Therapies targeting the immune checkpoint molecules programmed death ligand 1 (PD-L1) and indoleamine 2,3-dioxygenase 1 (IDO1) have been explored in various malignant tumours. In this study, we examined the relationship between PDL-1, IDO1 and JAK2 expression and the roles of these signal pathways in soft tissue leiomyosarcoma (LMS).

The next-generation sequencing data of 53 patients with LMS were obtained from an online public database and were used to assess PD-L1, IDO1 and JAK2 gene amplification and mRNA expression. Then, we determined the relationship between JAK-STAT pathway activation and PD-L1 and IDO1 expression in a LMS cell line. In addition, immunohistochemical staining of 69 cases of LMS was performed for PD-L1, IDO1, TDO2 and phosphorylated JAK2 (pJAK2).

Comprehensive gene expression analysis using microarray and RNA-Seq data revealed that PD-L1 and IDO1 mRNA expression positively correlated with JAK2 and STAT1 mRNA expression. Two of the 53 cases exhibited PD-L1 and JAK2 gene amplification; however, they were not related to their gene expression. Etanercept mw LMS cell line analysis revealed that IFN-γ supplementation induced IDO1 and PD-L1 expression; these effects were suppressed by JAK inhibition. Immunohistochemical analysis of the resected specimens revealed that TDO2 expression positively correlated with pJAK2 (P = 0.0490) and IDO1 expression (P < 0.0001). PD-L1-positive specimens tended to express pJAK2; however, the relationship did not reach statistical significance (P = 0.1477).

Viviparous species might be more susceptible to increasing temperatures because of their inability to meet their energetic demands following rapid environmental changes. Nevertheless, we do not yet fully understand how lizards with different reproductive modes can respond to climate change. We investigated the effect of both maternal thermal environment during pregnancy and incubation temperature on hatchling morphology and physiological performance of two populations of the lizard Saiphos equalis differing in their mode of reproduction, to test whether reproductive mode affects the ability to buffer against rising temperatures. Gravid females from both populations were subjected to current or projected end-of-century (future) thermal environments, to evaluate differences in the body size, growth rate, thermal preference, and locomotor performance of their offspring. Our results show that independently of the mode of reproduction, high temperatures accelerated gestation periods. Thermal environments did not affect hatchling thermal preference, but viviparous hatchlings consistently preferred lower temperatures. Unlike viviparous lizards, oviparous hatchlings incubated under future temperatures were smaller and had a lower growth rate compared to current-incubated hatchlings. Finally, thermal environments did not affect hatchling endurance and speed when controlling for body size. Our results show that global warming is likely to have a negative impact on S. equalis, but suggest that some of its effects may be ameliorated by maternal responses during pregnancy, particularly in viviparous populations.Providencia rettgeri infection has occurred occasionally in aquaculture, but is rare in turtles. Here, a pathogenic P. rettgeri strain G0519 was isolated from a diseased slider turtle (Trachemys scripta) in China, and qPCR assay was established for the RTX toxin (rtxD) gene. To date, little is known about the potential influencing factors on the outcome in small (< 2cm) umbilical hernia repair. This multivariable analysis of data from the Herniamed Registry now aims to assess these factors.

The data of patients with primary elective umbilical hernia repair and defect size < 2cm entered into the Herniamed Registry from September 1, 2009 to December 31, 2018 were analyzed to assess through multivariable analysis all confirmatory pre-defined potential influencing factors on the primary outcome criteria intraoperative and postoperative complications, general complications, complication-related reoperations, recurrence rate and rates of pain at rest, pain on exertion and chronic pain requiring treatment at 1-year follow-up.
