Plasmid and chromosomal-based complementation of MET25 in the met25 deficient cells on a double layer agar plate with nutrient gradients demonstrates delayed phenotype (white morphology) restoration, indicating post-transcriptional feedback regulation of methioninsm of the nonconventional yeast.After decades of being neglected, broad tapeworms now attract growing attention thanks to the increasing number of reports from humans but also thanks to many advancements achieved by application of molecular methods in diagnosis and epidemiological studies. Regarding sparganosis, unfortunately general uniformity of most species, their high intraspecific variability and lack of agreement among researchers has led to confusion about the classification of Spirometra/Sparganum species. For the first time we determined adult, eggs and plerocercoid life cycle stages and the molecular phylogeny of Sparganum proliferum obtained from endangered wild felids (Panthera onca, Leopardus pardalis, Leopardus guttulus and Herpailurus yagoauroundi) in one of the largest continuous remnants of worldwide biodiversity, the Atlantic Forest from South America. Our results showed that at least 57% of total species of wild felids in this natural area could act as definitive hosts of Sparganum proliferum. We conclude that the availability of more morphological characteristics are needed in order to secure reliable characterization and diagnosis of sparganosis. The integration of these data with molecular analysis of mitochondrial DNA sequences will be useful for species discrimination.Red deer (Cervus elaphus) carcasses showing grey-greenish discolouration have been increasingly observed in the canton of Grisons, Switzerland. We investigated whether Sarcocystis infections were associated with this pathology, and whether wild and domestic canids were involved in their transmission. Meat from affected red deer (n = 26), faeces and intestines from red foxes (Vulpesvulpes) (n = 126), and faeces from hunting dogs (n = 12) from the region, were analysed. Eosinophilic myositis and/or fasciitis were diagnosed in 69% of the deer, and sarcocysts were observed in 89% of the animals. Molecular typing targeting a ~700bp variable region of the 18S rRNA gene revealed Sarcocystis hjorti in 73%, S. venatoria/S. iberica in 54%, S. selleck chemicals llc linearis/S. taeniata in 12%, S. pilosa in 8% and S. ovalis in 4% of the deer samples. No inflammatory changes were observed in red deer carcasses with normal appearance (n = 8); however, sarcocysts were observed in one sample, and S. hjorti, S. venatoria/S. iberica or S. silva DNA was detected in five samples. Sarcocystis oocysts/sporocysts were observed in 11/106 faecal and 6/20 intestinal fox samples, and in 2/12 canine samples. Sarcocystis tenella (n = 8), S. hjorti (n = 2), S. gracilis (n = 2), and S. miescheriana (n = 1) were identified in foxes, and S. gracilis (n = 2), S. capreolicanis (n = 1) and S. linearis/S. taeniata (n = 1) in dogs. This study provides first molecular evidence of S. pilosa and S. silva infection in red deer and S. linearis/S. taeniata in dogs and represents the first record of S. ovalis transmitted by corvids in Central Europe. Although Sarcocystis species infecting red deer are not regarded as zoonotic, the affected carcasses can be declared as unfit for human consumption due to the extensive pathological changes.Fatigue is one of the most common symptoms associated with paroxysmal nocturnal hemoglobinuria (PNH), a rare acquired disorder of hematopoietic stem cells. While it directly impairs lifestyle leading to poor quality of life (QOL), it is not well recognized that fatigue could not depend on the disease activity or percentage of glycosylphosphatidylinositol-deficient granulocyte. We describe the case of a 78-year-old woman, with mild hemolysis and a 20-year history of severe sustained fatigue, whose QOL drastically improved with eculizumab followed by ravulizumab. We also used the novel aplastic anemia and PNH specific QOL tool to evaluate multiple statuses of the patient.Cytokine storm induced by the coronavirus 19 (COVID-19) profoundly activates the coagulation cascade causing venous thromboembolism (VTE). Initial studies from Wuhan, China showed increased incidence of VTE in patients with no standard deep vein thrombosis (DVT) prophylaxis in COVID-19 pneumonia patients. Few have argued for high intensity or intermediate DVT prophylaxis in COVID-19 patients with the incidence of VTE ranging from 16 to 27% despite standard DVT prophylaxis. However, no guideline recommendations presently exist to prescribe augmented DVT prophylaxis in these patients due to lack of evidence although the risk of VTE was clearly demonstrated. While there are ongoing trials to demonstrate the efficacy of intermediate dosing against standard DVT prophylaxis in the prevention of VTE, we present a 36-year-old male admitted with COVID-19 pneumonia who developed acute high-risk pulmonary embolism (PE) requiring emergent thrombolytic therapy despite intermediate dosing DVT prophylaxis.Pulmonary arteriovenous malformations (PAVMs) are rare vascular structures providing direct capillary-free communications between pulmonary arteries and veins. Embolotherapy is indicated as a front-line therapy. We report an unusual long-term complication of coil embolization for a 44-year-old woman with hereditary hemorrhagic telangiectasia (HHT) who had repeatedly undergone the procedures for her PAVMs. She expectorated the coil which had been placed 12 years earlier and migrated to the bronchus according to the chest radiogram and bronchoscopy. Histology of the resected lung segment suggested the cavity communicating with the bronchus was the consequence of abscess formation around the coils. Even after technically successful embolization to PAVMs, long term follow-up should be necessary paying attention to the symptoms and imaging to avoid massive hemoptysis and subsequent emergency surgery.
