RATIONALE A potential use of disaccharides was found in this research, as reference compounds in the kinetic method for chiral recognition and enantiomeric quantification. METHODS The experimental procedure consists of three steps (1) mixing a metal salt, an analyte and a disaccharide at a molar ratio of 122 with an analyte concentration of 20 μM; (2) introducing the mixture into an Electrospray Ionization (ESI) MS source; (3) isolating the ion [MII (ref*)2 (A)-H]+ and subjecting it to dissociation. Then through the relative ratio of two product ions combined with the kinetic method, chiral recognition and enantiomeric quantification can be achieved. RESULTS The method was verified with good chiral recognition for ten chiral amino acids and three chiral drugs. Among the ten amino acids, Tyr was observed to show best chiral selectivity (Rchiral =1.62) and good linearity with the correlation coefficient R2 =0.9991 for the quantitation of the enantiomeric excess (ee) of D-Tyr. Among the tested chiral drugs, naproxen showed best chiral selectivity with Rchiral =1.56 and good linearity with the correlation coefficient R2 =0.9997 for the quantitation of the enantiomeric excess (ee) of R-naproxen. CONCLUSIONS This established approach presents that a disaccharide can serve as a reference compound for the chiral recognition using the kinetic method. This article is protected by copyright. Vibrio bacteria are concentrated by oysters and, when consumed, infections can result with severe symptoms such as diarrhoea, lesions on the extremities, or even death. Vibrio spp. concentrations are strongly affected by season, location, and other factors such as temperature and salinity. Previous research in North Carolina oysters has been conducted on wild and farmed oysters but not at the same time. Farmed, or aquaculture raised, oysters are considerably different from wild oysters and could possibly pose different health risks. Farmed oysters are handled, raised from seed, and often grown using suspended grow-out systems called 'floating cages'. Therefore, farmed oysters can be grown at the surface of the estuary, while wild oysters typically grow at the bottom of the water column. This project compared the concentrations of Vibrio spp. in suspended, farm-grown oysters and wild oysters at three sites, using a paired approach with farmed and wild oysters sampled in proximity. An important part of this comparison was identifying pathogenicity of the bacteria isolated from the samples. Distinction was made between off- and on-bottom farming. However, little is known of microbial interactions with struvite which would result in element release. In this work, the geoactive fungus Aspergillus niger was investigated for struvite transformation on solid and in liquid media. Aspergillus niger was capable of solubilizing natural (fragments and powder) and synthetic struvite when incorporated into solid medium, with accompanying acidification of the media, and extensive precipitation of magnesium oxalate dihydrate (glushinskite, Mg(C2 O4 ).2H2 O) occurring under growing colonies. In liquid media, A. niger was able to solubilize natural and synthetic struvite releasing mobile phosphate (PO4 3- ) and magnesium (Mg2+ ), the latter reacting with excreted oxalate resulting in precipitation of magnesium oxalate dihydrate which also accumulated within the mycelial pellets. Struvite was also found to influence the morphology of A. niger mycelial pellets. These findings contribute further understanding of struvite solubilization, element release and secondary oxalate formation, relevant to the biogeochemical cycling of phosphate minerals, and further directions utilizing these mechanisms in environmental biotechnologies such as element biorecovery and biofertilizer applications. © 2020 The Authors.
