Polycystic ovary syndrome (PCOS) is the most common cause of female infertility. Growth differentiation factor-8 (GDF-8) is expressed in the ovary and can be detected in human follicular fluid which provides an important microenvironment for maintaining physiological functions of the ovarian follicle. To date, the relationship between GDF-8 levels in follicular fluid and the risk of PCOS is completely unknown. In the present study, we show that during the process of the controlled ovarian hyperstimulation (COH), serum GDF-8 levels are higher on the day of gonadotropin administration and 14 days after embryo transfer in in vitro fertilization (IVF) patients with PCOS than they are in IVF patients without PCOS. Importantly, GDF-8 levels in follicular fluid at oocyte retrieval are also higher in PCOS patients than in non-PCOS patients. Treatment of primary human granulosa-lutein (hGL) cells with GDF-8 downregulates steroidogenic acute regulatory protein (StAR) expression and the inhibition is more pronounced in hGL cells from PCOS patients than it is in cells from non-PCOS patients. Importantly, high GDF-8 levels and low progesterone (P4) levels were associated with poor pregnancy outcomes in PCOS patients. Our results provide the first evidence that aberrant expression of GDF-8 in the follicular fluid of PCOS patients results in abnormal P4 expression, which leads to poor pregnancy outcomes.BACKGROUND Papillary thyroid cancer (PTC) patients with clinical negative central lymph nodes (cN0), the use of prophylactic central lymph node dissection remains controversial. Patients with cN0 disease receive contralateral central lymph node metastasis (CCLNM) 3.88-30.63% of cases. Therefore, the present study aimed to obtain evidence for CCLNM risk factors in unilateral cN0 PTC. MATERIALS AND METHODS Relevant studies were searched in the PubMed, SCIE, and Wanfang databases up to 31 Oct 2019. The included patients had undergone lobectomy or total thyroidectomy with bilateral central lymph node dissection and were diagnosed pathologically with PTC. RESULTS Thirteen studies comprising 2449 patients were included. The factors associated with increased CCLNM risk in cN0 unilateral PTC were age less then 45 years (odds ratio (OR) = 1.89, 95% confidence interval (CI) = 1.43-2.49, P less then 0.00001), male sex (OR = 1.67, 95% CI = 1.24-2.24, P = 0.0007), extrathyroidal extension (OR = 1.63; 95% CI = 1.17-2.28; P = 0.004), tumor size ≥ 1 cm (OR = 2.63, 95% CI 1.85-3.74, P less then 0.00001), lymphovascular invasion (OR = 4.27, 95% CI =2.47-7.37, P less then 0.00001) and ipsilateral CLNM (OR = 11.42, 95% CI = 5.25-24.86, P less then 0.00001). However, no association was found for capsular invasion, multifocality and Hashimoto thyroiditis. CONCLUSION The study identified that age less then 45 years, tumor ≥1 cm, male sex, lymphovascular invasion, extrathyroidal extension and ipsilateral CLNM are related to CCLNM in CN0 PTC. These factors should influence the use of prophylactic central lymph node dissection in these patients.BACKGROUND In neuromyelitis optica spectrum disorder (NMOSD), clinical disability in NMOSD patients is relapse-related and progressive phase is rare. This observation raises the question whether there is any radiographic disease activity. selleck inhibitor The aim of present study was to determine the longitudinal changes in cerebral lesion number, lesion size, lesion-to-venule relationship, and morphological patterns of lesions in NMOSD using multiparametric 7T MR imaging. We also aimed to assess brain volume changes in NMOSD. METHODS A cohort of 22 patients with NMOSD underwent high-resolution 3D-susceptibility weighted imaging (SWI) and 2D-gradient-echo (GRE-T2*) weighted imaging on 7T MRI of brain at baseline and after ~2.8 years of follow-up. Morphologic imaging characteristics, and signal intensity patterns of lesions were recorded at both time points. Lesions were classified as "iron-laden" if they demonstrated hypointense signal on GRE-T2* images and/or SWI as well as hyperintense signal on quantitative susceptibility od. PURPOSE Adolescents with pediatric onset multiple sclerosis (MS) engage in low rates of physical activity. Behavioral interventions that target physical activity are most successful if grounded in theory. This study explored social cognitive theory (SCT) variables as correlates of physical activity among adolescents with pediatric onset MS. PROCEDURES Participants who had a diagnosis of pediatric onset MS, were between the ages of 14 and 18 years, and were fluent and literate in English were eligible. The electronic web-delivered survey included a demographics questionnaire, the Godin Leisure Time Exercise Questionnaire (GLTEQ), and measures of SCT variables. RESULTS The overall sample included 24 adolescents with pediatric onset MS (M3,F21) with a mean age of 16.1 years and disease duration of 2.6 years. The sample was predominately Caucasian (n = 14, 58%). Exercise planning (ρ=0.54), exercise outcome expectations (ρ=0.46), self-efficacy for overcoming barriers (ρ = 0.52), and exercise social support (ρ=0.58) were associated with GLTEQ scores. CONCLUSIONS Researchers might develop behavioral interventions that target exercise outcome expectations, exercise planning, self-efficacy for overcoming barriers, and exercise social support for changing physical activity in pediatric onset MS. V.Explaining biodiversity in nature is a fundamental problem in ecology. The fact that competitive exclusion is rarely observed in natural ecosystems has not been fully understood. Here we show that, by forming chasing pairs and chasing triplets among the consumers and resources in the consumption process, the Competitive Exclusion Principle can be naturally violated. The modeling framework developed here is broadly applicable and can be used to explain the biodiversity of many consumer-resource ecosystems and hence deepens our understanding of biodiversity in nature.
