Interventions aimed at counteracting burnout are the key to improving job satisfaction in nurses.Slow-wave sleep is one of the most important restorative components of sleep and central for our health and cognitive functioning. Although the amount of slow-wave sleep depends on sleep drive, age and other factors, also the pre-sleep mental state might influence sleep depth. We had shown that a pre-sleep hypnotic suggestion to sleep more deeply increased slow-wave sleep duration in hypnotizable subjects. In contrast, low-hypnotizable participants decreased sleep depth after this intervention. A possible reason might be an aversion to and active resistance against hypnosis. To overcome this potential opposition, we introduced the procedure as 'guided imagery'. We replaced the hypnotic induction by a breathing relaxation. Erdafitinib nmr Importantly, the suggestion 'to sleep more deeply' remained identical. We expected that these changes would make it easier for low-hypnotizable subjects to benefit from the suggestion. In contrast, young healthy low-hypnotizable participants did not show positive effects. Similar to our previous studies, they exhibited a reduced slow-wave sleep duration after the intervention. Additionally, the ratio between slow-wave activity and beta band power decreased. Subjective sleep quality remained unaffected. Our results indicate that suggestions to sleep more deeply result in decreased sleep depth in low-hypnotizable participants regardless of the mental technique (guided imagery versus hypnosis). Thus, the aversion against hypnosis per se cannot explain the detrimental effect of the intervention on slow-wave sleep in low-hypnotizable subjects. The results support the notion that our mental state before sleep can influence subsequent slow-wave sleep. However, the mechanisms of the contradictory decrease in low-hypnotizable subjects remain unknown.
TERT promoter mutation (TERTp
) has a strong association to recurrence and has been suggested to act as a driver mutation for malignant transformation of WHO grade I and II meningiomas. TERTp
has been investigated in selected high-grade meningioma samples. The existence of TERTp
across recurrent tumors in a population-based cohort needs to be investigated in order to identify when TERTp
emerges across recurrent samples and to validate prognostic impact among WHO grade III tumors.
We gathered material from a consecutive single-center cohort of 40 patients with malignant meningioma (WHO grade III) treated between 2000 and 2018, including specimens from primary and secondary malignant meningiomas with the corresponding earlier benign specimens and later malignant recurrences. In total 107 tumor samples were studied by Sanger sequencing for TERT promoter mutational status.
Seven of 40 patients (17.5%) harbored TERTp
thus validating the incidence of TERTp
in previous non-population-based cohorts. In 6/7 patients, the TERTp
was present at initial surgery (WHO grade I-III) while in one patient the TERTp
was found de novo when the meningioma became malignant. The incidences were 2/1.000.000/year for TERTp
WHO grade III meningioma and 8/1.000.000/year for TERTp

Noncatalytic extein residues near the splice junctions, such as the residues at the -1 and +2 positions, often strongly influence the protein-splicing efficiency. The substrate specificities of inteins have not been studied for many inteins. We developed a convenient mutagenesis platform termed "QuickDrop"-cassette mutagenesis for investigating the influences of 20 amino acid types at the -1 and +2 positions of different inteins. We elucidated 17 different profiles of the 20 amino acid dependencies across different inteins. The substrate specificities will accelerate our understanding of the structure-function relationship at the splicing junctions for broader applications of inteins in biotechnology and molecular biosciences.Bovine ephemeral fever (BEF), caused by the bovine ephemeral fever virus (BEFV), is associated with an acute febrile infection in cattle and widespread in tropical and subtropical areas, leading to great economic losses to cattle and milk industry. However, no efficacious BEF vaccine is currently available in China. Herein, we generated a recombinant rabies virus (RABV) expressing BEFV glycoprotein (LBNSE-BG), utilizing a reverse genetics system based on the recombinant rabies virus strain LBNSE. It was found that mice immunized with LBNSE-BG produced robust neutralizing antibodies against both BEFV and RABV, and developed complete protection from lethal RABV challenge. Further studies showed that LBNSE-BG activated more dendritic cells (DCs), B cells and T cells in immunized mice than the parent virus LBNSE. Collectively, these findings demonstrate that the recombinant LBNSE-BG described here has the potential to be developed as a cost-effective and efficacious bivalent vaccine for cattle use in endemic areas of BEF and rabies.The elongated, distally tapered limbs of horses are adapted for high-speed locomotion. Because these traits are artificially selected for in modern racehorses, they operate at a morphological extreme with a high risk of fracture. Racehorses are subject to different training and racing regimes depending on their breed and gait, and are therefore an interesting model to examine bone functional adaptation under variable biomechanically intense conditions. This study compares bone structural properties in the third metacarpal (MCIII) of Thoroughbred (n = 9) and Quarter Horse (n = 11) racehorses, using feral Assateague Island ponies (n = 6) as an untrained/unraced outgroup, to determine whether structural properties reflect variable racing and training regimes. Geometric section properties and bone mineral densities were determined using peripheral quantitative CT at two diaphyseal sites and through the distal epiphysis. Diaphyseal strength of the MCIII in all three breeds does not differ relative to body size, but in the mid-diaphyseal region Thoroughbreds have higher antero-posterior relative to medio-lateral bending strength than Quarter Horses, as well as higher bone mineral densities in left MCIII epiphyses (particularly in the lateral condyle).
