However, processing of the MRI data to obtain muscle-perfusion maps is time-consuming. This study proposes to substantially accelerate the mapping of muscle perfusion using a deep-learning method called artificial neural network (NN). Forty-eight MRI scans were acquired from 21 healthy subjects and patients with peripheral artery disease (PAD). For optimal training of NN, different training-data sets were compared, investigating the effect of data diversity and reference perfusion accuracy. Reference perfusion was estimated by tracer kinetic model fitting initialized with multiple values (multigrid model fitting). Result The NN method was much faster than tracer kinetic model fitting. To generate a perfusion map of matrix 128 × 128 on a same computer, multigrid model fitting took about 80 min, single-grid or regular model fitting about 3 min, while the NN method took about 1 s. Compared to the reference values, NN trained with a diverse group gave estimates with mean absolute error (MAE) of 15.9 ml/min/100g and correlation coefficient (R) of 0.949, significantly more accurate than regular model fitting (MAE 22.3 ml/min/100g, R 0.889, p less then .001). Conclusion the NN method enables rapid perfusion mapping, and if properly trained, estimates perfusion with accuracy comparable to multigrid model fitting.
Alport syndrome is a progressive hereditary kidney disease clinically presenting with haematuria, proteinuria, and early onset end-stage renal disease, and often accompanied by hearing loss and ocular abnormalities. The inheritance is X-linked in the majority of families and caused by sequence variants in the COL4A5 gene encoding the α5-chain of type-IV collagen. BSJ-03-123 The proportion of de novo COL4A5 sequence variants in X-linked Alport syndrome has been reported between 12 and 15% in previous studies.
In the present study we have systematically investigated the mosaic status of asymptomatic parents of six patients with X-linked Alport syndrome using next-generation sequencing of DNA extracted from different tissues. The deleterious COL4A5 sequence variants in these patients were previously assumed to be de novo, based on Sanger sequencing of the parents.
A low-grade (1%) parental mosaicism was detected in only one out of six families (17%). In addition, in one out of six families (17%), we found that the mutational event probably occurred postzygotic.
These findings highlight the importance of testing for mosaicism in unaffected parents of patients with sequence variants considered to be de novo, as it may have implications for the recurrence risk and thereby for the genetic counseling of the family.

In this study, resinol lignin models (syringaresinol and pinoresinol) and eucalyptus alkali lignin were treated under acid-catalyzed phenolation conditions to investigate the products derived from resinol (β-β) structures of lignins. The phenolation products were characterized by means of GC-MS and NMR spectroscopy following separation using flash chromatography and thin-layer chromatography. A series of new naphthalene products were identified from phenolation of syringaresinol, and the corresponding guaiacyl analogs were also identified by GC-MS. The C1-Cα bond of these resinol compounds was cleaved to release syringol or guaiacol during phenolation. In addition, diphenylmethane products formed from phenol or phenol and syringol/guaiacol were found in the phenolation products. Comparatively, more naphthalene products were obtained by phenolation from syringaresinol than those obtained from pinoresinol. HSQC NMR characterization of the phenolated alkali lignin revealed that naphthalene structures formed in the phenolated lignin.
In academic medicine, journal article authorship is central to career advancement and promotion. This study aimed to examine the contemporary representation of women as first and senior authors of rheumatology original research articles.
The gender of first and senior author, disease category, research design and funding source were extracted from rheumatology original research articles published in high impact rheumatology and general medical journals between 2015 and 2019.
7,651 original research articles were included in the analysis. In total, there were 51.5% [95% CI 50.4%, 52.6%] articles with women first authors and 35.3% [95% CI 34.2%, 36.4%] with women senior authors. Women were significantly less likely to be first and senior authors of articles reporting randomized controlled trials compared with other clinical research designs (P<0.001), and of articles reporting industry-funded/industry-initiated studies compared with studies not funded by industry (P≤0.01). For articles reporting industry-funded/industry-initiated randomized controlled trials, women were first authors in 18.5% [95% CI 13.8%, 24.0%] and senior authors in 23.9% [95% CI 18.6%, 29.8%].
In rheumatology research articles, there is gender parity for first authorship, but women are under-represented in senior authorship positions. Under-representation of women in authorship is particularly apparent in articles reporting randomized controlled trials, and especially those that are initiated by industry.
