115 isolates of lactic acid bacteria were screened from traditional fermented foods in Guizhou province, China. The cholesterol-removed rates of 86 isolates was in range 7.29%~25.66%, of which 18 isolates was more than 15%. According to the results of physiological and biological test, 13 isolates were selected to determined the fermentation performance. 9 isolates including MT-4,MT-2,PJ-15,SR2-2,SQ-4,SQ-7,ST2-2,ST2-6,NR1-7 had high tolerance of bile salt and acid. Moreover, ST2-2, SR2-2, NR1-7, SQ-4, MT-4 had high survival rate in different concentration of NaCl and NaNO 2 under different temperature. According to the result of BLAST comparison of 16S rRNA sequence in GeneBank database and calculation the genetic distance of 16S rRNA sequence with ortho connected algorithm, SR2-2, NR1-7, ST2-2 were identified as Lactobacillus plantarum , MT-4 was identified as Lactobacillus pentosus , SQ-4 was identified as Lactobacillus paraplantarum. Moreover, strains SQ-4 and MT-4 was added to fermented beef, the result showed that fermented beef had delicious taste and was popular in consumers because its proper pH, pleasant colors, high viable cell count, and suitable content bound water and immobilized water. These results gave important information for the development of new starter formulation for the production of high quality fermented meat products.NK cells play critical roles in protection against haematological malignancies but can acquire a dysfunctional state, which limits anti-tumour immunity. However, the underlying reasons for this impaired NK cell function remain to be uncovered. We found that NK cells in aggressive B cell lymphoma underwent substantial transcriptional reprogramming associated with increased lipid metabolism, including elevated expression of the transcriptional regulator PPAR-g. Exposure to fatty acids in the lymphoma environment potently suppressed NK cell effector response and cellular metabolism. NK cells from both diffuse large B cell lymphoma patients and Eµ-myc B cell lymphoma-bearing mice displayed reduced IFN-g production. Bardoxolone Overall our data indicate that increased lipid metabolism, while impairing their function, is a functional adaptation of NK cells to the fatty-acid rich lymphoma environment.The study used PCR to determine the molecular basis of the antibiotic resistance and virulence profiles of isolates of Salmonella by targeting genes encoding for carriage and persistence in the poultry. A total of 1503 cecal samples from poultry were collected, 91 (6.1%) were positive for Salmonella spp. Ten different serotypes were detected from Salmonella isolates. The study was also conducted to determine the occurrence of 13 virulence and 12 resistance genes in isolates of Salmonella spp. All 46 isolates of Salmonella spp. tested were positive to one or more of the 12 virulence genes detected, ranging from 0.0% (viaB) to 100.0% (invA, mgtB, pipA and spi4D) (P less then 0.05). Occurrence of virulence genes varied significantly (P less then 0.05) by serotype but not by animal species. Of the 12 resistance genes assayed, only 4 (33.3%), strA, ampC, cmy2 and qnrB were detected. Overall, the occurrence of detection resistance genes was 71.7% (33/46), with 87.1% and 40.0% of the isolates from chickens and ducks, respectively positive (p=0.0009). The occurrence of resistance gene ranged from 2.2% (cmy2) to 50.0% (qnrB) in resistance gene positive isolates. The findings provide evidence that poultry from pluck shops are colonized by pathogenic Salmonella harboring virulence and antimicrobial resistance genes which may have clinical and therapeutic consequences, if the genes detected are expressed. Although there is a need for prudent use of antimicrobial agents in poultry production systems which should be constant monitoring for the prevalence of resistance in Salmonella isolates using phenotypic methods, the importance and knowledge of the occurrence of resistance genes in the pathogen in Trinidad cannot be ignored.The cellular and molecular processes involved in acute graft-versus-host disease (aGVHD) development early after allogeneic hematopoietic cell transplantation (HCT) in humans remain largely unknown. We have performed multiparameter immunophenotyping and molecular profiling of CD4+ and CD8+ T cells in 2 independent cohorts of patients undergoing HCT, as well as in their HLA-identical sibling donors. Cellular profiling using spectral flow cytometry showed an incomplete reconstitution of the T-cell compartment in recipients without aGVHD early after transplantation, as well as a shift toward an effector memory phenotype, paralleled by depletion of the naive T-cell pool. Molecular profiling of T-cell populations in donors vs recipients without aGVHD revealed increased pathway activity of >40 gene modules in recipients. These pathways were associated in particular with T-cell activation, adhesion, migration, and effector functions. Cellular profiles from recipients developing aGVHD displayed an enrichment of cells with a T memory stem cell-like phenotype compared with recipients without aGVHD. Comparison of gene profiles from these recipients revealed that transforming growth factor-β (TGF-β) signaling was most significantly downregulated, whereas the pathway activity of NF-κB-associated transcription factors and signaling pathways were increased, at aGVHD onset. This study suggests that the integration of cellular and molecular profiles provides new insights into the development of aGVHD in humans.
It remains unclear whether adrenalectomy has more beneficial effects than treatment with a mineralocorticoid receptor antagonist on vascular function in patients with aldosterone-producing adenoma (APA).
The aim of this study was to compare the effects of adrenalectomy and treatment with eplerenone on vascular function in patients with APA.
Flow-mediated vasodilation (FMD), as an index of endothelium-dependent vasodilation, and nitroglycerine-induced vasodilation (NID), as an index of endothelium-independent vasodilation, were measured to assess vascular function before and after a 3-month treatment with eplerenone and at 3 months after adrenalectomy in 23 patients with APA.
Flow-mediated vasodilation and NID after adrenalectomy were significantly higher than those before treatment with eplerenone (5.4 ± 2.6% vs 2.7 ± 1.9% and 14.8 ± 4.7% vs 9.6 ± 4.6%, P < 0.01, respectively) and those after treatment with eplerenone (5.4 ± 2.6% vs 3.1 ± 2.3% and 14.8 ± 4.7% vs 11.0 ± 5.3%, P < 0.01 and P = 0.03, respectively), while treatment with eplerenone did not alter FMD and NID compared with those before treatment with eplerenone.
