Childhood ependymomas are heterogenous chemoresistant neoplasms arising from aberrant stem-like cells. Epigenome deregulation plays a pivotal role in ependymoma pathogenesis, suggesting that epigenetic modifiers hold therapeutic promise against this disease. Bromodomain and extraterminal domain (BET) proteins are epigenome readers of acetylated signals in histones and coactivators for oncogenic and stemness-related transcriptional networks, including MYC/MYCN (Proto-Oncogene, BHLH Transcritpion Factor)-regulated genes. We explored BET inhibition as an anticancer strategy in a panel of pediatric patient-derived ependymoma stem cell models by OTX015-mediated suppression of BET/acetylated histone binding. We found that ependymoma tissues and lines express BET proteins and their targets MYC and MYCN. In vitro, OTX015 reduced cell proliferation by inducing G0/G1-phase accumulation and apoptosis at clinically tolerable doses. Mechanistically, inhibitory p21 and p27 increased in a p53-independent manner, whereas the proliferative driver, phospho-signal transducer and activator of transcription 3 (STAT3), decreased. Upregulation of apoptosis-related proteins and survivin downregulation were correlated with cell line drug sensitivity. Minor alterations of MYC/MYCN expression were reported. In vivo, OTX015 significantly improved survival in 2/3 orthotopic ependymoma models. BET proteins represent promising targets for pharmaceutical intervention with OTX015 against ependymoma. As a comparable quality of feces and a high nutrient digestibility were observed when rye was used in the experimental diets-it can be considered an alternative carbohydrate source in dog foods.Identifying microbial fossils in the rock record is a difficult task because they are often simple in morphology and can be mimicked by non-biological structures. Biosignatures are essential for identifying putative fossils as being definitively biological in origin, but are often lacking due to geologic effects which can obscure or erase such signs. As such, there is a need for robust biosignature identification techniques. Here we show new evidence for the application of trace elements as biosignatures in microfossils. We found elevated concentrations of magnesium, aluminum, manganese, iron, and strontium colocalized with carbon and sulfur in microfossils from Drummond Basin, a mid-Paleozoic hot spring deposit in Australia. Our results also suggest that trace element sequestrations from modern hot spring deposits persist through substantial host rock alteration. Because some of the oldest fossils on Earth are found in hot spring deposits and ancient hot spring deposits are also thought to occur on Mars, this biosignature technique may be utilized as a valuable tool to aid in the search for extraterrestrial life.Single-point mutation in the ACTIN2 gene of the der1-3 mutant revealed that ACTIN2 is an essential actin isovariant required for root hair tip growth, and leads to shorter, thinner and more randomly oriented actin filaments in comparison to the wild-type C24 genotype. The actin cytoskeleton has been linked to plant defense against oxidative stress, but it is not clear how altered structural organization and dynamics of actin filaments may help plants to cope with oxidative stress. In this study, we characterized root growth, plant biomass, actin organization and antioxidant activity of the der1-3 mutant under oxidative stress induced by paraquat and H2O2. Under these conditions, plant growth was better in the der1-3 mutant, while the actin cytoskeleton in the der1-3 carrying pro35SGFPFABD2 construct showed a lower bundling rate and higher dynamicity. Biochemical analyses documented a lower degree of lipid peroxidation, and an elevated capacity to decompose superoxide and hydrogen peroxide. These results support the view that the der1-3 mutant is more resistant to oxidative stress. We propose that alterations in the actin cytoskeleton, increased sensitivity of ACTIN to reducing agent dithiothreitol (DTT), along with the increased capacity to decompose reactive oxygen species encourage the enhanced tolerance of this mutant against oxidative stress.Probiotics have become highly recognized as supplements for poultry.Since gut health can be considered synonymous withanimal health, the effects of probiotic Slab51® on the morphology and the glycan composition of guineafowlintestine were examined. Compared with control samples, probiotic group revealed (1) significant increase in villus height (p less then 0.001 in duodenum and ileum; p less then 0.05 in caecum), crypt depth (p less then 0.001 in duodenum and caecum; p less then 0.05 in ileum) and goblet cells (GCs) per villus (p less then 0.001) in all investigated tracts; (2) increase in galactoseβl,3N-acetylgalacyosamine(Galβl,3GalNAc)terminating O-glycans and αl,2-fucosylated glycans secretory GCs in the duodenum; (3) increase in α2,6-sialoglycans and high-mannose N-linked glycans secretory GCs but reduction in GCs-secreting sulfoglycans in the ileum; (4) increase in Galβl,3GalNAc and high-mannose N-linked glycans secretory GCs and decrease in GCs-producing sulfomucins in the caecum; (5) increase in the numbers of crypt cells containing sulfate and non-sulfated acidic glycans.
