We aimed to evaluate the analytical performance of the Daan HCV RNA quantitative assay and compare it with the COBAS AmpliPrep/COBAS TaqMan HCV Quantitative Test, v2.0. METHOD WHO HCV RNA standard, NIBSC 06/102 standard, and CLSI EP documents were used to evaluate the precision, accuracy, linearity, anti-interference ability, and cross-reactivity of the Daan HCV RNA quantitative assay. Overall 198 clinical serum specimens were used to make comparison between the Daan HCV RNA quantitative assay and the Roche Cobas test. RESULTS The within-run precision (Swithin ), and total precision (Stotal ) for 6.11 log IU/mL, 4.22 log IU/mL, and 2.32 log IU/mL HCV RNA were 0.13 and 0.15, 0.07 and 0.09, and 0.11 and 0.10, respectively. The linear range was 20-108 IU/mL, and the limit of detection was 15 IU/mL. 17β-Oestradiol It did not display any interference with commonly encountered conditions and cross-reactivity with some common virus. A good agreement was observed between the Daan HCV RNA quantitative assay and the Roche Cobas test. CONCLUSION The Daan HCV RNA quantitative assay has shown satisfactory performances and excellent agreement with COBAS HCV Quantitative Test on clinical specimens with lower cost, which provides an alternative choice for the diagnosis and monitoring of HCV infection in developing countries. © 2020 The Authors. We initially did not know each other; however, we were drawn by a mutual desire to further the conversation and sought connection with each other after the Conversations Around Diversity Platform Presentations. Through sustained, open dialogue we created a brave space for sharing our emotional and intellectual responses to the conference. Through this dialogue and through written reflections, we recognized an emboldened urgency to author a joint reflection on our shared responsibility as genetic counseling training program leaders to use our privilege in service to our students and future students. We have the evidence that we are not a diverse profession. We have more evidence now than we did before that our profession performs poorly with regards to inclusivity. Our inability to acknowledge, address, and discuss racism and other forms of oppression is damaging to each of us individually and as a group of professionals. We owe it to ourselves, our students, our patients, and colleagues to name our learned biases and behaviors, own them and interrupt them. © 2020 National Society of Genetic Counselors.Conservation treatment of degraded archaeological osseous materials is still an open challenge, since no specific conservation protocol is currently available for restorers or museum curators. This work aims to test the efficiency of two original consolidant solutions in consolidating archaeological material. Archaeological osseous materials remain rare and sparsely available, it is a real drawback for optimization of conservation treatments, therefore in the present work a set of representative samples was chosen. The consolidants tested were a solution of disodium sebacate and a novel polyalcohol (SG1.2) obtained by esterification of 5 succinic diacids with 6 molecules of glycerol at 150°C. Characterization studies of archaeological bones, combining SEM microscopy, IR spectroscopy and high-resolution solid-state 13 C NMR investigations, have been carried out to assess the effective permeation of bone by the consolidant solutions and to determine their chemical interactions with the residual components of archaeological bones. Although both water solutions significantly impregnate bone, we show that, the solution with disodium sebacate leads to chemical attack on the mineral component due to preferential precipitation of endogenous calcium by the sebacate ions. Such deleterious behaviour is not observed at all with the SG1,2 chemicals. The added value of the polyalcohol treatment as strengthening agent suitable for archaeological bony materials should be further demonstrated by mechanical and ageing tests. © 2020 John Wiley & Sons, Ltd.Human leukocyte antigen (HLA) class Ⅰ molecules play a central role in anticancer immunity, but their prognostic value in oral squamous cell carcinoma (OSCC) remains unclear. We examined HLA class Ⅰ expression in two distinct tumor compartments, namely, the tumor center and invasive front, and evaluated the association between its expression pattern and histopathological status in 137 cases with OSCC. HLA class Ⅰ expression was graded semi-quantitatively as high, low, and negative. At the invasive front of the tumor, HLA class I expression was high in 72 cases (52.6%), low in 44 cases (32.1%), and negative in 21 cases (15.3%). HLA class I expression in the tumor center was high in 48 cases (35.0%), low in 58 cases (42.4%), and negative in 31 cases (22.6%). The 5-year overall survival and disease-specific survival rates were good in cases with high HLA class I expression at the invasive front; however, there was no significant difference in survival based on HLA class I expression in the tumor center. In addition, high HLA class I expression was correlated with high CD8+ T cell density, whereas negative HLA class I expression was correlated with low CD8+ T cell density at the invasive front. These results suggest that it is easier for CD8+ T cells to recognize presented peptides in the case of high HLA class Ⅰ expression at the tumor invasive front and could be a prognostic factor for OSCC.
