Temporal lobe epilepsy (TLE) may represent an increased risk of developing depression, especially if the seizures do not generalize. The two-pore domain potassium channel-TWIK-related K+ channel (TREK-1) plays important roles in both epilepsy and depression. However, the changes in its expression in patients with epilepsy with comorbid depression remain unclear. In the present study, we analyzed depressive symptoms using neuropsychiatric scales in forty-two patients with drug-resistant TLE, who also underwent EEG in waking and sleeping states, as well as 3.0 T brain MRI. We tested for TREK-1 positive neurons and microglial cells in the anterior hippocampi of patients with drug-resistant TLE with and without comorbid depression (n=5/group). Approximately 31% of patients with TLE had comorbid depression (13/42). Meanwhile, the patients who had hippocampal sclerosis had much higher scores on the depression rating scale. The results indicated the contribution of hippocampal sclerosis to the development of depression. Immunostaining of TREK-1 channels was observed in neurons and glia in the anterior hippocampus. Increased immunoreactivity of TREK-1 neurons was observed in the hippocampi of patients with TLE with comorbid depression compared with nondepressed patients with TLE. TREK-1 was expressed in almost all microglia. Curiously, more activated TREK-1-positive microglia were observed in patients with TLE with depression than in those without depression. The results suggested that a change in TREK-1 immunoreactivity was involved, at least partly, in the development of depression as a comorbidity of TLE. Imbalance of the TREK-1 channel may be a potential target for the treatment of patients with epilepsy with comorbid depression.Toxicity in aquatic plants, caused by excess ammonium in the environment, is an important ecological problem and active research topic. Recent studies showed the importance of the enzyme Glutamate Dehydrogenase (GDH) in detoxifying ammonium. However, these results mainly derived from species comparisons, hence some mechanisms may have been obscured due to species differences. Our recent finding that young leaves of Potamogeton lucens were less sensitive to ammonium enrichment, than mature leaves allowed us to study ammonium detoxification within a species. We found that, unlike mature leaves, ammonium-tolerant young leaves of P. lucens could assimilate ammonium mainly through GDH. There was a 38% increase of NADH-dependent GDH in 50 mg/L ammonium concentration compared with 0.1 mg/L. Therefore, this study confirms the hypothesis that the GDH pathway plays a major role in the detoxification of ammonium in freshwater macrophytes.Plants native to extreme habitats often face changes in environmental conditions such as salinity level and water availability. In response, plants have evolved efficient mechanisms allowing them to survive or recover. In the present work, effects of high salinity and salt-stress release were studied on the halophyte Cakile maritima. Four week-old plants were either cultivated at 0 mM NaCl or 200 mM NaCl. After one month of treatment, plants were further irrigated at either 0 mM NaCl, 200 mM NaCl, or rewatered to 0 mM NaCl (stress release). Upon salt stress, C. maritima plants exhibited reduced biomass production and shoot hydration which were associated with a decrease in the amount of chlorophyll a and b. However, under the same stressful conditions a significant increase of anthocyanin and malonyldialdehyde concentrations was noticed. Trometamol mouse Salt-stressed plants were able to maintain stable protein complexes of thylakoid membranes. Measurement of chlorophyll fluorescence and P700 redox state showed that PSI was more susceptible for damage by salinity than PSII. PSII machinery was significantly enhanced under saline conditions. All measured parameters were partially restored under salt-stress release conditions. Photoinhibition of PSI was also reversible and C. maritima was able to successfully re-establish PSI machinery indicating the high contribution of chloroplasts in salt tolerance mechanisms of C. maritima. Overall, to overcome high salinity stress, C. maritima sets a cascade of physio-biochemical and molecular pathways. Chloroplasts seem to act as metabolic centers as part of this adaptive process enabling growth restoration in this halophyte following salt stress release.Drought tolerant plant growth-promoting rhizobacteria (PGPR) can confer drought tolerance in plants, when inoculated, and this effect can be more pronounced by their combined application with silicon oxide nanoparticles (SiO2 NPs). In this research, drought-tolerant and plant growth-promoting rhizobacterial strains were isolated from the rhizospheric soil of wheat plants growing in the arid region of Pakistan. Out of 30 isolated strains, three rhizobacterial strains were selected based on their drought tolerance, higher phytohormones (indole acetic acid (IAA), abscisic acid (ABA), and cytokinin (CK), and osmolyte (proline and sugar) production ability. These strains were identified as Bacillus sp. Azospirillum lipoferum and Azospirillum brasilense by 16S rRNA sequencing and accession numbers (MT482404, MT742664, and MT 742666, respectively) were obtained. Inoculation of these strains, alone and in combination, improved the germination attributes of wheat seeds under drought stress conditions. However, the combination of all three bacterial strains gave the best results.
