Fungal infections and toxicoses caused by insecticides may alter microbial communities and immune responses in the insect gut. We investigated the effects of Metarhizium robertsii fungus and avermectins on the midgut physiology of Colorado potato beetle larvae. We analyzed changes in the bacterial community, immunity- and stress-related gene expression, reactive oxygen species (ROS) production, and detoxification enzyme activity in response to topical infection with the M. robertsii fungus, oral administration of avermectins, and a combination of the two treatments. Avermectin treatment led to a reduction in microbiota diversity and an enhancement in the abundance of enterobacteria, and these changes were followed by the downregulation of Stat and Hsp90, upregulation of transcription factors for the Toll and IMD pathways and activation of detoxification enzymes. Fungal infection also led to a decrease in microbiota diversity, although the changes in community structure were not significant, except for the enhancement of Serratia. Fungal infection decreased the production of ROS but did not affect the gene expression of the immune pathways. In the combined treatment, fungal infection inhibited the activation of detoxification enzymes and prevented the downregulation of the JAK-STAT pathway caused by avermectins. The results of this study suggest that fungal infection modulates physiological responses to avermectins and that fungal infection may increase avermectin toxicosis by blocking detoxification enzymes in the gut.In the long history of development and elimination, the creatures have derived a variety of exquisite structures and unique properties, typically natural nacre, marine mussel and Glycera to adapt to the environment and resist the predation of the enemy. We raised queen bees on four Hawaiian Islands (Kaua'i, O'ahu, Maui, and Hawai'i) and subsequently collected their offspring to determine queen mating frequency and insemination success. No significant difference for mating success was found between the islands with and without varroa mites, and relatively high levels of polyandry was detected overall. We also found a significant association between the number of sperm stored in the queens' spermathecae and the number of managed colonies within the localities of the queens mated. Our findings suggest that varroa mites, as they currently occur in Hawai'i, may not significantly reduce mating success of honey bee queens, which provides insight for both the reproductive biology of honey bees as well as the apiculture industry in Hawai'i.We aimed to compare the (1) clinical outcomes including composite cardiovascular outcomes, cardiovascular death, and all-cause death, and (2) healthcare costs of using liraglutide and basal insulin as an initial treatment for patients with type 2 diabetes mellitus (T2DM) and high cardiovascular diseases (CVD) risk. This is a retrospective cohort study using Taiwan's Health and Welfare Database. A total of 1057 patients treated with liraglutide were identified and matched with 4600 patients treated with basal insulin. The liraglutide group had a lower risk of a composite CVD outcome (hazard ratio (HR) 0.65; 95% confidence interval (CI) 0.50-0.85; p  less then  0.01), all-cause mortality (HR 0.40; 95% CI 0.28-0.59; p  less then  0.0001), and nonfatal stroke (HR 0.54; 95% CI 0.34-0.87; p = 0.01). Compared to the basal insulin group, the liraglutide group had lower median per-patient-per-month (PPPM) inpatient, emergency room (ER), and total medical costs, but higher median PPPM outpatient, total pharmacy, and total costs (all p  less then  0.0001). In conclusion, compared to basal insulin, liraglutide was found to be associated with reduced risk of a composite CVD outcome, nonfatal stroke, and all-cause mortality among high CVD risk patients with T2DM. In addition, liraglutide users had lower inpatient, ER, and total medical costs, but they had higher outpatient and total pharmacy costs.High-performance polymer materials that can exhibit distinguished mechanical properties have been developed based on material design considering energy dissipation by sacrificial bond dissociation. We now propose co-continuous network polymers (CNPs) for the design of tough polymer materials. CNP is a new composite material fabricated by filling the three-dimensionally continuous pores of a hard epoxy monolith with any cross-linked polymer having a low glass transition temperature (Tg). The structure and mechanical properties of the CNPs containing epoxy resins, thiol-ene thermosets, and polyacrylates as the low-Tg components were investigated by differential scanning calorimetry, dynamic mechanical analysis, tensile tests as well as scanning electron microscopic observations and non-destructive 3D X-ray imaging in order to clarify a mechanism for exhibiting an excellent strength and toughness. It has been demonstrated that the mechanical properties and fractural behavior of the CNPs significantly depend on the network structure of the filler polymers, and that a simultaneous high strength and toughness are achieved via the sacrificial fracture mechanism of epoxy-based hard materials with co-continuous network structures.
