The estimation of bloodstain formation time is still an unsolved problem in forensic science and lacks accurate quantitative methods. Whether DNA can be adopted to estimate bloodstain formation time is still controversial, and there is no study to confirm the potential of mtDNA markers. To address these issues, a triple quantification method based on the ratio of mtDNA fragments of different lengths of COⅠ (mitochondrially encoded cytochrome c oxidase Ⅰ) for estimating bloodstain formation time was established. A total of 152 samples (140 old samples, 12 fresh samples) were collected and tested, and the absolute copies of different-sized fragments of COⅠ (304 bp, 120 bp, 41 bp) in all samples were quantified by SYBR Green real-time qPCR. The natural logarithms of two copy number ratios (304 bp/41 bp, 120 bp/41 bp) of COⅠ in old samples were calculated, which were used as degradation indexes to evaluate the degradation degree of mtDNA. The 140 old human blood samples from 1 to 14 years of storage were accumulated from casework of forensic practice to establish the method of estimating bloodstain formation time and used to analyze the impact of gender factors on the two degradation indexes, and 10 animal samples and 2 fresh human samples were collected to verify the human specificity of the method. There was a high correlation between degradation indexes and bloodstain formation time (the absolute values of correlation coefficients of these two degradation indexes were 0.901 and 0.758 respectively). A method with triple quantification and dual indexes estimating bloodstain formation timewas successfully established, which was highly human-specific. There was no statistically significant difference in degradation indexes between different gender samples (P > 0.05). This study confirmed that mtDNA can be utilized to estimate bloodstain formation time, which provides a new solution to the forensic problem of estimating the time of bloodstain formation.This case describes a 74-year-old male who presented with rapid atrial flutter in association with large atrial lipoma along the interatrial septum. Strawberry plants (Fragaria x ananassa) were grown in Hoagland's nutrient solution in a greenhouse with two levels of Fe 0 and 10 μM Fe(III)-EDDHA. After 20 days, plants growing without Fe showed typical symptoms of Fe deficiency chlorosis in young leaves. Then, the adaxial and abaxial sides of one mature or one young leaf in each plant were brushed with 10 mM malic (MA), citric (CA) or succinic (SA) acids. Eight applications were done over a two-week period. At the end of the experiment, the newly emerged (therefore untreated), young and mature leaves were sampled for nutritional and metabolomic analysis, to assess the effectiveness of treatments. Leaf regreening was monitored using a SPAD-502 apparatus, and the activity of the ferric chelate-reductase activity (FCR) was measured using root tips. Iron deficiency negatively affected biomass and leaf chlorophyll but did not increase FCR activity. Application of succinic acid alleviated the decrease in chlorophyll observed in other treatments, and the overall nutritional balance in the plant was also changed. The concentrations of two quinic acid derivatives increased under Fe deficiency and decreased in plants treated with succinic acid, and thus they are proposed as Fe stress markers. Data suggest that foliage treatments with carboxylates may be, in some cases, environmentally friendly alternatives to Fe(III)-chelates. The importance of Fe mobilization pathways in the formulation of new fertilizers is also discussed.The contamination of agricultural soils with Arsenic (As) is a significant environmental stress that restricts plant growth, metabolism, and productivity worldwide. The present study examined the role of elemental sulfur (S0) in protecting Brassica napus plants from Arsenic (As) toxicity. Arsenic (100, and 200 mg As kg-1 soil) in soil caused detrimental effects on five Brassica napus cultivars (Neelam, Teri-Uttam Jawahar, Him Sarson, GSC-101, and NUDB 26-11). The As toxicity inhibited the growth and photosynthesis indices in all cultivars with more deterioration effects in NUDB 26-11. Plant absorption and uptake of As caused the generation of oxidative injury by accumulating the reactive oxygen species (ROS), which simultaneously decreased the plant defence capability and ultimately the photosynthesis. Application of sulfur (S0, 100 or 200 mg S kg-1 soil) alleviated the negative impacts and toxicity of As on the photosynthesis and growth matrices of plants, especially under high S level. S0 also boosted the antioxidant potential of plants and toned-down lipid peroxidation and ROS aggravation such as superoxide anion (O2•-) and H2O2, hydrogen peroxide, in As affected plants. In general, S0 at 200 mg kg-1 soil more perceptibly increased the functionality of antioxidant enzymes, and non-enzymatic antioxidants, metal chelators and non-protein thiols. Further amendment of soil with S0 at fifteen days before seed sowing affected by As-induced toxic effects (added to soil at the time of sowing) considerably intensified the endogenous hydrogen sulfide (H2S) content and its regenerating enzymes D-cysteine desulfhydrase (DCD) and L-cysteine desulfhydrase (LCD) that further strengthened the defense capability of plants to withstand As-stress. Our results suggest the role of H2S in the S-induced defense operation of the B. napus plants in restraining As toxicity. The current study shows that S0 as a source of S might be used to promote the growth of B. napus plants in polluted agricultural soils.
Common titration strategies for vagus nerve stimulation (VNS) prioritize monitoring of tolerability during small increases in stimulation intensity over several months. This study aimed to characterize the relationship between titration speed and the onset of clinical response to VNS.

To assess differences between more aggressive titration strategies and more conservative ones, we analyzed the relationship between time-to-dose and time-to-response using a weighted Cox regression. The target dose was empirically defined as 1.625mA output current delivered at 250 microsecond pulse widths at 20Hz. Patient-level outcomes and dosing data were segregated into fast (<3 months), medium (3-6months), and slow (>6 months) cohorts based on their titration speed.

The statistical model revealed a significant relationship between titration speed and onset of clinical response, defined as a 50% reduction from baseline in seizure frequency. Frequency of adverse events reported between each cohort trended toward higher rates of adverse events in adults who were titrated quickly; however, the pediatric population appeared to be more tolerant of titration at any speed.

Homebound individuals with advanced Parkinson's disease (PD) are underrepresented in research and care. We tested the impact of interdisciplinary, telehealth-enhanced home visits (IN-HOME-PD) on patient quality of life (QoL) compared with usual care.

Nonrandomized controlled trial of quarterly, structured, telehealth-enhanced interdisciplinary home visits focused on symptom management, home safety, medication reconciliation, and psychosocial needs (ClinicalTrials.gov NCT03189459). We enrolled homebound participants with advanced PD (Hoehn & Yahr (HY) stage ≥3). Usual care participants had ≥2 visits in the Parkinson's Outcomes Project (POP) registry. We compared within- and between-group one-year change in QoL using the Parkinson's Disease Questionnaire.

Sixty-five individuals enrolled in IN-HOME-PD (32.3% women; mean age 78.9 (SD 7.6) years; 74.6% white; 78.5% HY≥4) compared with 319 POP controls, with differences in age, race, and PD severity (37.9% women; mean age 70.1 (7.8) years; 96.2% white; 15e and outcomes in advanced PD.
Prodromal constipation (PC) at Parkinson's disease (PD) onset may mark a distinct neurodegenerative trajectory; accordingly, presenting phenotype, biochemical signature, and progression of PD patients with PC (PD+PC) might differ from those without (PDwoPC). We compared the clinical-biochemical profile of de novo PD patients with and without PC, and the respective mid-term progression, to establish the grouping effect of PC.

Motor and non-motor scores were collected at diagnosis in n=57 PD+PC patients and n=73 PDwoPC. Paired CSF biomarkers (α-synuclein, amyloid and tau peptides, lactate, CSF/serum albumin ratio or AR) were assessed into a smaller sample and n=46 controls. Clinical progression was estimated as Hoehn and Yahr stage (HY) and levodopa equivalent daily dose (LEDD) change 2.06±1.35 years after diagnosis.
