Using shotgun proteomics, the mRNA binding protein Cold Shock Domain Containing E1 (CSDE1), also called Upstream-of-N Ras (UNR), was detected in pancreatic cancer cell lines but not in normal pancreatic epithelial cells. The expression of CSDE1 in pancreatic cancer cells was confirmed by Western-blotting and immunohistochemistry of human pancreatic tumors. In vitro functional assays showed that siRNA downregulation of CSDE1 or gene knockout using CRISPR-Cas9 significantly reduced the invasiveness of pancreatic cancer cells. Together, this study reveals that CSDE1 is overexpressed in pancreatic cancer and is a potential therapeutic target to inhibit pancreatic cancer cell invasion. This article is protected by copyright. Early detection has been described as the best approach to manage frailty; however, frailty instruments are not widely available, particularly in the Thai language. The purpose of this cross-sectional study was to develop a culturally-adapted Thai version of the Reported Edmonton Frail Scale and to validate the psychometric properties of the new instrument in hospitalized older Thai adults. Reliability and validity were examined. Participants completed questionnaires that included demographic and health information, and the Reported Edmonton Frail Scale-Thai version. Results revealed that the new instrument was reliable and had good content validity. Inter-rater reliability was strong. Confirmatory factor analysis showed a fair fit for the whole model, but most domains were strongly associated with frailty. On average, the instrument was completed in under 7 minutes. The Thai version of the frailty instrument may be a practical tool for frailty evaluation, and could inform inpatient care, both locally and internationally; future research is needed to confirm predictability and feasibility in other clinical settings and populations. The stability of this peculiar product has been ascribed largely to strong antiferromagnetic interaction of the two spins. Further oxidation of this diradical produced the corrole tape, suggesting its involvement as a reaction intermediate to the corrole tape. As another example, oxidation of 10-10'-linked bis-pyridine-coordinated CoIII corrole dimer with DDQ produced a cobalt corrole radical dimer and a doubly-linked corrole dimer both as stable compounds bearing pyridine and cyanide axial ligands. Bis-pyridine-coordinated CoIII corrole monomer was also converted to the corresponding corrole radical upon treatment with cyanide anion. Further oxidation of the cobalt corrole radical dimer with p-chloranil gave the doubly-linked cobalt corrole dimer exclusively. KGaA, Weinheim.Histiocytic sarcoma (HS) is a comparatively rare hematolymphoid neoplasm. Differential diagnosis of this entity has generally been proven to be so difficult that extensive immunohistochemical staining is required to confirm its presence. We herein present a case of HS manifesting as a retroperitoneal mass in which imprint cytology (IC) contributed to the diagnosis. A 77-year-old man was referred for the evaluation of a colonic tubulovillous polypoid lesion. Preoperative imaging revealed a 3 cm-sized polypoid lesion in the transverse colon. Incidentally, a retroperitoneal mass of 8 cm in the left para-aortic region was also detected. Diff-Quik stained IC slides were prepared as an adjunct to intraoperative pathology consultation. The smear exhibited moderate cellularity involving loosely cohesive cell clusters with intervening delicate vasculature. Most of the tumor cells consisted of large single cells with abundant eosinophilic cytoplasm that was vacuolated and foamy. The nuclei were predominantly large and eccentrically located. Mitotic figures were minimal in number and finely granular brown pigments were occasionally observed. mTOR inhibitor A frozen section diagnosis was reported as a malignant neoplasm, and a histologic diagnosis of HS was confirmed by conducting a battery of immunohistochemical stains negative for pan-cytokeratin, EMA, CD117, HMB45, Melan A, CD30, PAX5, CD20, S-100, and CD23 but strong positive for vimentin and CD68. This entity is rarely described in terms of cytopathologic features. This case study highlights how cytopathologists should be aware of HS as a retroperitoneal lesion and should become familiar with cytomorphologic features. © 2020 Wiley Periodicals, Inc.SCOPE Diabetes is thought to primarily represent a disturbance of carbohydrate metabolism; however, population studies employing metabolomics have mainly identified plasma amino acids and lipids, or their products, as biomarkers. In this pilot study, we aimed to analyze a wide spectrum of sugar compounds in fasting state and during an oral glucose tolerance test (OGTT) in healthy, prediabetic and type 2 diabetic volunteers. METHODS AND RESULTS The three volunteer groups underwent a standard OGTT. Plasma samples obtained in fasting state and 30 and 90 min after the OGTT were subjected to semitargeted GC-MS sugar profiling. Overall, 40 sugar compounds were detected in human plasma, of which some were so far unknown to change during an OGTT. Several sugar compounds (i.a. trehalose) revealed significant differences between the volunteer groups both in fasting plasma and in distinct time courses after the OGTT. Thus, suggesting an endogenous production from orally absorbed glucose and/or an insulin-dependent production/removal from plasma.
