Ventilation of the middle ear and mastoid air cells is believed to play an important role in the pathogenesis of chronic ear disease. Traditionally, ventilation is assessed by computed tomography. However, this exposes patients to cumulative radiation injury. In cases with a perforation in the tympanic membrane, tympanometry potentially presents a non-invasive alternative to measure the ventilated middle-ear and mastoid air cell volume. This study hypothesised that total tympanometry volume correlates with ventilated middle-ear and mastoid air cell volume.
Total tympanometry volume was compared with ventilated middle-ear and mastoid air cell volume on computed tomography scans in 20 tympanic membrane perforations.
There was a high correlation between tympanometry and computed tomography volumes (r = 0.78; p < 0.001). A tympanometry volume more than 2 ml predicted good ventilation on computed tomography.
These results may help reduce the need for pre-operative computed tomography in uncomplicated cases with tympanic membrane perforations.
These results may help reduce the need for pre-operative computed tomography in uncomplicated cases with tympanic membrane perforations.Despite several extensive and exhaustive efforts, search for potential therapy against leishmaniasis has not made much progress. In the present work, we have employed mining strategy to screen Leishmania donovani proteome for identification of promising vaccine candidate. We have screened 21 potential antigenic proteins from 7960 total protein of L. donovani, based on the presence of signal peptide, GPI anchor, antigenicity prediction and substractive proteomic approach. Selleckchem Ipatasertib Secondly, we have also performed comprehensive immunogenic epitope prediction from the screened 21 proteins, using IEDB-AR tools. Out of the 21 antigenic proteins, we obtained 11 immunogenic epitopes from 9 proteins. The final results revealed that four predicted epitopes namely; YPAFAALVF, VAVAATVAY, AAAPTEAAL and MYPLVAVVF, have significantly better binding potential with respective alleles and could elicits immune responses. Docking analysis using PATCHDOCK server and molecular dynamic simulation using GROMACS revealed the potential of the sequences as immunogenic epitopes. In silico studies also suggested that the epitopes occupied almost same binding cleft with the respective alleles, when compared with the reference peptides. It is also suggested from the molecular dynamic simulation data that the peptides were intact in the pocket for longer periods of time. Sarma.Galectin-1 (Gal-1) is the first member of galectin family, which has a carbohydrate recognition domain, specifically binds towards β-galactoside containing oligosaccharides. Owing its association with carbohydrates, Gal-1 is involved in many biological processes such as cell signaling, adhesion and pathological pathways such as metastasis, apoptosis and increased tumour cell survival. The development of β-galactoside based inhibitors would help to control the Gal-1 expression. In the current study, we carried out molecular dynamics (MD) simulations to examine the structural and dynamic behaviour Gal-1-thiodigalactoside (TDG), Gal-1-lactobionic acid (LBA) and Gal-1-beta-(1→6)-galactobiose (G16G) complexes. The analysis of glycosidic torsional angles revealed that β-galactoside analogues TDG and LBA have a single binding mode (BM1) whereas G16G has two binding modes (BM1 and BM2) for interacting with Gal-1 protein. We have computed the binding free energies for the complexes Gal-1-TDG, Gal-1-LBA and Gal-1-G16G using MM/PBSA and are -6.45, -6.22 and -3.08 kcal/mol, respectively. This trend agrees well with experiments that the binding of Gal-1 with TDG is stronger than LBA. Further analysis revealed that the interactions due to direct and water-mediated hydrogen bonds play a significant role to the structural stability of the complexes. The result obtained from this study is useful to formulate a set of rules and derive pharmacophore-based features for designing inhibitors against galectin-1.Communicated by Ramaswamy H. Sarma.
Since 2014, many companies have followed the lead of Apple and Facebook and now offer financial support to female employees to access egg freezing. Australian companies may soon make similar offers. Employer-sponsored egg freezing (ESEF) has raised concerns and there is academic debate about whether ESEF promotes reproductive autonomy or reinforces the 'career vs. family' dichotomy. Despite the growing availability of ESEF and significant academic debate, little is known about how ESEF is perceived by the public. The aim of this study was to explore women's attitudes toward ESEF.
Women aged 18-60
years who resided in Victoria, Australia were invited to complete an online, cross-sectional survey investigating views toward egg freezing. Associations between participant demographics and their views about ESEF were assessed using multinominal logistic regression, adjusted for age and free text comments were analyzed using thematic analysis.
The survey was completed by 656 women, median age 28
years (rangemen ESEF is not viewed as theoretically wrong, but rather it may be acceptable under certain conditions; such as with protections around reproductive freedoms and assurances that ESEF is offered alongside other benefits that promote career building and family. We suggest that there may be a role for the State in ensuring that these conditions are met.Situation awareness is the main reason behind different patterns of unsafe behavior among workers and can play a mediating role in the relationship between predictive situational variables and human error. This questionnaire-based study carried out with Iranian workers investigated the direct and indirect effects of work pressure, mental workload, human-systems interaction and environmental distractions on three types of human error (i.e., slips, lapses and mistakes). The results of path analysis showed that, based on model fit indexes, the model is appropriately fit (χ2 / df = 3, comparative fit index = 099, Tucker-Lewis index = 0.99, root mean square error of approximation = 026). It was also observed that the effects of four independent variables on all three types of human error are indirect and mediated by situation awareness. Consequently, it is confirmed that situation awareness plays a key role in the occurrence of human error and is a sharp-end causal factor for human error in industrial workplaces.
Gene drive technologies (GDTs) bias the inheritance of a genetic element within a population of non-human organisms, promoting its progressive spread across this population. If successful, GDTs may be used to counter intractable problems such as vector-borne diseases. A key issue in the debate on GDTs relates to what governance is appropriate for these technologies. While governance mechanisms for GDTs are to a significant extent proposed and shaped by professional experts, the perspectives of these experts have not been explored in depth.
A total of 33 GDT experts from different professional disciplines were interviewed to identify, better understand, and juxtapose their perspectives on GDT governance. link3 The pseudonymized transcripts were analyzed thematically.
Three main themes were identified (1) engagement of communities, stakeholders, and publics; (2) power dynamics, and (3) decision-making. There was broad consensus amongst respondents that it is important to engage communities, stakeholders, and puns.
The insights obtained in this interview study give rise to recommendations for the design and evaluation of GDT governance. Moreover, these insights point to unresolved normative questions that need to be addressed to move from general commitments to concrete obligations.The aims of this study were; 1) to identify different movement profiles in sixth graders, and 2) to investigate if there are differences in their mathematical basic (BasicMath) and problem solving (ProbSol) skills between existing movement profiles. The sample included 461 (223 girls, 238 boys) students with a mean age of 11.27 ± .32 years from southern and middle Finland. A latent profile analysis (LPA) revealed four movement profiles "poor movers", "average movers", "skilled movers" and "expert movers". These profiles differed substantially in their motor competence (MC) and health-related fitness (HRF). A multivariate analysis of variance (MANOVA) also revealed that "poor movers" and "average movers" obtained lower results in BasicMath comparing with "skilled movers". The results of this study suggest that cardiorespiratory and muscular fitness, MC and BasicMath are interrelated.Coping skills represent cognitive, emotional, and behavioral resources to overcome developmental challenges and tasks. Based on Cloninger's model of personality, the main aim of this study was to analyze the relation among temperament, character, and coping skills in nonclinical pre- and early adolescents by also considering adolescents' gender and age. One hundred and thirty-eight Italian pre- and early adolescents (52.2% boys and 47.8% girls), aged between 10-14 years (M = 12.33 ± 0.87 SD), filled out the Junior Temperament and Character Inventory and the Children's Coping Strategies Checklist Revision 1. Regression and relative weights analyses showed that several of adolescents' personality dimensions did predict their coping skills. Specifically, Harm Avoidance was negatively related to Positive Reframing and Distraction; this latter (i.e., Distraction) was also negatively associated with Novelty Seeking, Self-Directedness, and Cooperativeness. On the contrary, Cooperativeness was positively related to the use of Problem Focus strategy, and Reward Dependence was positively related to Distraction and Social Support. Significant gender and age differences in personality as well as in coping skills emerged. Girls scored higher on Harm Avoidance, Reward Dependence, Persistence and Problem Focus, and boys scored higher on Novelty Seeking and Distraction. Moreover, pre-adolescents (10-12 years) reported lower scores on Novelty Seeking and higher on Problem Focus than early adolescents (13-14 years). Overall, the results suggest that bio-psycho-social individual factors linked to personality, gender, and age have an important role in shaping pre- and early adolescents' coping and adaptation responses. Practical implications of the results and future developments are discussed.Titanium dioxide (TiO2) nanoparticles (NPs) are used extensively in a variety of commercial, industrial, and medical products, due to which human exposure is inevitable. This study aimed to explore the potential of eugenol and thymoquinone (TQ), two well-known antioxidants, in counteracting the NP-induced toxicity in human blood cells in vitro. Fresh lymphocytes and erythrocytes were isolated from volunteer human blood donors and incubated with 50 μg/mL of TiO2 NPs in the presence and absence of 50 μM of TQ and 20 μg/mL of eugenol for 3 h. Results showed that NP-treatment-induced hemolysis, oxidative stress, lactate dehydrogenase (LDH) leakage, and reduced ATPase activity in the erythrocytes. In the lymphocytes treated with NPs alone (50 μg/mL), cytotoxicity in MTT assay and DNA damage in comet assay were observed; in addition, mitochondrial membrane potential collapsed and ADP/ATP ratio increased indicating mitochondrial function impairment. However, in the presence of antioxidants, all these NP-induced changes were mitigated significantly.
