The objective of this study was to characterize a soft red winter wheat (SRWW) breeding population that has been subjected to intense germplasm introduction and alien introgression for FHB resistance in the past. The population was evaluated under misted FHB nurseries inoculated with Fusarium graminearum infested corn spawn for two years. Phenotypic data included disease incidence (INC), disease severity (SEV), Fusarium damaged kernels (FDK), FHB index (FHBdx), and deoxynivalenol concentration (DON). Genome-wide association studies by using 13,784 SNP markers identified twenty-five genomic regions at -logP ≥ 4.0 that were associated with five FHB-related traits. Of these 25, the marker trait associations that explained more than 5% phenotypic variation were localized on chromosomes 1A, 2B, 3B, 5A, 7A, 7B, and 7D, and from diverse sources including adapted SRWW lines such as Truman and Bess, and unadapted common wheat lines such as Ning7840 and Fundulea 201R. Furthermore, individuals with favorable alleles at the four loci Fhb1, Qfhb.nc-2B.1 (Q2B.1), Q7D.1, and Q7D.2 showed better FDK and DON scores (but not INC, SEV, and FHBdx) compared to other allelic combinations. Our data also showed while pyramiding multiple loci provides protection against FHB disease, it has significant trade-off with grain yield.Hydrangea macrophylla (Thunb.) Ser. (Hydrangeaceae) is the most popular hydrangea species grown in home gardens and landscapes in China. Plants of H. macrophylla with symptoms of powdery mildew were found in a commercial wholesale nursery in Huzhu, Haidong (36°49'11.87" N, 101°57'03.36″E, alt. 2490 m), in May 2020, with disease incidence reaching 80%. Symptoms included yellowing and necrosis of leaves. Upon microscopic observation, masses of conidia and mycelium were observed covering the symptomatic tissues. Fungal isolates displayed nipple-shaped hyphal appressoria, often poorly developed, conidiophores erect, arising laterally or from the upper surface of hyphal mother cells, and positioned almost centrally or towards one end of the cells, up to about 160 μm long (n = 30), with foot cells straight or flexuous, 32 to 86 × 8 to 13 μm (n = 50), followed by one to three shorter cells about 11 to 24 × 10 to 15 μm (n = 50), forming catenescent conidia in usually predominantly chains, conidia doliiform to limonifnd Cook 2012).Maize (Zea mays L.) is one of the major crops in China. In July 2020, leaf blight was observed on approximately 18% of maize plants at the 852 Farm, Heilongjiang province. Symptoms appeared as yellow necrotic lesions on leaf tips and margins, which later expanded to the entire. The disease was first observed on the lower leaves of the plants and then progressed up the plant. Thirty symptomatic leaves were collected in 2020, and tissue samples between healthy and necrotic area (4 × 2 mm) were surface disinfected with 1% NaOCl for 3 min, 70% ethanol for 10 s, and washed three times with sterile water. Disinfected tissues were placed on potato dextrose agar (PDA) amended with streptomycin sulfate (50 mg/liter) and incubated at 25°C for 1 week. Cultures were purified using the hyphal-tip technique for morphological and molecular analyses. Morphological characteristics were observed on 1-week-old PDA cultures grown at 25°C. Mycelium changed from cream colored to pale pink or reddish, and the back of plate turned pnts, and species identification was confirmed by the morphological and molecular method described. L. roseipellis has been reported to infect Cynodon dactylon, Lolium perenne and Festuca arundinacea, respectively. To our knowledge, this is the first report of the identification of L. 5'-N-Ethylcarboxamidoadenosine clinical trial roseipellis as a pathogen of maize in China, and this report will assist with monitoring distribution of the disease to assist with developing management recommendations.Viruses and viroids prevalent in a population of 42 wild grapevines (i.e., free-living, uncultivated grapevines; Vitis spp.) were compared to those in a population of 85 cultivated grapevines collected in Tennessee, USA by RNA-seq analysis of pools of ribosomal RNA-depleted total RNA. The sequences of 10 viruses (grapevine fleck virus, grapevine leafroll-associated virus 2, grapevine rupestris stem pitting-associated virus, grapevine Syrah virus 1, grapevine vein-clearing virus, grapevine virus B, grapevine virus E, tobacco ringspot virus, tomato ringspot virus and a novel nano-like virus) and two viroids (hop stunt viroid and grapevine yellow speckle viroid 1) were detected in both grapevine populations. Sequences of four viruses (grapevine associated tymo-like virus, grapevine leafroll-associated virus 3, grapevine red blotch virus and grapevine virus H) were identified only from cultivated grapevines. High, moderate and low numbers of sequence reads were identified only from wild grapevines for a novel caulimovirus, an enamovirus, and alfalfa mosaic virus, respectively. The presence of most virus sequences and both viroids was verified independently in the original samples by reverse transcription-polymerase chain reaction followed by Sanger sequencing. Comparison of viral sequences shared by both populations showed that cultivated and wild grapevines harbored distinct sequence variants, which suggests that there was limited virus movement between the two populations. Collectively, this study represents the first unbiased survey of viruses and viroids in both cultivated and wild grapevines within a defined geographic region.
