The embryonic rat dorsal root ganglion (DRG) neuron-derived 50B11 cell line is a promising sensory neuron model expressing markers characteristic of NGF and GDNF-dependent C-fibre nociceptors. Whether these cells have the capacity to develop into distinct nociceptive subtypes based on NGF- or GDNF-dependence has not been investigated. Here we show that by augmenting forskolin (FSK) and growth factor supplementation with NGF or GDNF, 50B11 cultures can be driven to acquire differential functional responses to common nociceptive agonists capsaicin and ATP respectively. In addition, to previous studies, we also demonstrate that a differentiated neuronal phenotype can be maintained for up to 7 days. Western blot analysis of nociceptive marker proteins further demonstrates that the 50B11 cells partially recapitulate the functional phenotypes of classical NGF-dependent (peptidergic) and GDNF-dependent (non-peptidergic) neuronal subtypes described in DRGs. Further, 50B11 cells differentiated with NGF/FSK, but not GDNF/FSK, show sensitization to acute prostaglandin E2 treatment. Finally, RNA-Seq analysis confirms that differentiation with NGF/FSK or GDNF/FSK produces two 50B11 cell subtypes with distinct transcriptome expression profiles. Gene ontology comparison of the two subtypes of differentiated 50B11 cells to rodent DRG neurons studies shows significant overlap in matching or partially matching categories. This transcriptomic analysis will aid future suitability assessment of the 50B11 cells as a high-throughput nociceptor model for a broad range of experimental applications. In conclusion, this study shows that the 50B11 cell line is capable of partially recapitulating features of two distinct types of embryonic NGF and GDNF-dependent nociceptor-like cells.Background The association between regional volumes and resting-state functional networks was tested within the default-mode network (DMN), influenced by Alzheimer pathology, salience network (SalN), not under similar pathological influence, and sensorimotor network (SMN), usually spared by pathology. Methods A total of 148 participants, with Alzheimer's disease (AD) dementia, mild cognitive impairment (MCI), and healthy controls underwent multimodal brain magnetic resonance imaging (MRI). Functional network identification was achieved with group-level independent-component analysis of functional MRI (fMRI) scans. T1 weighted images were also analyzed. Ten regions of interest (ROI) were defined in core hubs of the three networks. Gray-matter volume/functional network strength association was tested within-ROI and cross-ROI in each group by using partial-correlation models and ROI-to-ROI, ROI-to-voxel, and voxel-to-voxel correlations. Results In controls, a negative association was found between right inferior-parietal volumes and SMN expression in the left precentral gyrus, as revealed by ROI-to-ROI models. In AD, DMN expression was positively associated with the volume of the left insula and the right inferior parietal lobule, and SalN expression was positively associated with volume of the left inferior parietal lobule. ROI-to-voxel models revealed significant associations between the volume of the posterior cingulate cortex and SMN expression in sensorimotor and premotor regions. No significant findings emerged in the MCI nor from voxel-to-voxel analyses. Discussion Regional volumes of main network hubs are significantly associated with hemodynamic network expression, although patterns are intricate and dependent on diagnostic status. Since distinct networks are differentially influenced by Alzheimer pathology, it appears that pathology plays a significant role in influencing the association between regional volumes and regional functional network strength.The aim of this study was to analyze the influence of reproductive aging on executive functions. We assessed executive functions in three groups of healthy women in the premenopausal (n = 45, mean age = 30.89, SD = 10.5), perimenopausal (n = 31, mean age = 50.06, SD = 3.6) and postmenopausal (n = 24, mean age = 63.39, SD = 6.5) phase. No differences between groups were observed in working memory, verbal fluency, inhibitory control, planning, and cognitive flexibility. However, when the analyses were repeated with participants with occupations with lower intellectual demands, perimenopausal and postmenopausal women performed worse than premenopausal women in semantic verbal fluency. This study provides important evidence to understand the effects of reproductive aging on cognitive performance in healthy women. Our findings indicate that cognitive reserve-related factors may be important to understand the differences in executive functions associated with reproductive aging.In the period 1 January 2012 to 30 June 2020, 76 doctors whose names/entries had been erased from the UK Medical Register by a disciplinary tribunal applied for restoration, and 23 out of 76 (30.3%) applications granted. In 5 of the 53 of those refused restoration, the tribunal suspended indefinitely the right to make further applications. The most frequent reasons for refusal were failure to demonstrate insight (seen in 96%), failure to demonstrate remediation (seen in 79%), and failure to demonstrate that knowledge and skills were up to date (24.5%). Success was more common in UK graduate applications (14/29 - 48.3%) than non-UK graduate applications (9/37 - 24.3%), and in those legally represented (16/29 - 55.2%) than in those without legal representation (7/29 - 24.1%), but the data does not indicate the reasons for these differences. Disciplinary erasure need not necessarily be for life as doctors who learn from their experience, change their ways, and provide evidence of genuine insight and remediation along with up to date knowledge and skills can successfully be reinstated on the register.I consider the Doerig, Schruger, Herzog (this issue) paper to be a very important paper on the multiple empirical theories of consciousness (ToCs). MS-L6 cell line The paper compared 13 different approaches. It would be wonderful to create a forum for all interested parties to discuss the many issues this paper brought up. Equally important is what the paper didn't bring up. Namely, how do the different empirical approaches connect to qualia, the subjective aspects of consciousness. In my mind, qualia is the biggest mystery facing present science. It would be good to see how the multiple interpretations might connect to qualia.
