The rate of Ca2+ transient recovery following PRL treatment was significantly increased in myocytes from diabetic and control heart. Amplitude of L-type Ca2+ current was not significantly altered by diabetes or by PRL. PRL increased the amplitude of shortening and Ca2+ transients in myocytes from control and diabetic heart. Increased fractional release of sarcoplasmic reticulum Ca2+ may partly underlie the positive inotropic effects of PRL in ventricular myocytes from control and STZ-induced diabetic rat. © 2020 Published by Elsevier Ltd.Over the years, the inability of pharmaceutical companies to achieve optimum maintenance of their equipment, and enhanced machine availability for better resource and maintenance utilization, has adversely affected their competitive advantage. The need to adopt a unique production technique that will curb their numerous equipment maintenance challenges, and also re-position them for world-class manufacturing will not only reduce their losses, but will also increase their throughput and profitability. To address a Pharmaceutical firm's maintenance challenges, data were obtained for the Overall Equipment Effectiveness (OEE) factors after Total Productive Maintenance (TPM) implementation in the company. Minitab 16.0 software was used to analyze the data collected, and the results showed that the highest value of 98.90 and 96.39 in the descriptive statistics for the maximum and mean respectively, underscore the importance of quality in the company's products. The percentage of mean for quality, availability, and performance obtained were 96.3906, 60.4938, and 27.6188 respectively. This once again showed that quality of products is the greatest OEE factor that pharmaceutical companies must take seriously in order to reduce the six big losses in their manufacturing processes. Response Surface Method (RSM), with the application of Design Expert software with Box-Behnken as the design type was used to model, analyze, and optimize the Overall Equipment Effectiveness (OEE) using availability, quality, and performance as the input parameters. The analysis of both the actual and coded values, which is the main contribution of the study showed that quality has the greatest value followed by availability and performance. Also, OEE must be set at 10.3 and 629.5 for both the lower and upper limits respectively, in order to effectively reduce downtime, setup cost, the inherent wastes, as well as the six big losses. © 2020 The Author(s).Amoxicillin dispersible tablet (DT) is now recommended by the WHO as a first-line drug for the treatment of pneumonia in children below 5 years. The study aim was to compare acceptability, adherence and clinical outcome of amoxicillin DT and amoxicillin oral suspension (OS) in the treatment of children aged 2-59 months with pneumonia in Kenya. We conducted a two-arm cluster randomized controlled trial and utilized quantitative methods. The community unit was the unit of randomization. Children aged 2-59 months with pneumonia were enrolled and treated with either amoxicillin DT or OS. Acceptability was defined as the perception of taste of medication as the same or better compared to other medicines and expression of willingness of caregivers to use DT/OS in future, adherence was measured based on the dose, frequency, and duration of treatment, and clinical outcome as complete resolution of symptoms without change of antibiotic treatment. Equivalence was defined as a difference of ≤8% between study arms. We found high levels of acceptability among both DT (93.9%) and OS (96.1%) arms (difference 2.3%, 90% CI -2.6-7.3). The objective measure of adherence on day four and the overall objective measure were significantly higher among children on DT compared to children on OS (88.7% vs. 41.5% (difference 47.2%, 90% CI 31.0-63.3) & 83.5% vs. 39% (difference 44.5%, 90% CI 27.9-60.9), respectively). Cure rates were high in both arms (DT (99.5%), OS (98.1%), difference 1.4%, 90% CI -0.2-3.2). There is reported better adherence to Amoxicillin DT compared to OS and equivalence in acceptability and clinical outcomes. © 2020 The Author(s).In this study, we report the synthesis of the magnetite/silica nanocomposites and their structural and functional groups, magnetic properties, morphology, antimicrobial activity, and drug delivery performance. The X-ray diffraction characterization showed that magnetite formed a spinel phase and that silica formed an amorphous phase. The particle sizes of magnetite increased from 8.2 to 13.2 nm with increasing silica content, and the particles were observed to be superparamagnetic. The nanocomposites tended to agglomerate based on the scanning electron microscopy images. The antimicrobial activity of the magnetite/silica nanocomposites revealed that the increasing silica content increased the inhibition zones by 74%, 77%, and 143% in case of Gram-positive bacteria (B. subtilis), Gram-negative bacteria (E. coli), and fungus (C. albicans), respectively. Furthermore, doxorubicin was used as the model compound in the drug loading and release study, and drug loading was directly proportional to the silica content. Thus, the increasing silica content increased the drug loading owing to the increasing number of OH- bonds in silica, resulting in strong bonds with doxorubicin. Based on this study, the magnetite/silica nanocomposites could be applied as drug delivery vehicles. © 2020 The Author(s).The paper suggests a method for optimization of the process of integrated tests for complex technical equipment of automotive, aviation, and rocket systems based on an analysis of efficiency dynamics models. The method makes it possible to determine scopes of ground tests for complex technical equipment, which are minimally required to start field tests under the necessity to combine ground and field development. Exponential models of complex systems' development are used as efficiency dynamics models for the whole testing process as well as efficiency dynamics models at various levels of the hierarchy of tests. The paper addresses an optimization task when the structure of tests at each level of the hierarchy is specified, i.e. efficiency dynamics models are determined for each level. selleck The authors determine optimal points of transition from one level of tests to another, considering the random nature of efficiency dynamics parameters. A method for optimization using determination of the optimal scope of field tests is given.
