The commercial formulation including gels, injections and ointments showed short duration of action (1 to 2 h). In this paper, the efforts have being made to develop tailored lidocaine-microemulsion (o/w), which on penetration in the skin layer cause micro-depots formation due to destabilization of the microemulsion system. To identify the microemulsion region, pseudo ternary diagrams were constructed using Capmul MCM as oil, Pluronic F68 as tri-block surfactant, polyethylene glycol 200 as co-surfactant at 14 and 16 ratios (SCo-S). The selected 5%w/v lidocaine loaded microemulsion [Ld-ME-2(14)] was stable in thermodynamic test and during shelf life period (3 months). In ex vivo permeability study, the lidocaine release from Ld-ME-2(14) microemulsion was sustained in comparison to the marketed lidocaine ointment. The skin irritation study confirmed the safety of lidocaine loaded microemulsion. Tail flick test showed improved and sustain local anaesthetic effect in comparison to the market ointment. The improved efficacy of microemulsion system, was due to high penetration in the skin layer due to local precipitation of lidocaine from microemulsion. The findings suggest that the tailored microemulsion could be a potential strategy to prolong the local anaesthesia.This study aimed to test the effects of a 25-session version of the Growing Pro-Social (GPS-25) program over schemas and schema-related emotions in male young offenders. Participants included 123 youth aged between 14 and 19 years, placed in eight Portuguese detention facilities. Youth were allocated to receive GPS (n = 63) or treatment as usual (n = 60), and answered a self-report measure assessing schemas and schema-related emotions at baseline and posttreatment. Two-factor mixed multivariate analysis of variance (MANOVA; group change) and the Reliable Change Index (individual change) revealed nonsignificant differences between groups for the schema's endorsement. Significant differences between groups were found for the schema-related emotions Treatment participants presented lower scores and/or higher clinical improvements after GPS, when compared with controls. GPS-25 produced change at an emotional level but not in schema's endorsement, suggesting that longer interventions should be tested in their capability to promote cognitive and emotional change in young offenders.Introduction Arenavirus are unique category-A pathogens that are also classified as Orphan diseases. The current review provides detailed patent landscape and a description of selected technologies developed for combating category-A Arenavirus. Currently, Arenavirus infections are epidemic [2] but could cause widespread pandemics due to ease of dissemination and lack of immunity against these viruses.Areas covered The key strings for selected Arenavirus VHF were run separately in MCPaIRS®, PatSeer, and Questel database. The search was limited to Title, Abstract and Claim fields; one member per patent family was considered for analysis.Expert opinion Synthetic molecules dominate the patent landscape, while natural products have not been extensively claimed for the treatment of Arenavirus infection. The broad-spectrum activity has been highly desired for Arenavirus treatment, but few reports have experimentally tested it. With each year, a constant increase in number of patents published is seen, while the maximum number of applications was filed in 2017. The research in VHF is driven by public funds; the maximum numbers of patents were filed by publicly funded organizations.Introduction Autoimmune hemolytic anemia (AIHA) is due to autoantibodies against erythrocytes that may arise either because of primary tolerance breakage or along with several associated conditions, including genetic predispositions, congenital syndromes, environmental triggers, autoimmune diseases, immunodeficiencies, and neoplasms.Areas covered This review evaluated the risk of AIHA development in associated conditions and summarized disease-intrinsic risk factors for relapse and outcome. Diagnostic procedures were analyzed to properly identify primary and secondary forms. A Medline including clinical trials, meta-analyses, guidelines, consensus, and case reports, published in the last 30 years was performed.Expert opinion The several associated conditions listed above constitute a risk for AIHA development and should be considered since disease course and therapy may be different. Particularly, AIHA developing after transplant or novel checkpoint inhibitors is an emerging complex entity whose proper therapy is still an unmet need.
