Regenerative methods for IVD degeneration are largely cell-therapy-based or involve viral vectors, which are associated with mutagenesis and undesired immune responses. The present study used bulk electroporation and engineered extracellular vesicles (EVs) to deliver forkhead-box F1 (FOXF1) mRNA to degenerate human nucleus pulposus (NP) cells as a minimally invasive therapeutic strategy for IVD regeneration. Bulk electroporation was used to investigate FOXF1 effects on human NP cells during a 4-week culture in 3D agarose constructs. Engineered EV delivery of FOXF1 into human IVD cells in monolayer was determined, with subsequent in vivo validation in a pilot mouse IVD puncture model. FOXF1 transfection significantly altered gene expression by upregulating healthy NP markers [FOXF1, keratin 19 (KRT19)], decreasing inflammatory cytokines [interleukin (IL)-1β, -6], catabolic enzymes [metalloproteinase 13 (MMP13)] and nerve growth factor (NGF), with significant increases in glycosaminoglycan accumulation in human NP cells. Engineered EVs loaded with FOXF1 demonstrated successful encapsulation of FOXF1 cargo and effective uptake by human NP cells cultured in monolayer. Injection of FOXF1-loaded EVs into the mouse IVD in vivo resulted in a significant upregulation of FOXF1 and Brachyury, compared to controls at 7 d post-injection, with no evidence of cytotoxicity. This is the first study to demonstrate non-viral delivery of FOXF1 and reprogramming of human NP cells in vitro and mouse IVD cells in vivo. This strategy represents a non-addictive approach for treating IVD degeneration and associated back pain.
Properly executed hand washing by food service employees can greatly minimize the risk of transmitting foodborne pathogens to food and food contact surfaces in restaurants. However, food service employee hand washing is often not done correctly or does not occur as often as it should. The purpose of this study was to assess the relative impact of (i) the convenience and accessibility of hand washing facilities; (ii) the maintenance of hand washing supplies, (iii) multiunit status, (iv) having a certified food protection manager, and (v) having a food safety management system for compliance with proper hand washing. Results revealed marked differences in hand washing behaviors between fast-food and full-service restaurants; 45% of 425 fast-food restaurants and 57% of 396 full-service restaurants were out of compliance for washing hands correctly, and 57% of fast-food restaurants and 78% of full-service restaurants were out of compliance for employee hands being washed when required. Logistic regression results indicated the benefits of accessibility and maintenance of the hand washing sink and of a food safety management system for increasing the likelihood of employees washing hands when they are supposed to and washing them correctly when they do.

Bacillus cereus is one of the important foodborne pathogens that can be found in various foodstuffs, causes diarrheal and/or emetic syndromes, and can cause severe systemic diseases that may lead to death. Of 165 food samples investigated in this study, 39 (24%) were positive for B. cereus, with contamination levels of 2 to 6 log CFU/g or mL and a higher prevalence of levels >3 log CFU. Aquatic products are favored by people all over the world, but the potential quality and safety issues cannot be ignored. To determine the risk of veterinary drug residues in aquatic products in the Yangtze River Delta, this study used geographic information system method to analyze Chinese veterinary drugs in aquatic products in Shanghai, Jiangsu, Zhejiang, and Anhui (Yangtze River Delta Urban Agglomerations) from 2017 to 2019. A study of the spatial distribution pattern, hot spot detection and analysis, and spatiotemporal cluster analysis of the residual excess rate and detection rate showed a random spatial distribution in the overall excess rate and detection rate of veterinary drug residues in aquatic products from 2017 to 2019. The results of hot spot analysis and spatiotemporal cluster analysis showed that the rate of detection of veterinary drug residues and the rate of detection of residues in excess of regulatory standards were clustered. This study provides a scientific basis for food safety evaluation and risk management suggestions.

In most developing countries, mining is one of the major sources of toxic heavy metals, some of which are a major health concern to the surrounding populace through soil pollution and food contamination. We investigated the levels of Ti, Mn, Ni, Cu, Zn, Pb, Rb, Sr, Se, Nb, and V in 64 soil samples, 320 samples of regularly consumed staple food crops and vegetables (Manihot esculenta [cassava] tubers, Dioscorea rotundata [white yam], Arachi hypogea [groundnut] seed, Telfariria occidentalis [fluted pumpkin leaf]), and leaves of the medical plant Zanthoxulum zanthoxyloides on farmlands near Enyigba mining sites in Ebonyi state, Nigeria. We also examined the possible health risks due to consumption of these staple foods. Energy dispersive X-ray fluorescence was used to determine the metals present in the samples. The mean levels of metals, except for Mn, Pb, and Zn, were below the threshold limits recommended by various institutions for agricultural soils. Our data also showed that food crops cultivated near these sites bioaccumulated toxic levels of these metals above the proposed values set by the World Health Organization, National Agency for Food and Drug Administration and Control, and Codex Alimentarius Commission, especially for Pb, Zn, Cu, Ni, Se, and Rb.
