BACKGROUND AND AIMS Osteoarthritis (OA) is the most common type of arthritis disease. Previous etiological studies indicated that both environmental and genetic factors play important roles in the occurrence and development of knee OA (KOA). In the present study, we aimed to investigate the association between the FGF18 gene and susceptibility to KOA in Han Chinese population. METHODS A total of 2,556 unrelated Han Chinese individuals, including 866 KOA patients and 1,688 healthy controls, were recruited. Nine tag SNPs located within the gene region of FGF18 were selected for genotyping. Logistic models were fitted for single marker-based association analyses. Age, gender and BMI were included in each model as covariates. To investigate the functional consequences of significant SNP, we extracted expression quantitative trait loci (eQTL) data from the GTEx database. RESULTS SNP rs3884606 was significantly associated with the risk of KOA (OR [95% CI] = 1.25 [1.11-1.41], p = 0.0002) after adjusting for age, gender and BMI. The G allele was significantly associated with an increased risk of KOA. No significant eQTL signals could be identified for SNP rs3884606 on FGF18. CONCLUSION In this study, we identified a genetic polymorphism in FGF18 that was significantly associated with the risk of KOA based on samples with Chinese Han ancestry. Our findings replicated the recent GWAS report and supported an association between KOA and FGF18. BACKGROUND Maternal substance use can pose a risk to the fetal health. We studied the background characteristics of women with substance use disorders (SUDs) and selected neonatal outcomes in their children. MATERIAL AND METHODS A database-linkage study was performed. The sample consisted of pregnant women with a SUD during pregnancy (ICD-10 diagnosis F10-F19 except F17, n = 1710), women not diagnosed with a SUD (n = 1,511,310) in Czechia in 2000-2014, and their children. Binary logistic regression adjusted for age, marital status, education, concurrent substance use, and prenatal care was performed. RESULTS Women with illicit SUDs were younger, more often unmarried, with a lower level of education, a higher abortion rate, a higher smoking rate, and lower compliance to prenatal care than women with a SUD related to alcohol, or sedatives and hypnotics (SH). Women with a SUD had worse socioeconomic situations, poorer pregnancy care, and worse neonatal outcomes than women without a SUD. After adjustment, we found no difference in SGA between the illicit SUD groups and the alcohol and the SH groups. The newborns from all SUD groups had a higher risk of SGA when compared to women without a SUD. However after adjustment, the difference remained significant just in the alcohol group (OR = 1.9, 95 % CI = 1.4-2.6). CONCLUSION Mother's SUD during pregnancy increased risk of fetal growth restriction as measured by SGA. The role of maternal socioeconomic and lifestyle factors for the risk of SGA was substantial. V.INTRODUCTION Marijuana use is common among persons living with HIV, but whether it's use increases the risk of type 2 diabetes in this population has not been explored. OBJECTIVE To determine whether self-reported marijuana use is associated with incident type 2 diabetes in women and men living with and at risk for HIV. METHODS We analyzed data from the Women's Interagency HIV Study (WIHS) and Multicenter AIDS Cohort Study (MACS), between 2000-2017 (WIHS) and 1999-2017 MACS. The association between self-reported marijuana use and incident type 2 diabetes was analyzed using time-dependent Cox regression models among 3578 and 2682 participants in the WIHS and MACS respectively. RESULTS Over the follow-up period, 452 (WIHS) and 326 (MACS) incident type 2 diabetes cases occurred. In multivariable models, the hazard ratios, collectively indicate a reduced risk of type 2 diabetes, in marijuana users compared to none users, although all associations were not statistically significant. The results were similar for HIV-positive and HIV-negative participants in both cohorts. CONCLUSIONS In this prospective analysis of nearly 20 years of data for women and men with and at risk for HIV in the WIHS and MACS, although we found a pattern of reduced risk of type 2 diabetes among self-reported marijuana users, the associations were not statistically significant. To better inform clinical decisions and legal policy regarding marijuana use in this population, further longitudinal investigations that biologically quantify marijuana use to assess risk for incident diabetes is warranted. V.Human milk oligosaccharides (HMOs) are complex carbohydrates that exist naturally in mother's milk. Due to their myriad beneficial biological effects, HMOs are in the focus of current researches. Some of these complex carbohydrates are already commercially available utilizing their health-promoting benefits not just in infant formulas but in dietary supplements. Among the acidic oligosaccharides 3'- and 6'-sialyllactoses (SLs) are the most abundant in human milk. Extending the health benefits of these trisaccharides, 3'-SL and 6'-SL are expected to be active ingredients in infant- and follow-on formula and novel foods, therefore reliable and easy-to-perform analytical methods are urgently needed for their characterization. Herein we report a graphitized carbon liquid chromatography-mass spectrometry-based method for the separation and quantitation of sialyllactoses in human milk. Compounds were separated as their alditol forms and were quantified using maltotriose as internal standard. The developed bioanalytical method was validated in terms of selectivity, accuracy, precision, recovery, carry-over, matrix effects and stability. Two consecutive lactation periods of the same individual were monitored for the first time and a slight decrease in SL concentrations were found during the first month of the second lactation. A high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) method was successfully developed for the simultaneous detection of gabapentin (GBP) and cetirizine (CTZ) in cell lysates. Multiple reaction monitoring (MRM) transitions were found for GBP, CTZ and deuterated internal standards of both, allowing a selective and sensitive detection. Limits of detection (LODs) of 0.04 ng/mL and 0.07 ng/mL were achieved for CTZ and GBP respectively, and a linear range could be confirmed to the highest tested concentration of 50 ng/mL. Furthermore, with a total runtime of 2.5 min, this method allows a high throughput analysis. The applicability of the method was demonstrated by using it for the quantification of CTZ and GBP in cell lysates to examine the effects of renal transporters on these two analytes.
