Development of thermoelectrics usually involves trial-and-error investigations, including time-consuming synthesis and measurements. Here, we identify the electronic quality factor BE for determining the maximum thermoelectric power factor, which can be conveniently estimated by a single measurement of Seebeck coefficient and electrical conductivity of only one sample, not necessarily optimized, at an arbitrary temperature. We demonstrate that thousands of experimental measurements in dozens of materials can all be described by a universal curve and a single material parameter BE for each class of materials. Furthermore, any deviation in BE with temperature or doping indicated new effects such as band convergence or additional scattering. This makes BE a powerful tool for evaluating and guiding the development of thermoelectrics. We demonstrate the power of BE to show both p-type GeTe alloys and n-type Mg3SbBi alloys as highly competitive materials, at near room temperature, to state-of-the-art Bi2Te3 alloys used in nearly all commercial applications.The surfaces of Jupiter and Saturn have magnificent vortical storms that help shape the dynamic nature of their atmospheres. Land- and space-based observational campaigns have established several properties of these vortices, with some being similar between the two planets, while others are different. Shallow-water hydrodynamics, where the vortices are treated as shallow weather-layer phenomenon, is commonly evoked for explaining their formation and properties. Here, we report novel formation mechanisms for vortices where the primary driving mechanism is the deep planetary convection occurring in these planets. Using three-dimensional simulations of turbulent convection in rotating spherical shells, we propose two ideas (i) Rotating turbulent convection generates deep axially aligned cyclones and anticyclones; (ii) a deep planetary dynamo acts to promote additional anticyclones, some as large as Jupiter's Great Red Spot, in an overlying atmospheric layer. We use these ideas to interpret several observational properties of vortices on Jupiter and Saturn.Immune checkpoint inhibitors (ICIs) show promise, but most patients do not respond. We identify and validate biomarkers from extracellular vesicles (EVs), allowing non-invasive monitoring of tumor- intrinsic and host immune status, as well as a prediction of ICI response. We undertook transcriptomic profiling of plasma-derived EVs and tumors from 50 patients with metastatic melanoma receiving ICI, and validated with an independent EV-only cohort of 30 patients. Plasma-derived EV and tumor transcriptomes correlate. EV profiles reveal drivers of ICI resistance and melanoma progression, exhibit differentially expressed genes/pathways, and correlate with clinical response to ICI. We created a Bayesian probabilistic deconvolution model to estimate contributions from tumor and non-tumor sources, enabling interpretation of differentially expressed genes/pathways. EV RNA-seq mutations also segregated ICI response. EVs serve as a non-invasive biomarker to jointly probe tumor-intrinsic and immune changes to ICI, function as predictive markers of ICI responsiveness, and monitor tumor persistence and immune activation.High-performance lightweight materials are urgently needed, given the pressing quest for weight reduction and the associated energy savings and emission reduction. Here, by incorporating the multi-principal element feature of compositionally complex alloys, we develop the concept of lightweight steels further and propose a new class of compositionally complex steels (CCSs). This approach allows us to use the high solid solution strengthening and shift the alloys' compositions into previously unattainable phase regions where both nanosized shearable κ-carbides and non-shearable B2 particles are simultaneously formed. The achievement of dual-nanoprecipitation in our CCSs leads to materials with ultrahigh specific tensile strength (up to 260 MPa·cm3 g-1) and excellent tensile elongation (13 to 38%), a combination outperforming all other high-strength high-entropy alloys and advanced lightweight steels. Topoisomerase I cleavage complexes (TOP1ccs) are trapped by camptothecin and TOP2ccs by etoposide. Proteolysis of the trapped topoisomerase DNA-protein cross-links (TOP-DPCs) is a key step for some pathways to repair these lesions. We describe a pathway that features a prominent role of the small ubiquitin-like modifier (SUMO) modification for both TOP1- and TOP2-DPC repair. Both undergo rapid and sequential SUMO-2/3 and SUMO-1 modifications in human cells. The SUMO ligase PIAS4 is required for these modifications. RNF4, a SUMO-targeted ubiquitin ligase (STUbL), then ubiquitylates the TOP-DPCs for their subsequent degradation by the proteasome. This pathway is conserved in yeast with Siz1 and Slx5-Slx8, the orthologs of human PIAS4 and RNF4.Identifying the systems-level mechanisms that lead to Alzheimer's disease, an unmet need, is an essential step toward the development of therapeutics. In this work, we report that the key disease-causative mechanisms, including dedifferentiation and repression of neuronal identity, are triggered by changes in chromatin topology. Here, we generated human induced pluripotent stem cell (hiPSC)-derived neurons from donor patients with early-onset familial Alzheimer's disease (EOFAD) and used a multiomics approach to mechanistically characterize the modulation of disease-associated gene regulatory programs. We demonstrate that EOFAD neurons dedifferentiate to a precursor-like state with signatures of ectoderm and nonectoderm lineages. RNA-seq, ATAC-seq, and ChIP-seq analysis reveals that transcriptional alterations in the cellular state are orchestrated by changes in histone methylation and chromatin topology. Furthermore, we demonstrate that these mechanisms are observed in EOFAD-patient brains, validating our hiPSC-derived neuron models. The mechanistic endotypes of Alzheimer's disease uncovered here offer key insights for therapeutic interventions.
