The main source of mercury (Hg) exposure in the general population is fish. Another possible source is dental amalgam. Here, we compare the levels of Hg and selenium (Se) in samples of maternal and fetal origin collected shortly after childbirth of healthy postpartum women in the coastal (n = 96) and continental (n = 185) areas of Croatia related to maternal seafood/fish consumption. We also evaluated Hg concentrations and maternal serum metallothionein (MT2) concentrations in relation to the number of dental amalgam fillings, and MT2A-5A/G (rs28366003) polymorphism. The levels of Hg and Se in maternal hair and blood/serum, placenta and cord blood/serum increased in relation to increasing fish consumption with the highest values in subjects from the coast. The concentrations of each element and between elements correlated across the matrices. Increasing amalgam number correlated linearly with increased Hg levels in maternal and cord serum and was not associated with serum MT2. No association of MT2A-5A/G polymorphism and Hg or Se levels were found. The results confirmed higher fish consumption in coastal vs. continental Croatia and increases of both Hg and Se related to fish consumption in all analyzed samples. Increased blood Hg reflected the predominant MeHg share from seafood, while increased serum Hg matched exposure from dental amalgams.Estrogen-receptor-negative breast cancer (BCER-) is mainly treated with chemotherapeutics. Leptin signaling can influence BCER- progression, but its effects on patient survival and chemoresistance are not well understood. We hypothesize that leptin signaling decreases the survival of BCER- patients by, in part, inducing the expression of chemoresistance-related genes. The correlation of expression of leptin receptor (OBR), leptin-targeted genes (CDK8, NANOG, and RBP-Jk), and breast cancer (BC) patient survival was determined from The Cancer Genome Atlas (TCGA) mRNA data. Leptin-induced expression of proliferation and chemoresistance-related molecules was investigated in triple-negative BC (TNBC) cells that respond differently to chemotherapeutics. Leptin-induced gene expression in TNBC was analyzed by RNA-Seq. The specificity of leptin effects was assessed using OBR inhibitors (shRNA and peptides). The results show that OBR and leptin-targeted gene expression are associated with lower survival of BCER- patients. Importantly, the co-expression of these genes was also associated with chemotherapy failure. Leptin signaling increased the expression of tumorigenesis and chemoresistance-related genes (ABCB1, WNT4, ADHFE1, TBC1D3, LL22NC03, RDH5, and ITGB3) and impaired chemotherapeutic effects in TNBC cells. OBR inhibition re-sensitized TNBC to chemotherapeutics. In conclusion, the co-expression of OBR and leptin-targeted genes may be used as a predictor of survival and drug resistance of BCER- patients. Targeting OBR signaling could improve chemotherapeutic efficacy.Background We compared the efficiency of two MRI diffusion weighted imaging (DWI) techniques DWI with SPIR (DWISPIR) and DWI with STIR (DWISTIR), to estimate their eligibility for quantitative assessment of Crohn's disease activity in children and adults. Methods In inflamed terminal ileum segments (n = 32 in adults, n = 46 in children), Magnetic Resonance Index of Activity (MaRIA) was calculated, ADC values of both DWI techniques were measured, and the corresponding Clermont scores calculated. ADC values of both DWI techniques were compared between both and within each patient group, assessing their mutual correlation. Correlations between MaRIA and the corresponding ADC values, and Clermont scores based on both DWI techniques were estimated. Results No correlation between ADC of DWISPIR and DWISTIR was observed (rho = 0.27, p = 0.13 in adults, rho = 0.20, p = 0.17 in children). The correlation between MaRIA and Clermont scores was strong in both techniques-in SPIR, rho = 0.93; p less then 0.0005 in adults, rho = 0.98, p less then 0.0005 in children, and, in STIR, rho = 0.89; p less then 0.0005 in adults, rho = 0.95, p less then 0.0005 in children. The correlation between ADC and MaRIA was moderate negative for DWISTIR (rho = 0.93, p less then 0.0005 in adults, rho = 0.95, p less then 0.0005 in children), but, in DWISTIR, no correlation between ADC and MaRIA score was observed in adults (rho = -0.001, p = 0.99), whereas children presented low negative correlation (rho = -0.374, p = 0.01). check details Conclusions DWISTIR is not suitable for quantitative assessment of Crohn's disease activity both in children and adult patients.This investigation aimed to determine the countermovement jump (CMJ) neuromuscular performance (NMP) qualities that differentiate between athletes with high or low horizontal deceleration ability. Twenty-seven male university team sport athletes performed a CMJ on vertical axis force plates and a maximal horizontal deceleration following a 20 m maximal horizontal sprint acceleration. The instantaneous velocity throughout the maximal horizontal deceleration test was measured using a radar device. The deceleration ability was evaluated using the average deceleration (HDEC, m·s-2) and change in momentum-referred to as the horizontal braking impulse (HBI, N·s·kg-1). Participants were dichotomised into high and low HDEC and HBI according to a median-split analysis, and CMJ variables calculated for the overall eccentric, eccentric-deceleration and concentric phases. When horizontal deceleration ability was defined by HDEC, the CMJ concentric (effect size (ES) = 0.95) and eccentric (ES = 0.72) peak forces were the variables with the largest difference between groups. However, when defined using HBI, the largest difference was the concentric (ES = 1.15) and eccentric (ES = -1.00) peak velocities. Only the concentric mean power was significantly different between the high and low groups for both HDEC (ES = 0.85) and HBI (ES = 0.96). These findings show that specific eccentric and concentric NMP qualities may underpin the horizontal deceleration abilities characterised by HDEC and HBI. Specific NMP training interventions may be beneficial to target improvements in either of these measures of horizontal deceleration abilities.
