Obesity can cause chronic metabolic disorders such as type 2 diabetes, hyperlipidemia, and nonalcoholic fatty liver diseases. The aim of this study was to investigate the antiobesity and antidiabetic effects of the dairy bacterium P. freudenreichii MJ2 isolated from raw milk using 3T3-L1 cells and high-fat diet (HFD)-induced obese mice. Lipid accumulation and the expression levels of genes related to lipid metabolism, such as preadipocytic gene (Pref-1), adipogenic genes (PPARγ and C/EBPα), and lipogenic genes (FAS, SCD-1, and ACC), significantly decreased in heat-killed P. freudenreichii MJ2 (hkMJ2)-treated adipocytes. Live P. freudenreichii MJ2 (MJ2), hkMJ2, and Lactobacillus plantarum (LP) decreased body weight gain in HFD-induced obese mice compared with the model group. The liver and epididymal white adipose tissue weights in the MJ2-, hkMJ2- and LP-treated groups were significantly lower than those in the model group. PF573228 The expression levels of genes and proteins related to adipogenesis and lipogenesis significantly decreased and lipolysis (HSL and ATGL) increased in the MJ2-, hkMJ2-, and LP-treated groups. The expression levels of genes related to fatty acid β-oxidation (CPT-1α and ACOX1) increased in the MJ2-, hkMJ2-, and LP-treated groups. In addition, blood glucose and fasting insulin levels in the MJ2- and hkMJ2-treated groups decreased compared with those in the model group. In conclusion, both MJ2 and hkMJ2 alleviate obesity and metabolic syndrome.We designed and fabricated a photonic crystal surface emitting laser (PCSEL) with vertically integrated diffractive optical elements on their top to study the mechanism of static beam steering on a single chip. The deflected output beam by the self-formed periodic ITO cladding layer of the PCSEL can be further steered by changing the grating period and azimuthal angle of the diffractive gratings relative to the photonic crystal. Through the analysis of photonic band structure and lasing characteristics, the periodic ITO structure is coupled to the photonic crystal band, whereas the integrated grating serves the diffractive function only. The findings pave the way for the design of PCSELs enabling single or multiple output beam with varying direction capability. This process generates heterogeneity. Recently, we proposed a random effect model meta-regression method (MR) to study the effect of single nucleotide polymorphism (SNP)-environment interactions. This method takes heterogeneity into account and produces high power. We also proposed a fixed effect model overlapping MR in which the overlapping data is taken into account. In the present study, a random effect model overlapping MR that simultaneously considers heterogeneity and overlapping data is proposed. This method is based on the random effect model MR and the fixed effect model overlapping MR. A new way of solving the logarithm of the determinant of covariance matrices in likelihood functions is also provided. Tests for the likelihood ratio statistic of the SNP-environment interaction effect and the SNP and SNP-environment joint effects are given. In our simulations, null distributions and type I error rates were proposed to verify the suitability of our method, and powers were applied to evaluate the superiority of our method. Our findings indicate that this method is effective in cases of overlapping data with a high heterogeneity.AlphaB crystallin (αB-crystallin) is a key protein for maintaining the long-term transparency of the eye lens. In the eye lens, αB-crystallin is a "dynamical" oligomer regulated by subunit exchange between the oligomers. To elucidate the unsettled mechanism of subunit exchange in αB-crystallin oligomers, the study was carried out at two different protein concentrations, 28.5 mg/mL (dense sample) and 0.45 mg/mL (dilute sample), through inverse contrast matching small-angle neutron scattering. Interestingly, the exchange rate of the dense sample was the same as that of the dilute sample. From analytical ultracentrifuge measurements, the coexistence of small molecular weight components and oligomers was detected, regardless of the protein concentration. The model proposed that subunit exchange could proceed through the assistance of monomers and other small oligomers; the key mechanism is attaching/detaching monomers and other small oligomers to/from oligomers. Moreover, this model successfully reproduced the experimental results for both dense and dilute solutions. It is concluded that the monomer and other small oligomers attaching/detaching mainly regulates the subunit exchange in αB-crystallin oligomer.Nitrate pollution in groundwater is a serious problem in many parts of the world. However, due to the diffuse and common spatially over-lapping character of potential several non-point pollution sources, it is often difficult to distinguish main nitrate sources responsible for the pollution. For this purpose, we present a novel methodology applied to groundwater for an intensely polluted area. Groundwater samples were collected monthly from April 2017 to March 2018 in Shimabara City, Nagasaki, Japan. Soil samples were collected seasonally at soil surface and 50 cm depth at 10 locations during the same period. Sequential extraction by water and extract agents was performed using calcium phosphate for anions and strontium chloride for cations. Mean nitrate concentration in groundwater close to a livestock waste disposal site (hereinafter called "LWDS") was 14.2 mg L-1, which is exceeding Japanese drinking water standards (10 mg L-1). We used coprostanol concentration, which is a fecal pollution indicator, to identify pollution sources related to livestock waste. For this purpose, we measured coprostanol (5β) and cholestanol (5α) and then calculated the sterol ratio (5β/(5β + 5α)). The ratios for three groundwater sampling sites were 0.28, 0.26, and 0.10, respectively. The sterol ratios indicated no pollution ( less then  0.3). However, the detection of coprostanol originating from animal and human waste showed that groundwater was clearly affected by this pollution source. Nitrate levels in the soil were relatively high in samples collected close to the LWDS and coprostanol contents were affected by livestock waste. Soil and groundwater nitrate concentrations displayed a complex but strong relationship. Nitrate contents were shown to be transported downstream from source areas in both soil and groundwater.
