This study, for the first time, aims to evaluate the situation of air quality in Pakistan critically; through a detailed assessment of sources, policies, and key challenges to identify the plausible way forward. Air pollution and particulate matter have merged as a global challenge in recent years because of its growing health and socio-economic risks. The intensity and impacts of these risks have become more pronounced, especially in developing countries like Pakistan that lack adequate warning, protection, and management systems. Various epidemiological studies have linked poor air quality with different health disorders and increasing death rates. In Pakistan, mortality rates as a result of exposure to increased levels of air pollutants, especially particulate matter, are alarming. According to the World Bank's estimates, Pakistan's annual burden of disease from outdoor air pollution is responsible for around 22,000 premature adult deaths and 163,432 DALYs (disability-adjusted life years) lost. Herein, we propose a fluorescent immunoassay for the highly sensitive and rapid analysis of chloramphenicol (CAP) based on carbon dots (CDs)-encapsulated CaCO3 nanospheres and magnetic nanoparticles (MNPs). The fluorescent immunoprobes were prepared by coupling the anti-CAP antibodies to carboxymethyl cellulose-functional CDs@CaCO3 nanospheres. Chitosan-modified MNPs with "core-shell" structures were prepared and then conjugated to the CAP hapten, acting as the nano-carrier and interface for the immunoreaction. With the assistance of MNPs, the established fluorescent immunoassay achieved the sensitive detection of CAP in chicken with a limit of detection of 0.03 μg kg-1 and recoveries ranging from 83.7%-105.0%. The analysis results of the fluorescent immunoassay were evaluated by the enzyme-linked immunosorbent assay, having a correlation coefficient of 0.981. Our work provides a rapid, facile, and reliable strategy for the highly sensitive analysis of food contaminants based on "green" fluorescent nanoprobes.A NiCo2O4-Au composite was fabricated following the calcination-reduction method and testified to exhibit the intrinsic high peroxidase- and oxidase-like activities. The composite could activate a low level of H2O2 (100 μM) to kill E. coli and S. selleckchem aureus. NiCo2O4-Au composite could be easily separated by an external magnet from the media and reused several times. NiCo2O4-Au composite could also effectively damage the existing biofilms and prevent the formation of new biofilms. The radical trapping experiment confirmed that OH and 1O2 were the main radicals in the antibacterial process for NiCo2O4-Au in the presence of H2O2. A NiCo2O4-Au based Band-Aid was also designed, which exhibited high anti-infective and wound-healing properties. This study has demonstrated that NiCo2O4-Au composite can be a promising antibacterial agent in environmental and clinical applications.Microplastics, which are new types of environmental pollutants, are recently receiving widespread attention worldwide. Hydrogen sulfide (H2S) as the third endogenous gaseous mediator had protective effects in multiple physiological and pathological conditions. However, the protective role of H2S in microplastics-induced hepatotoxocity remain unclear. In this study, our data showed that H2S significantly suppressed inflammation, apoptosis and oxidative stress induced by polystyrene microplastics (mic-PS) (20 mg/kg b.w.) in the liver. Strikingly, although mic-PS exposure increased the expression of nuclear factor-E2-related factor (Nrf2), it did not influence the levels of heme oxygenase-1 (HO-1) and NAD(P)H quinone oxidoreductase 1 (NQOl) in the L02 hepatocytes. Immunofluorescence assay showed that sodium hydrosulfide (NaHS) reduced micro-Ps-induced hepatic apoptosis by facilitating nuclear accumulation of Nrf2. Simultaneously, flow cytometry also showed that NaHS could prevent mic-PS-induced accumulation of reactive oxygen species (ROS) by increasing the expression of HO-1 and NQO1. Furthermore, inhibition of HO-1 could reverse the hepatic protective effects of NaHS during mic-PS exposure. Mechanistically, H2S elevating the HO-1 and NQO1 expression by facilitating nuclear accumulation of Nrf2, and consequently reducing mic-PS-induced hepatic apoptosis and inflammation. This study unveils the hepatotoxic effects of MPs and suggest NaHS have protective effects on mic-PS-induced liver damage.Bisphenol A (BPA), an environmental endocrine-disrupting compound, has been revealed associated with metabolic disorders such as obesity, prediabetes, and type 2 diabetes (T2D). However, its underlying mechanisms are still not fully understood. Here, we provide new evidence that BPA is a risk factor for T2D from a case-control study. To explore the detailed mechanisms, we used two types of diet models, standard diet (SD) and high-fat diet (HFD), to study the effects of long-term BPA exposure on prediabetes in 4-week-old mice. We found that BPA exposure for 12 weeks exacerbated HFD-induced prediabetic symptoms. Female mice showed increased body mass, serum insulin level, and impaired glucose tolerance, while male mice only exhibited impaired glucose tolerance. No change was found in SD-fed mice. Besides, BPA exposure enhanced astrocyte-dependent hypothalamic inflammation in both male and female mice, which impaired proopiomelanocortin (POMC) neuron functions. Moreover, eliminating inflammation by toll-like receptor 4 (TLR4) knockout significantly abolished the effects of BPA on the hypothalamus and diet-induced prediabetes.
