TRPV1 activation induced only a small and transient increase in DAG levels, unlike the robust and more sustained increase induced by muscarinic receptor activation. DAGK inhibition substantially increased the DAG signal evoked by TRPV1 activation, but not that evoked by M1 muscarinic receptor activation. Our results show that Ca2+influx through TRPV1 activates both PLC and DAGK enzymes and that the latter limits the formation of DAG and thereby negatively regulates TRPV1 channel activity. Our findings uncover a role for DAGK in ion channel regulation. GA homeostasis is achieved via complex biosynthetic and catabolic pathways, whose exact activities remain to be elucidated. Here, we isolated two cDNAs from mature or imbibed cucumber seeds with high sequence similarity to known GA 3-oxidases. We found that one enzyme (designated here CsGA3ox5) has GA 3-oxidation activity. However, the second enzyme (designated CsGA1ox/ds) performed multiple reactions, including 1β-oxidation and 9,11-desaturation of GAs, but was lacking the 3-oxidation activity. CsGA1ox/ds overexpression in Arabidopsis plants resulted in severely dwarfed plants that could be rescued by the exogenous application of bioactive GA4, confirming that CsGA1ox/ds catabolizes GAs. Substitution of three amino acids in CsGA1ox/ds, Phe-93, Pro-106, and Ser-202, with those typically conserved among GA 3-oxidases, Tyr-93, Met-106, and Thr-202, respectively, conferred GA 3-oxidase activity to CsGA1ox/ds and thereby augmented its potential to form bioactive GAs besides catabolic products. Accordingly, overexpression of this amino acid-modified GA1ox/ds variant in Arabidopsis accelerated plant growth and development, indicating that this enzyme variant can produce bioactive GAs in planta. mTOR inhibition Furthermore, a genetically modified GA3ox5 variant in which these three canonical GA 3-oxidase amino acids were changed to the ones present in CsGA1ox/ds was unable to convert GA9 to GA4, highlighting the importance of these three conserved amino acids for GA 3-oxidase activity. We propose an appeal-aversion assessment tool to help regulators choose among graphic packaging options. METHODS After familiarisation with different cigarette packaging, adult daily smokers (n=338) from San Diego, California, USA completed a discrete choice appeal-aversion purchasing task and provided information on nicotine dependence and sociodemographics (2017-2019). The conjoint analysis estimated the importance and price utility for product attributes (ie, packaging, price, tobacco origin and quitline number). The price premiums that smokers would be willing to pay to avoid purchasing graphic packaging were calculated. RESULTS Among purchase determinants, the price was the most important attribute (65.5%), followed by packaging design (27.1%). Compared with blank packaging without marketing, branded industry packs had appeal valuations (US$0.54; 95% CI US$0.44 to US$0.65), whereas graphic warning packs had aversion valuations that varied with the salience of the image (blindness=-US$2.53, 95% CI -US$2.76 to -US$2.31; teeth damage=-US$2.90, 95% CI -US$3.17 to -US$2.63; and gangrenous foot=-US$3.70, 95% CI -US$4.01 to -US$3.39). The aversion was such that 46.2% of participants were willing to pay a 50+% premium over their current cigarette price to have their branded packs rather than a graphic pack. These appeal-aversion valuations were moderated by sex, income and nicotine dependence (p less then 0.05). CONCLUSIONS Smokers indicated a willingness to pay substantial premiums to avoid purchasing graphic packaging. Results suggest that mandating graphic warnings on US cigarette packs would induce price aversion and may deter cigarette purchasing. Published by BMJ.BACKGROUND Near-infrared fluorescence (NIRF) imaging is a relatively novel technique that can aid surgeons during intraoperative tumour identification. METHODS Nine canine oncology patients (five mammary gland tumours, three mast cell tumours and one melanoma) received intravenous indocyanine green (ICG). After 24 hours, tumours were resected and fluorescence intensities of tumours and surroundings were evaluated. Additional wound bed tissue was resected if residual fluorescence was present after tumour resection. Ex vivo, fluorescence-guided dissection was performed to separate tumour from surrounding tissue. RESULTS Intraoperative NIRF-guided tumour delineation was feasible in four out of nine dogs. Wound bed imaging after tumour removal identified nine additional fluorescent lesions, of which four contained tumour tissue. One of these four true positive in vivo lesions was missed by standard-of-care inspection. Ex vivo fluorescence-guided tumour dissection showed a sensitivity of 72 per cent and a specificity of 80 per cent in discriminating between tumour and surrounding tissue. CONCLUSION The value of ICG for intraoperative tumour delineation seems more limited than originally thought. Although NIRF imaging using ICG did identify remaining tumour tissue in the wound bed, a high false positive rate was also observed. © British Veterinary Association 2020. No commercial re-use. See rights and permissions. Published by BMJ.BACKGROUND Excessive fear in farm animals can lead to chronic stress and thus impair animal welfare. In laying hens, fear responses in several behavioural tests have also been associated with the occurrence of feather pecking. The aim of the present study was to comparatively assess fear-related responses of conventional layer hybrids (Lohmann Brown plus, LB+) and dual-purpose hens (Lohmann Dual, LD), which seem to be less prone to injurious pecking. METHODS A novel object (NO) and an avoidance distance (AD) test were carried out in both hybrids at a group level and at different ages during the laying period in order to measure their fear-related responses. RESULTS On most study days, more LD hens approached the NO and they approached it sooner than the LB+ hens. Similarly, the LD hens retreated at smaller distances from a human being in the AD test. CONCLUSION The results indicate that dual-purpose hens act less fearful in the performed behavioural tests compared with conventional layer hybrids. Therefore, dual-purpose hens might experience less stress during daily management routines, which would affect animal welfare positively.
