8 vs ≤median) were inversely associated with all-cause mortality (HR = 0.74, 95% CI = 0.56 to 0.98) among women with breast cancer. Reduced hazard was also observed for breast cancer mortality (HR = 0.73, 95% CI = 0.45 to 1.17) and cardiovascular diseases mortality (HR = 0.76, 95% CI = 0.47 to 1.23), although the 95% confidence intervals included the null. Similar findings were also observed for women without breast cancer. The association with all-cause mortality was more pronounced among breast cancer participants who began chemotherapy before urine collection (n = 118, HR = 0.42, 95% CI = 0.22 to 0.81) than among those who had not (n = 559, HR = 0.98, 95% CI = 0.72 to 1.34; P interaction = .008). Conclusions The urinary 2-OHE1 to 16-OHE1 ratio may be inversely associated with long-term all-cause mortality, which may depend on cancer treatment status at the time of urine collection.Background Chronically higher inflammation, likely contributed to by dietary and lifestyle exposures, may increase risk for colorectal cancer (CRC). To address this, we investigated associations of novel dietary (DIS) and lifestyle (LIS) inflammation scores with incident CRC in the prospective National Institutes of Health-American Association of Retired Persons Diet and Health Study (N = 453 465). Methods The components of our previously developed and externally validated 19-component DIS and 4-component LIS were weighted based on their strengths of associations with a panel of circulating inflammation biomarker concentrations in a diverse subset (N = 639) of participants in the REasons for Geographic and Racial Differences in Stroke Study cohort. We calculated the components and applied their weights in the National Institutes of Health-American Association of Retired Persons cohort at baseline, summed the weighted components (higher scores reflect a higher balance of proinflammatory exposures), and investigated associations of the scores with incident CRC using Cox proportional hazards regression. All statistical tests were two-sided. Results Over a mean 13.5 years of follow-up, 10 336 participants were diagnosed with CRC. Among those in the highest relative to the lowest DIS and LIS quintiles, the multivariable-adjusted hazards ratios (HRs) and their 95% confidence intervals (CIs) were HR = 1.27 (95% CI = 1.19 to 1.35; P trend less then .001) and 1.38 (95% CI = 1.30 to 1.48; P trend less then .001), respectively. The associations were stronger among men and for colon cancers. The hazards ratio for those in the highest relative to the lowest joint DIS and LIS quintile was HR = 1.83 (95% CI = 1.68 to 1.99; P interaction less then .001). TAK165 Conclusions Aggregates of proinflammatory dietary and lifestyle exposures may be associated with higher risk for CRC.Metastasis, the leading cause of death in cancer patients, requires the invasion of tumor cells through the stroma in response to migratory cues, in part provided by the extracellular matrix (ECM). Recent advances in proteomics have led to the identification of hundreds of ECM proteins, which are more abundant in tumors relative to healthy tissue. Our goal was to develop a pipeline to easily predict which ECM proteins are more likely to have an effect on cancer invasion and metastasis. We evaluated the effect of four ECM proteins upregulated in breast tumor tissue in multiple human breast cancer cell lines in three assays. There was no linear relationship between cell adhesion to ECM proteins and ECM-driven 2D cell migration speed, persistence, or 3D invasion. We then used classifiers and partial-least squares regression analysis to identify which metrics best predicted ECM-driven 2D migration and 3D invasion responses. We find that ECM-driven 2D cell migration speed or persistence did not predict 3D invasion in response to the same cue. However, cell adhesion, and in particular cell elongation and shape irregularity, accurately predicted the magnitude of ECM-driven 2D migration and 3D invasion. Our models successfully predicted the effect of novel ECM proteins in a cell-line specific manner. Overall, our studies identify the cell morphological features that determine 3D invasion responses to individual ECM proteins. This platform will help provide insight into the functional role of ECM proteins abundant in tumor tissue and help prioritize strategies for targeting tumor-ECM interactions to treat metastasis.[This corrects the article DOI 10.1021/acsomega.9b04231.].Various types of seaweed are potential functional foods as they contain multiple bioactive compounds. N-Methyltaurine (NMT) is a taurine derivative metabolite found in a type of red algae. The functional actions of NMT in mammalian animals have not been investigated, but the parent compound, taurine, possesses a variety of cellular actions. To explore the beneficial role of NMT in animals, the present study analyzed the effect of NMT against glucocorticoid-induced skeletal muscle atrophy. Glucocorticoids are one of the major causes of pathological muscle atrophy. Initially, we assessed the bioavailability of ingested NMT by determining its concentration in mouse blood. The bioavailability of orally administered NMT was found to be 96.1% that of intravenously administered NMT. Mice maintained on water containing 0.5% NMT for several days lead to the distribution of the taurine derivative to various tissues, including skeletal muscles. Like taurine, the delivery of NMT to skeletal muscles or myoblast cells is cytoprotective. The treatment with NMT prevents dexamethasone-induced atrophy of myotubes derived from C2C12 cells. Similarly, the addition of 0.5% NMT to drinking water attenuates dexamethasone-mediated reduction in muscle mass of the treated mice. The present study supports the hypothesis that orally administered NMT partially reverses skeletal muscle atrophy.The ring stain phenomenon is a critical hindrance to the distribution of the solute during drying for biochemical assays and materials deposition. Herein, we developed a substrate, characterized with hydrophilic spots surrounded by hydrophobic areas, to suppress the ring stain effect, and fabricated four kinds of patterned surfaces to investigate the relationship between the surface free energy and ring-suppressing performance. We found that during the evaporation process, a drop was constrained on the hydrophilic spot with a pinned contact line, and the ring stain effect was suppressed significantly. The suppressing performance of the ring stain effect increases with surface free energy differences between the hydrophilic and hydrophobic regions.
