Despite increasing evidence that cannabinoids elicit antitumor effects in adult cancers, there is minimal data available on their effects in children or in pediatric cancers despite public and clinical demand for information. Here we describe a comprehensive and critical review of what is known about the effects of cannabinoids on pediatric cancers, highlight current gaps in knowledge and identify the critical issues that need addressing before considering these promising but controversial drugs for use in pediatric oncology.The maternal-to-zygotic transition (MZT), which controls maternal signaling to synthesize zygotic gene products, promotes the preimplantation development of mouse zygotes to the two-cell stage. Our previous study reported that mouse granzyme g (Gzmg), a serine-type protease, is required for the MZT. In this study, we further identified the maternal factors that regulate the Gzmg promoter activity in the zygote to the two-cell stage of mouse embryos. A full-length Gzmg promoter from mouse genomic DNA, FL-pGzmg (-1696~+28 nt), was cloned, and four deletion constructs of this Gzmg promoter, Δ1-pGzmg (-1369~+28 nt), Δ2-pGzmg (-939~+28 nt), Δ3-pGzmg (-711~+28 nt) and Δ4-pGzmg (-417~+28 nt), were subsequently generated. Different-sized Gzmg promoters were used to perform promoter assays of mouse zygotes and two-cell stage embryos. The results showed that Δ4-pGzmg promoted the highest expression level of the enhanced green fluorescent protein (EGFP) reporter in the zygotes and two-cell embryos. The data suggested thscription activity in preimplantation mouse embryos. It plays an important role in the maternal-to-zygotic transition during early embryonic development.Resistance to chemotherapeutic agents is a major obstacle in cancer treatment. A recently proposed strategy is to target the collateral sensitivity of multidrug resistant (MDR) cancer. Paradoxically, the toxicity of certain metal chelating agents is increased, rather than decreased, by the function of P-glycoprotein (Pgp), which is known to confer resistance by effluxing chemotherapeutic compounds from cancer cells. We have recently characterized and compared the solution's chemical properties including ligand protonation and the metal binding properties of a set of structurally related 8-hydroxyquinoline derived Mannich bases. Here we characterize the impact of the solution stability and redox activity of their iron(III) and copper(II) complexes on MDR-selective toxicity. Our results show that the MDR-selective anticancer activity of the studied 8-hydroxyquinoline derived Mannich bases is associated with the iron deprivation of MDR cells and the preferential formation of redox-active copper(II) complexes, which undergo intracellular redox-cycling to induce oxidative stress.The study aimed to evaluate the effect of replacing cottonseed meal by dried distiller's grains (DDG) in terms of efficiency in the productive aspects of beef cattle finishing in pasture versus feedlot. The experiment was conducted in a completely randomized design in a 2 × 3 factorial arrangement, with two production systems (pasture versus feedlot) and three supplements CM, conventional supplement with cottonseed meal (CM) as a protein source; 50DDG supplement with 50% replacement of CM by DDG; and 100DDG 100% replacement. The effect of replacing CM by DDG on dry matter and nutrients intake and nutrients digestibility depends on the finishing system (p less then 0.05). While in the pasture system animal consumed more nutrients in the CM, a greater intake was observed in the 100DDG in feedlot. The nutrients digestibility was lower in the pasture (p less then 0.05). Animal performance and final body weight were higher in the feedlot (p less then 0.0001), with averages of 1.57 kg/d and 566 kg of final body weight (FBW) for feedlot, and 0.99 kg/d and 504 kg FBW for pasture. The use of DDG does not change the animal performance finished in pasture or feedlot, and it is a viable alternative to replace conventional supplements in finishing phase in both systems in tropical environment.The role of autoimmunity in central nervous system (CNS) disorders is rapidly expanding. Several of these antibodies are directed against NMDA receptors (NMDAR), mostly in autoimmune encephalitis, whereas a growing field of research has identified antibodies against AMPA receptor (AMPAR) subunits in patients with different types of epilepsy or frontotemporal dementia. Several in vitro and in vivo studies performed in the last decade have dramatically improved our understanding of the molecular and functional effects induced by both NMDAR and AMPAR autoantibodies at the excitatory glutamatergic synapse and, consequently, their possible role in the onset of clinical symptoms. In particular, the method by which autoantibodies can modulate the localization at synapses of specific target subunits leading to functional impairments and behavioral alterations has been well addressed in animal studies.
