this behavioral regulation highlight a role of the peptide hormone oxytocin (OXT), but it remains unclear whether the facilitating effects of OXT vary as a function of facial familiarity. Here, a total of 73 healthy participants was enrolled in an eye-tracking experiment specifically designed to test whether intranasal OXT (24 IU) augments gaze duration toward the eye-region across four different face categories the participants' own face, the face of their romantic partner, the face of a familiar person (close friend) or an unfamiliar person (a stranger). We found that OXT treatment induced a tendency to spend more time looking into the eyes of familiar persons (partner and close friend) as compared to placebo. This effect was not evident in the self and unfamiliar conditions. Independent of treatment, volunteers scoring high on autistic-like traits (AQ-high) spent less time looking at the eyes of all faces except their partner. Collectively, our results show that the OXT system is involved in facilitating an attentional bias towards the eye region of familiar faces, which convey safety and support, especially in anxious contexts. In contrast, autistic-like traits were associated with reduced attention to the eye region of a face regardless of familiarity and OXT-treatment.To verify the viability and functionality of cryopreserved thyroid autotransplantation in rats who underwent total thyroidectomy in the treatment of postoperative hypothyroidism. Thirty-two Wistar rats were randomly assigned into groups (G) with eight animals each control (CG); simulation (SG); hypothyroidism (HTG) and transplanted (TG). At the beginning and in the 13th week of the experiment, serum levels of total T3, free T4, TSH and calcium were determined. In both the first and 14th weeks, scintigraphic examinations, 99m-Tc pertechnetate radioisotope biodistribution and histopathology were performed. In the 14th week, the expression of proliferating cell nuclear antigen (PCNA) and cellular apoptosis (caspase-3) were also evaluated. In the 13th week, the transplanted animals had normal serum levels of total T3 and free T4. TSH levels showed a tendency towards normality. In the 14th week, scintigraphic exams displayed graft isotopic uptake in all animals in the TG group. Histological examinations 13 weeks after transplantation showed the viability and functionality of thyroid follicles.
