Guided by the top ε4-attenuating signals from methylome-wide association analyses (N = 572, ε4+ and ε4-) of neurofibrillary tangles and neuritic plaques, we conducted a meta-analysis for pathological AD within the ε4+ subgroups (N = 235) across four independent collections of brains. Cortical RNA-seq and microglial morphology measurements were used in functional analyses. Three out of the four significant CpG dinucleotides were captured by one principal component (PC1), which interacts with ε4 on AD, and is associated with expression of innate immune genes and activated microglia. In ε4 carriers, reduction in each unit of PC1 attenuated the odds of AD by 58% (odds ratio = 2.39, 95% confidence interval = [1.64,3.46], P = 7.08 × 10-6 ). Because of the tremendous increase in collected data in breeding programs and the slow rate of genetic gain increase, it is required to explore the potential of artificial intelligence in analyzing the data. The main objectives of this study include optimization of multitrait (MT) machine- and deep-learning models for predicting grain yield and grain protein content in wheat (Triticum aestivum L.) using spectral information. This study compares the performance of four machine- and deep-learning-based unitrait (UT) and MT models with traditional genomic best linear unbiased predictor (GBLUP) and Bayesian models. The dataset consisted of 650 recombinant inbred lines (RILs) from a spring wheat breeding program grown for three years (2014-2016), and spectral data were collected at heading and grain filling stages. The MT-GS models performed 0-28.5 and -0.04 to 15% superior to the UT-GS models. Random forest and multilayer perceptron were the best performing machine- and deep-learning models to predict both traits. Four explored Bayesian models gave similar accuracies, which were less than machine- and deep-learning-based models and required increased computational time. Green normalized difference vegetation index (GNDVI) best predicted grain protein content in seven out of the nine MT-GS models. Overall, this study concluded that machine- and deep-learning-based MT-GS models increased prediction accuracy and should be employed in large-scale breeding programs.Sonic hedgehog (Shh)-driven medulloblastoma (Shh MB) cells are dependent on constitutive Shh signaling, but targeted treatment of Shh MB has been ineffective due to drug resistance. The purpose of this study was to address the critical role of signal transducer and activator of transcription 3 (STAT3) in Shh signaling and drug resistance in Shh MB cells. Herein, we show that STAT3 is required for Smoothened (Smo)-dependent Shh signaling and, in turn, is reciprocally regulated by Shh signaling, and demonstrate that STAT3 activity is critical for expression of HCK proto-oncogene, Src family tyrosine kinase (Hck) in Shh MB. We also demonstrate that maintained STAT3 activity suppresses p21 expression and promotes colony formation of Shh MB cells, whereas dual treatment with inhibitors of both Smo and STAT3 results in marked synergistic killing and overcomes drug resistance in vitro of Smo antagonist-resistant Shh MB cells. Finally, STAT3 inhibitor treatment significantly prevents in vivo tumor formation in genetically engineered Shh MB mice. Collectively, we show that STAT3 is necessary to maintain Shh signaling and thus is a potential therapeutic target to treat Shh MB and overcome anti-Smo drug resistance.

This integrative review examined research studies addressing physical activity in women with a history of preeclampsia or other hypertensive disorders of pregnancy. Using four databases, the search strategy included published studies through December 31, 2019. Identified studies were assessed using Joanna Briggs Institute critical appraisal tools.
The initial search identified 1,166 publications. Seven studies including two observational, four quasi-experimental, and one experimental study were included in the review. Physical activity promotion among women with a history of hypertensive disorders of pregnancy has been studied by a small group of researchers. Current literature on the topic is characterized by limited sample diversity, lack of exercise-based interventions, and over reliance on self-report to measure physical activity. It is imperative that further research be conducted to facilitate improved cardiovascular outcomes.The ability to differentiate between two languages sets the stage for bilingual learning. Infants can discriminate languages when hearing long passages, but language switches often occur on short time scales with few cues to language identity. As bilingual infants begin learning sequences of sounds and words, how do they detect the dynamics of two languages? In two studies using the head-turn preference procedure, we investigated whether infants (n = 44) can discriminate languages at the level of individual words. In Study 1, bilingual and monolingual 8- to 12-month-olds were tested on their detection of single-word language switching in lists of words (e.g., "dog… lait [fr. milk]"). In Study 2, they were tested on language switching within sentences (e.g., "Do you like the lait?"). We found that infants were unable to detect language switching in lists of words, but the results were inconclusive about infants' ability to detect language switching within sentences. No differences were observed between bilinguals and monolinguals. Given that bilingual proficiency eventually requires detection of sound sequences across two languages, more research will be needed to conclusively understand when and how this skill emerges. Efforts to model the cost-effectiveness of managing/modifying cognitive impairment lack reliable, objective, baseline medical, and nursing-home (NH) costs.
A stratified-random sample of Olmsted County, MN, residents ages 70-89 years (N=3545), well-characterized as cognitively unimpaired, mild cognitive impairment (MCI), or dementia, were followed forward ≤1 year in provider-linked billing data and the Centers for Medicare &-Medicaid Services NH assessments. Direct medical/nursing home/medical + NH costs were estimated. Costs were stratified by vital status and NH-use intensity (NH days/follow-up days [0%, 1% to 24%, 25% to 99%, and 100%]). Between-category mean-annual cost differences were adjusted for patient characteristics and follow-up days.
Costs/follow-up day distributions differed significantly across cognitive categories. Mean costs/follow-up days were 2.5 to 18 times higher for decedents versus survivors. Among all persons with MCI, <9% with any NH use accounted for 18% of all total annual medical + NH costs. Adjusted-between-category comparisons revealed significantly higher medical and medical + NH costs for MCI versus cognitively unimpaired.
Cost-effectiveness for managing/modifying both MCI and dementia should consider end-of-life costs and NH-use intensity. Results can help inform cost-effectiveness models, predict future-care needs, and aid decision-making by individuals/providers/payers/policymakers.

This consensus document by the Heart Failure Association (HFA) of the European Society of Cardiology (ESC) presents current evidence-based indications relevant to integration of imaging techniques and biomarkers in HF. The document first focuses on application of circulating biomarkers together with imaging findings, in the broad domains of screening, diagnosis, risk stratification, guidance of treatment and monitoring, and then discusses specific challenging settings. In each section we crystallize clinically relevant recommendations and identify directions for future research. The target readership of this document includes cardiologists, internal medicine specialists and other clinicians dealing with HF patients.The review investigated the barriers and facilitators associated with assessing and engaging with mental health in a rural setting. The aim is to describe and synthesize the literature that examines the experiences of adults who access or attempt to access mental health services in rural settings. A systematic search from 2010 to 2020 was conducted using CINAHL, PsycINFO, Web of Science Core Collection, PubMed, Psychology and Behavioural Sciences Collection, Google Scholar, and Scopus. PRISMA protocols located 32 relevant papers from the overall 573 first selected. Braun and Clarke (Qualitative Research in Psychology, 377-101, 2006) thematic analysis methodology was applied to the data resulting in two themes first theme identified help-seeking with subthemes of stigma and locality of health services. The second theme was connectedness, inclusive of subthemes of support systems and personal identity. The review identified gender-related perspectives concerning accessing mental health support, exposing the need for more research to examine the diverse social connections and support networks in rural communities. The findings suggest the need to further explore the impediments that reduce the likelihood of accessing mental health services in rural communities.
