Protein- and lipid oxidation were investigated in whey protein based oleogels with varying water addition. Lipid oxidation was low (~30 mmol O2/kg lipid hydroperoxides after 6 weeks) in gels with 2.4% water addition fluorescence (excitation 325 nm / emission 410 nm) as indicator of tyrosine oxidation and carbonyl content significantly increased and remained at low levels in oleogels with less then 0.23% water addition. Primary amines as indicator for protein backbone breakage increased in early stages of oxidation in high water oleogels and decreased after 28 days. Degradation has been suggested to occur through interactions with reactive secondary lipid oxidation products and was confirmed by spiking experiments using respective compounds. The results suggest that secondary lipid oxidation markers are masked dependent on water addition in the presence of proteins.Glycation between target proteins and saccharides is time-consuming or requires high temperatures. Here, a promising reaction medium, natural deep eutectic solvents (NADES), for glucose glycation with bovine serum albumin (BSA) was applied to improve the grafting of glucose-glycated BSA by shifting reaction equilibrium. Two types of glucose-glycated BSA products were prepared using NADES and water systems. SDS-PAGE and MALDI-TOF-MS revealed that BSA and glucose were covalently bonded. Compared with in water system, glycated BSA products in NADES system had more -OH groups, more disordered secondary structures, lower intrinsic fluorescence intensity, and higher ultraviolet-visible absorption. Lower surface hydrophobicity (1100 versus 1356), higher emulsifying activity index (66.17 versus 46.49 m2/g), higher emulsion stability index (79.62 versus 63.61%), and lesser free sulfhydryl (8.07 versus 8.98 μmol/g) groups were obtained with NADES system than with water system. The results suggest that NADES is a suitable alternative reaction medium for promoting the glycation of BSA.Stable carbon (δ13C) and nitrogen (δ15N) isotope ratios and multi-element signatures of pooled fish scales were used to differentiate wild, lake-farmed and pond-farmed carp from Dongting Lake, China. Fish scales were found to be important archives for the dietary history and trophic level of wild and farmed fish, indicating their food sources. δ13C and δ15N values of pond-farmed carp scales were comparatively enriched to wild and lake-farmed carp due to their animal protein-derived feeds. Multi-element compositions of fish scales also showed significant differences between wild and farmed fish. A partial least squares discriminant analysis (PLS-DA) model correctly discriminated the three carp groups. Discrimination accuracies of wild, lake-farmed and pond-farmed carp were 100%, 95%, and 100% for the training set, and 100% for the testing set. This strategy provides a promising non-lethal alternative method to combat mislabeling of freshwater carp from different farming methods.Sudan I is a synthetic-azo dye commonly used to adulterate foods to increase sensory appearance. However, it is banned due to its carcinogenic, mutagenic and genotoxic properties, which represent a serious risk to human health. Thus, this paper proposes a feasibility study to identify and quantify Sudan I dye in ketchup samples using colour histograms (obtained from digital images) and multivariate analysis. The successive projections algorithm coupled with linear discriminant analysis (SPA-LDA) classified correctly all samples, while the partial least squares coupled with SPA for interval selection (iSPA-PLS) quantified adequately the adulterant, attaining values of RMSEP of 11.64 mg kg-1, R2 of 0.96, RPD of 5.28, REP of 13.63% and LOD of 39.45 mg kg-1. Therefore, the proposed methodology provides a simple, fast, inexpensive, promising analytical tool for the screening of both the quality and safety of ketchup samples. As a consequence, it can help to protect the consumer's health. The tunillo (Stenocereus stellatus [Pfeiffer] Riccobono) is a relatively little known cactus fruit with a significant pharmacological potential. However, all currently known variants are identified visually mostly on the basis of pulp color. Differences in chemical composition and pharmacological properties also remain largely unknown. Support vector machine classifiers were applied to UV-Visible spectra of liquid samples to obtain the following, color-based categories of tunillo fruits A1-white, A2-red, A3-purple, and A4-orange. The spectrum of A2-red could be duplicated by combining those from A3-purple and A4-orange, while UPGMA-based hierarchical clustering of psbA-trnH and matK suggested that certain differences in color might actually have a genetic basis. The pigment quantification established A2-red and A3-purple as the most suitable candidates for the extraction of betalains and complex colored matrices, respectively. A2-red also had the highest content of phenols and flavonoids and displayed a noticeable anti-hyperglycemic effect. Objectives Resistance training combined with consumption of a high-protein diet (HPD) is typically recommended to increase muscle mass, as both acute resistance exercise (RE) and dietary protein intake stimulate mechanistic target of rapamycin complex 1 (mTORC1) and muscle protein synthesis (MPS). However, the effect of chronic HPD consumption on MPS response to an acute RE remains to be determined. Methods Male Sprague-Dawley rats aged 10 wk were fed HPD (50 kcal % protein, for 4 wk) or normal protein diet (NPD; 20 kcal % protein). After the 4-wk dietary intervention, the rats were fasted overnight and the right gastrocnemius muscle was subjected to percutaneous electrical stimulation to mimic acute RE, whereas the left gastrocnemius muscle served as control. The rats were sacrificed 6 h after exercise and the tissues were sampled immediately. Results The HPD group showed significantly lower fat mass and higher skeletal muscle mass than the NPD group without affecting body weight. Resting mTORC1 activity did not differ between the groups. Additionally, resting MPS was also unchanged after HPD. Acute RE significantly increased mTORC1 activity and MPS in both groups. However, differences in diet did not influence the response of mTORC1 activation to acute RE. check details Furthermore, HPD did not affect the response of MPS to acute RE. Conclusion The present results suggested that although 4 wk of HPD reduces body fat and increases skeletal muscle mass, it does not affect muscle protein synthesis at basal state, and in response to acute RE.
