Food and nutritional security continue to be the issues of concern in developing countries like ours. Exploring the reservoir of high potential unexplored genetic resources could address the world's food and nutritional insecurity. The availability of diverse data and the population structure of any crop germplasm is a valuable genetic resource for discovering genes that can help achieve food and nutritional stability. We used seven ISSR and seven SSR markers to investigate diversity among 63 buckwheat genotypes, including landraces from India's northwestern Himalayas. Various parameters such as percent polymorphism, PIC, resolving power, and marker index was used to evaluate the inequitable efficacy of these markers. We foundthat both marker systems are effective in detecting polymorphism in buckwheat germplasm. Seven ISSRs produced 55 polymorphic bands, while seven SSRs produced 32bands. When compared to ISSRs, SSRs had a greater average PIC value (0.43) than that of (0.36). ISSRs, on the other hand, had a resolving power of (4.38) compared to (1.42) for SSRs. The hierarchical cluster analysis dendrogram divided genotypes into three major clusters. We found that both marker systems were equally accurate in grouping buckwheat genotypes according to their geographical origins. Using 7 ISSR and 7 SSR markers, the model-based STRUCTURE analysis established a population with two sub-populations that correspond to species-based groupings. Within the population, there was a high level of genetic diversity. These results have consequences for both buckwheat breeding and conservation efforts.Colorectal cancer (CRC) is one of the leading causes of death in Saudi Arabia. CRC mostly affects older age groups, but now a days it also appears frequently at a young age. However, the complete genetic etiology of CRC remains unknown. To identify the genetic factors responsible for this cancer type and to search for biomarkers for early diagnosis and prevention, we collected sixteen CRC tumor tissue samples and six normal colon tissues and extracted mRNA and synthesized cDNA. We then performed microarray transcriptomic profiling of Saudi patients with colon cancer. Gene expression was analyzed using Partek Genomics Suite, and principal component analysis (PCA) was performed to separate the different clusters of colon cancer and healthy tissues. Distinct differences in gene expression profiles were observed between colon cancer and normal tissue samples. Subsequently, we validated gene expression using real-time PCR. Aminocaproic chemical structure We found that the C-X-C motif chemokine ligand 8 (CXCL8) gene was expressed most in CRC samples. CXCL8 expressed 25.6 folds more in CRC tissues than in healthy tissues. In conclusion, we found that CXCL8 is the chief biomarker gene that is expressed most in CRC and plays an important role in tumor progression and metastasis.Azima tetracantha, a traditional medicinal plant included in the order Brassicales and family Salvadoraceae, is widely used as a dietary supplement in folklore medicines. The plant is also used for the treatment of rheumatism, diarrhea and other inflammatory disorders. The present investigation focused on the phytochemical composition, radical scavenging, reducing potential and anti-proliferative activities of the A. tetracantha leaves. Quantitative estimation of the polyphenols and flavonoids revealed significantly elevated levels in the methanol extract. Corroborating with this, methanol extract exhibited higher in vitro anti-radical scavenging effect against 2,2-diphenyl-1- picrylhydrazyl (34.14 ± 2.19 μg/mL), and hydrogen peroxide (44.96 ± 1.77 μg/mL), as well as ferric reducing properties (58.24 ± 6.98 μg/mL). The methanolic extract also showed strong lipoxygenase (71.42 ± 6.36 μg/mL) and nitric oxide inhibitory activities (94.23 ± 8.11 μg/mL). Cytotoxic activity against MCF7 cells was found to be higher (IC50= 37.62 ± 2.94 μg/mL), than that of MDAMB231 cells (IC50= 69.11 ± 5.02 μg/mL). The qPCR-based analysis indicated dose-dependent increase in the expression of the pro-apoptotic genes such as executioner caspases and apoptotic protease activating factor-1. Overall, the results indicated the possible use of methanol extract of A. tetracantha leaves as a chain-breaking antioxidant molecule and are capable of inhibiting inflammatory enzymes and the proliferative potential of breast cancer cells. The long distant, transcontinental migration of shorebirds entails many well identified costs in terms of time, energy, and direct mortality risk. Injuries from debris or from human structures and activities were observed as the major reasons for the direct mortality of shorebirds during migration worldwide. We recorded injured birds in major coastal wetlands of Kerala, for a period of 15 years from 2005 to 2019. The injured birds were observed in 9 different sites in various districts of Kerala. The highest instances of injuries were observed in Kadalundi-Vallikunnu Community Reserve, the major wintering and stop over site of migrant shorebirds in the west coast of India. During the study period, fifty-eight individuals of shorebirds belonging to four families were found to be injured. The highest proportion of injuries was recorded among the families Scolopacidae and Charadriidae comprising long distance migrant shorebird species and the lowest among Laridae and Ardeidae. We recommend that environmental authorities pay special attention to minimize anthropogenic debris along the flyways used by migratory birds thereby reducing the risk of injuries to some of these species. Proactive measures such as removal of discarded fishing gear or plastic debris from wintering areas as well as stopover areas could greatly reduce injuries in migratory birds arising from anthropogenic sources.Long non-coding RNAs (lncRNAs) play pivot roles in regulating mRNA expression in eukaryotic organisms without coding any proteins. In the current study, a comprehensive analysis of 260 published RNA-Seq datasets collected from different tissues (fruits, leaves, stems, and roots) of Coffea arabica L. was performed to discover potential lncRNAs. A total of 10,564 unique lncRNAs were identified. Our results showed that 77.14% of the lncRNAs were intergenic and 60.39% of them are located within 5 Kbp from the partner gene. In general, all the identified lncRNAs showed shorter lengths, fewer number of exons, and lower expression levels as compared to mRNAs in different studied tissues. Several lncRNAs were determined as differentially expressed (DE) in fruits as compared to leaves, stems, or roots. The functional characterization of the DE lncRNAs revealed their roles in regulating significant biological processes in different tissues of C. arabica. The current study provides a comprehensive analysis and dataset of lncRNAs in C. arabica that could be utilized in further studies concerning the roles of these molecules in plant cells.
We present six datasets of saproxylic beetles collected between 2012 and 2018 in Central and Southern Italian forests. Saproxylics represent one of the main components in forest ecosystems in terms of diversity, species richness and functional traits and, for this reason, they are an important target group for studying the modification of forests over time. The datasets consist of annotated checklists and were published on Zenodo repository.
Overall, 1,171 records are published, corresponding to 918 taxa (taxonomy at species or subspecies level). The taxa are scarcely shared amongst the areas, 80.2% of them are exclusive, indicating that the beetle communities are substantially different. In consideration of the biodiversity crisis we are passing through, which is especially dramatic for the insects, we want to promotecollaboration amongst researchers for making datasets available in open repositories. This will improve the possibility for researchers and forest managers of analysing the state of species me and space.
The genus
Heller, 1912 is the most speciose and taxonomically complex genus in the tribe Pachyrhynchini. It is known to be endemic in the Philippines, with most species having a very narrow range of distribution. There are already more than 230 species of
documented in the Philippines.
sp. nov., a new species of the genus
Heller, 1912 from Davao de Oro, Mindanao Island, Philippines, is described with brief notes about its ecology. Its specific epithet is from the Cebuano word "dagtum" which means pitch black referring to the colour of the integument of the species.
Metapocyrtusdagtum sp. nov., a new species of the genus Metapocyrtus Heller, 1912 from Davao de Oro, Mindanao Island, Philippines, is described with brief notes about its ecology. Its specific epithet is from the Cebuano word "dagtum" which means pitch black referring to the colour of the integument of the species.Vitisshizishanensis (Vitaceae), a new species from Hubei, China, is described and illustrated. It is morphologically similar to V.flexuosa and V.bryoniifolia, but differs in leaf lobing and pubescence. It can be easily distinguished from the two species based on its glabrous or with very sparse arachnoid tomentum on the abaxial mature leaf surface, and its unlobed to 3-7 lobed leaves. A detailed description, along with photographs for the new species, and a table for morphological comparisons with similar Vitis species, are also provided.
Ginsenoside Rh2, a major saponin derivative in ginseng extract, is recognized for its anticancer activities. Compared to coding genes, studies on long noncoding RNAs (lncRNAs) and microRNAs (miRNAs) that are regulated by Rh2 in cancer cells, especially on competitive endogenous RNA (ceRNA) are sparse.
LncRNAs whose promoter DNA methylation level was significantly altered by Rh2 were screened from methylation array data. The effect of STXBP5-AS1, miR-4425, and RNF217 on the proliferation and apoptosis of MCF-7 breast cancer cells was monitored in the presence of Rh2 after deregulating the corresponding gene. The ceRNA relationship between STXBP5-AS1 and miR-4425 was examined by measuring the luciferase activity of a recombinant luciferase/STXBP5-AS1 plasmid construct in the presence of mimic miR-4425.
Inhibition of STXBP5-AS1 decreased apoptosis but stimulated growth of the MCF-7cells, suggesting tumor-suppressive activity of the lncRNA. MiR-4425 was identified to have a binding site on STXBP5-AS1 and proven to be downregulated by STXBP5-AS1 as well as by Rh2. In contrast to STXBP5-AS1, miR-4425 showed pro-proliferation activity by inducing a decrease in apoptosis but increased growth of the MCF-7cells. MiR-4425 decreased luciferase activity from the luciferase/STXBP5-AS1 construct by 26%. Screening the target genes of miR-4425 and Rh2 revealed that Rh2, STXBP5-AS1, and miR-4425 consistently regulated tumor suppressor RNF217 at both the RNA and protein level.
LncRNA STXBP5-AS1 is upregulated by Rh2
promoter hypomethylation and acts as a ceRNA, sponging the oncogenic miR-4425. Therefore, Rh2 controls the STXBP5-AS1/miR-4425/RNF217 axis to suppress breast cancer cell growth.
